ABSTRACT. The separation of Pneumocystis carinii life-cycle stages while preserving infectivity is a hitherto unresolved challenge. We describe an original, reproducible, and efficient method for separating trophic from cystic forms of P. carinii using a high-speed cell sorter. The large amounts of highly purified (99.6±0.3%) infectious trophic and cystic forms can now be used to elucidate the poorly understood P. carinii life cycle.  相似文献

Abstract.  The Brazilian apple leafroller, Bonagota cranaodes (Meyrick) (Lepidoptera: Tortricidae) is reared in the laboratory under a long-day (LD 14 : 10 h) and a short-day (LD 7 : 17 h) photoperiod at 22 °C, and under two different temperatures (10–13 °C and 21–22 °C). The development time from larval to adult eclosion do not differ between the two photoperiods, but did between the two temperature regimes. However, the larvae do not enter diapause, even under short day conditions and low temperatures. The number of adults obtained does not differ with temperature and light conditions. Field captures with pheromone traps show that Brazilian apple leafroller occurs in apple orchards throughout the year and the population densities are lower in winter. Accordingly, control measures should be taken during the off-season.  相似文献

Mean summer water temperatures in the Fraser River (British Columbia, Canada) have increased by ~1.5 °C since the 1950s. In recent years, record high river temperatures during spawning migrations of Fraser River sockeye salmon (Oncorhynchus nerka) have been associated with high mortality events, raising concerns about long‐term viability of the numerous natal stocks faced with climate warming. In this study, the effect of freshwater thermal experience on spawning migration survival was estimated by fitting capture–recapture models to telemetry data collected for 1474 adults (captured in either the ocean or river between 2002 and 2007) from four Fraser River sockeye salmon stock‐aggregates (Chilko, Quesnel, Stellako‐Late Stuart and Adams). Survival of Adams sockeye salmon was the most impacted by warm temperatures encountered in the lower river, followed by that of Stellako‐Late Stuart and Quesnel. In contrast, survival of Chilko fish was insensitive to the encountered river temperature. In all stocks, in‐river survival of ocean‐captured sockeye salmon was higher than that of river‐captured fish and, generally, the difference was more pronounced under warm temperatures. The survival–temperature relationships for ocean‐captured fish were used to predict historic (1961–1990) and future (2010–2099) survival under simulated lower river thermal experiences for the Quesnel, Stellako‐Late Stuart and Adams stocks. A decrease of 9–16% in survival of all these stocks was predicted by the end of the century if the Fraser River continues to warm as expected. However, the decrease in future survival of Adams sockeye salmon would occur only if fish continue to enter the river abnormally early, towards warmer periods of the summer, as they have done since 1995. The survival estimates and predictions presented here are likely optimistic and emphasize the need to consider stock‐specific responses to temperature and climate warming into fisheries management and conservation strategies.  相似文献

