The kinase domain alters the kinetic properties of the myosin IIIA motor   总被引:2,自引:0,他引:2  
Myosin IIIA is unique among myosin proteins in that it contains an N-terminal kinase domain capable of autophosphorylating sites on the motor domain. A construct of myosin IIIA lacking the kinase domain localizes more efficiently to the stereocilia tips and alters the morphology of the tips in inner ear hair cells. Therefore, we performed a kinetic analysis of myosin IIIA without the kinase domain (MIII DeltaK) and compared these results with our reported analysis of myosin IIIA containing the kinase domain (MIII). The steady-state kinetic properties of MIII DeltaK indicate that it has a 2-fold higher maximum actin-activated ATPase rate (kcat = 1.5 +/- 0.1 s-1) and a 5-fold tighter actin affinity (KATPase = 6.0 +/- 1.4 microM, and KActin = 1.4 +/- 0.4 microM) compared to MIII. The rate of ATP binding to the motor domain is enhanced in MIII DeltaK (K1k+2 approximately 0.10 +/- 0.01 microM-1.s-1) to a level similar to the rate of binding to MIII in the presence of actin. The rate of ATP hydrolysis in the absence of actin is slow and may be rate limiting. Actin-activated phosphate release is identical with and without the kinase domain. The transition between actomyosin.ADP states, which is rate limiting in MIII, is enhanced in MIII DeltaK. MIII DeltaK accumulates more efficiently at the tips of filopodia in HeLa cells. Our results suggest a model in which the activity and concentration of myosin IIIA localized to the tips of actin bundles mediates the morphology of the tips in sensory cells.  相似文献

Summary Standard methods to estimate heritability by half-sib correlation are biased if selection has operated in the parental generation. In this paper a simple method to correct for selection of animals used as sires is described. By selection of both the top and the bottom ranking sires, the sampling variances of the corrected estimates of heritability are substantially reduced. Algebraic expressions to predict the sampling variance of the estimates of heritability using selected sires are derived. Theoretical predictions were checked by Monte-Carlo simulation. The results may have application in the design of experiments to estimate heritabilities.  相似文献

Rapid advances in human genomic research are increasing the availability of genomic data for secondary analysis. Particularly in the case of vulnerable African populations, ethics and informed consent processes need to be transparent-both to ensure participant protection, as well as to share skills and to evolve best practice for informed consent from a shared knowledge base. One of the main mechanisms for double stranded DNA break (DSB) repair is through the non-homologous end-joining (NHEJ) pathway. Using plasmid and chromosomal repair assays, we showed that deletion mutant strains for interacting proteins Pph3p and Psy2p had reduced efficiencies in NHEJ. We further observed that this activity of Pph3p and Psy2p appeared linked to cell cycle Rad53p and Chk1p checkpoint proteins. Pph3/Psy2 is a phosphatase complex, which regulates recovery from the Rad53p DNA damage checkpoint. Overexpression of Chk1p checkpoint protein in a parallel pathway to Rad53p compensated for the deletion of PPH3 or PSY2 in a chromosomal repair assay. Double mutant strains Δpph3/Δchk1 and Δpsy2/Δchk1 showed additional reductions in the efficiency of plasmid repair, compared to both single deletions which is in agreement with the activity of Pph3p and Psy2p in a parallel pathway to Chk1p. Genetic interaction analyses also supported a role for Pph3p and Psy2p in DNA damage repair, the NHEJ pathway, as well as cell cycle progression. Collectively, we report that the activity of Pph3p and Psy2p further connects NHEJ repair to cell cycle progression.  相似文献

Question: What are the effects of grazing abandonment on the vegetation composition of Estonian coastal wetlands? Location: Vormsi Island and Silma Nature Reserve in western Estonia, Europe. Methods: Local knowledge and field reconnaissance were used to identify current and historical management levels of wetland sites within the west Estonian study area. Nine study sites, with varying management histories, were selected comprising an area of 287 ha. A total of 198 quadrats were taken from 43 distinct vegetation patches in five of the sites. TWINSPAN analysis was used to identify community type, and a phytosociological key was constructed for character taxa. This vegetation classification was then applied within a GIS‐based context to classify all the study sites, using a ground survey technique and 1:2000 scale air photos. Results: We identified 11 different brackish coastal wetland community types. Indicator species were defined with community characteristics for the seven main vegetation types readily recognisable in the field. Coastal wet grasslands were most extensive in grazed sites, or sites that had been more intensively grazed, while abandoned sites were largely composed of Phragmites australis stands, tall grassland, and scrub. Site variations based on vegetation composition were significantly correlated with past grazing intensity. Plant community types showed significant edaphic differences, with particularly low soil moisture and high conductivity and pH for open pioneer patches compared to other vegetation types. Conclusion: Abandonment of traditionally grazed coastal grasslands threatens their characteristic biodiversity. This study found that grazing abandonment reduced the extent of coastal wetland grasslands of particular conservation value. Nevertheless, plant species of conservation interest were found across the sequence of community types described. The study shows that grazing is an important factor influencing coastal wetland plant communities but suggests that vegetation distribution is affected by environmental variables, such as topography.  相似文献

Myosin IIIA is specifically expressed in photoreceptors and cochlea and is important for the phototransduction and hearing processes. In addition, myosin IIIA contains a unique N-terminal kinase domain and C-terminal tail actin-binding motif. We examined the kinetic properties of baculovirus expressed human myosin IIIA containing the kinase, motor, and two IQ domains. The maximum actin-activated ATPase rate is relatively slow (k(cat) = 0.77 +/- 0.08 s(-1)), and high actin concentrations are required to fully activate the ATPase rate (K(ATPase) = 34 +/- 11 microm). However, actin co-sedimentation assays suggest that myosin III has a relatively high steady-state affinity for actin in the presence of ATP (K(actin) approximately 7 microm). The rate of ATP binding to the motor domain is quite slow both in the presence and absence of actin (K(1)k(+2) = 0.020 and 0.001 microm(-1).s(-1), respectively). The rate of actin-activated phosphate release is more than 100-fold faster (85 s(-1)) than the k(cat), whereas ADP release in the presence of actin follows a two-step mechanism (7.0 and 0.6 s(-1)). Thus, our data suggest a transition between two actomyosin-ADP states is the rate-limiting step in the actomyosin III ATPase cycle. Our data also suggest the myosin III motor spends a large fraction of its cycle in an actomyosin ADP state that has an intermediate affinity for actin (K(d) approximately 5 microm). The long lived actomyosin-ADP state may be important for the ability of myosin III to function as a cellular transporter and actin cross-linker in the actin bundles of sensory cells.  相似文献