The delimitation of species is a major issue in systematic biology and has been a re-emerging discipline in the last decade. A number of studies have shown that the use of multiple data sets is critical for the identification of cryptic species, particularly in groups with complex evolutionary histories. Liolaemus monticola is a montane lizard species distributed in central Chile (32°–42°S), with four described subspecies in a latitudinal gradient from north to south: L. m. monticola , L. m. chillanensis , L. monticola ssp. and L. m. villaricensis . In order to test the systematic status and phylogenetic relationships of the taxa included in the L. monticola group, we analysed morphological (morphometric and meristic) and molecular (allozyme and mitochondrial DNA) data sets. The results of the morphological analyses showed that meristic variables correctly assigned individuals with higher accuracy than did morphometric characters. The results of the analyses of allozyme data revealed eight diagnostic loci that are evidence for significant differences among the four L. monticola subspecies. Phylogenetic analyses with mitochondrial DNA data, including additional species, showed that the L. monticola group is polyphyletic. We postulate that the four current subspecies represent independent evolutionary lineages and must be raised to the specific level as L. monticola , L. chillanensis and L. villaricensis . Variation at five polymorphic microsatellite loci was used to investigate genetic diversity and differentiation of two tetraploid Canarian endemics, Bencomia exstipulata and B. caudata. Data were analysed and are discussed in terms of tetrasomic (autotetraploid) and disomic (allotetraploid) inheritance. In both cases, genetic diversity values were similar to those described in other tetraploid plant species. High genetic differentiation between the only two described natural populations of B. exstipulata was detected (FST = 0.411). Bayesian cluster analysis revealed a geographical structure with distinct genetic groups from each island. High genetic differentiation and low genetic diversity of the B. exstipulata population from Tenerife suggest a recent population bottleneck, perhaps caused by the most recent major volcanic eruption, for this natural locality. This may be heightened by possible inbreeding depression and the monoecy of these species. Polymorphic microsatellite loci were also tested across all species in the Bencomia alliance. Male coleoid cephalopods produce spermatophores that can attach autonomously on the female's body during a complex process of evagination called the ‘spermatophoric reaction’, during which the ejaculatory apparatus and spiral filament of the spermatophore are everted and exposed to the external milieu. In some deepwater cephalopods, the reaction leads to the intradermal implantation of the spermatophore, a hitherto enigmatic phenomenon. The present study builds upon several lines of evidence to propose that spermatophore implantation is probably achieved through the combination of (1) an ‘evaginating‐tube’ mechanism performed by the everting ejaculatory apparatus and (2) the anchorage provided by the spiral filament's stellate particles. The proposed theoretical model assumes that, as it is exposed to the external milieu, each whorl of the spiral filament anchors to the surrounding tissue by means of its sharp stellate particles. As the ejaculatory apparatus tip continues evaginating, it grows in diameter and stretches lengthwise, enlarging the diameter of the whorl and propelling it, consequently tearing and pushing the anchored tissue outward and backward, and opening space for the next whorl to attach. After the ejaculatory apparatus has been everted and has perforated tissue, the cement body is extruded, possibly aiding in final attachment, and the sperm mass comes to lie inside the female tissue, encompassed by the everted ejaculatory apparatus tube. It is proposed that this unique, efficient spermatophore attachment mechanism possibly evolved in intimate relationship with the adoption of an active mode of life by coleoids. Miamiensis avidus is a marine polymorphic scuticociliate that undergoes microstome to tomite transformation when nutrients are depleted. This transformation takes place as a result of two consecutive divisions and does not involve a cyst. Tomitogenesis was induced by nutrient deprivation. When returned to nutrient medium tomites transformed into microstomes. Changes in population were noted at one-hour intervals. Tomites of Miamiensis avidus Ma/2 are compared to dispersal stages in other scuticociliates and hymenostomes.  相似文献

SYNOPSIS. The visual system of the horseshoe crab, Limulus polyphemus,provides an excellent opportunity for studying the neural basisof behavior. Quantitative analysis of the animal's visual behavioris now possible as is theoretical analysis of information processingin its retina. We combine these theoretical and behavioral approachesto investigate the nature of the signals the eye transmits tothe brain for the animal to see. Over the years theoretical studies of the Limulus eye were restrictedby the limited capabilities of single processor digital computers.However, a breakthrough in technology with the advent of parallelcomputers greatly enhances the analysis of large neural networkssuch as that of the retina. We have developed a time-dependentmodel of the Limulus retina on the Connection Machine (ModelCM-2), which is a massively parallel computer containing 32,768processors. The model represents a matrix of 64 x 128 receptorsand simulates interactions among receptors with digital filtersand transduction and adaptation within a receptor by a multistagecascade. Neural response patterns computed with the ConnectionMachine model replicate to a first approximation the patternsof neural activity recorded in the laboratory. Behavioral studies of Limulus vision carried out in the fieldcan be simulated on the Connection Machine. Neural responsesrecorded from behaving animals serve to test the accuracy ofthe model. Thus far we have developed just one model of theretina, but it eventually will have two forms, "daytime" and"nighttime," to account for the known circadian rhythms in retinalfunction. With a combination of field, physiological, and theoreticalstudies, we hope to gain a better understanding of the neuralmechanisms that underlie the animal's visually-guided behavior.  相似文献
