Summary Four monoclonal antibodies specific for somatostatin have been produced and characterized. These antibodies were used to assess the anatomical relationship of somatostatin-containing cells in the pancreas and gastrointestinal tract of man, baboon and rat with ten other peptide-containing endocrine cells. The peptides investigated were gastrin, cholecystokinin, motilin, secretin, neurotensin, gastric inhibitory polypeptide, gut-glucagon, pancreatic glucagon, pancreatic polypeptide and insulin.The only regions in which somatostatin cells were seen in close contact with another endocrine cell were in the pancreas and the gastric antrum. In the pancreas somatostatin cells were commonly seen in close contact with insulin, glucagon and pancreatic polypeptide cells and infrequent contact was demonstrable with the gastrin-immunoreactive cells in the antrum of both rat and man. In all other cases no evidence was obtained for a close anatomical relationship between somatostatin cells and the other enteroendocrine cells.  相似文献

Most behavioural traits show plastic responses to changes in internal or external conditions. Similarly, animal personality is not necessarily fixed during an individual's lifetime, leading to age-related changes. Both individual plasticity (i.e., within-individual effect) and non-random selective (dis)appearance of behavioural types (i.e., between-individual effect) may contribute to age-related changes observed at the population level. Here, we investigated how boldness changes with age in a woodland population of the Speckled wood butterfly (Pararge aegeria L.) using a capture–mark–recapture approach. We used wing wear as an index for age and we show that fresh individuals are bolder than worn individuals. Using the subsample of recaptured butterflies, we found that this pattern is most likely driven by individual plasticity. Our design also allowed us to explore some aspects of the species' spatial ecology and how it relates to personality. We found no relationship between boldness and net displacement between successive captures, a finding which we discuss within a movement ecology framework.  相似文献

Habitats selected for development may have important fitness consequences. This is relevant within the framework of niche shifts in human-dominated landscapes. Currently, the peacock butterfly (Aglais io) occurs ubiquitously, covering many habitat types, whereas its distribution used to be much more restricted. Indeed, its host plant (stinging nettle Urtica dioica) was limited to natural forest gaps on relatively nitrogen-rich soil, but due to land use changes and eutrophication, host plants are now quasi-omnipresent in Western Europe. In order to assess the impact of specific anthropogenic habitat types on host plant quality and environmental conditions for phenotypic trait values, an experiment was conducted in woodlands, field margins, and urban gardens. Larval development was studied in field enclosures, and adult traits were analyzed to test predicted effects of warmer and more nitrogen-rich conditions in field margins compared to woodlands and urban gardens. Survival to the adult stage was highest in woodlands and lowest in field margins, and whilst development time did not differ amongst habitat types, butterflies that developed in field margins were larger and had higher lipid content and wing loadings than conspecifics from woodlands and urban gardens. Nettles in field margins provided warmer microclimates. However, and contrary to predictions, the nitrogen level within host plant leaves was highest in woodlands. Hence, anthropogenic landscapes may pose a conflict for choosing what is ultimately the best breeding habitat, as survival was highest in woodlands (followed by urban gardens), but adults with highest fitness predictions were produced in field margins (and secondarily urban gardens).  相似文献

Autophagic and proteasomal degradation constitute the major cellular proteolysis pathways. Their physiological and pathophysiological adaptation and perturbation modulates the relative abundance of apoptosis-transducing proteins and thereby can positively or negatively adjust cell death susceptibility. In addition to balancing protein expression amounts, components of the autophagic and proteasomal degradation machineries directly interact with and co-regulate apoptosis signal transduction. The influence of autophagic and proteasomal activity on apoptosis susceptibility is now rapidly gaining more attention as a significant modulator of cell death signalling in the context of human health and disease. Here we present a concise and critical overview of the latest knowledge on the molecular interplay between apoptosis signalling, autophagy and proteasomal protein degradation. We highlight that these three pathways constitute an intricate signalling triangle that can govern and modulate cell fate decisions between death and survival. Owing to rapid research progress in recent years, it is now possible to provide detailed insight into the mechanisms of pathway crosstalk, common signalling nodes and the role of multi-functional proteins in co-regulating both protein degradation and cell death.  相似文献

Most bacteria in the ocean can be motile. Chemotaxis allows bacteria to detect nutrient gradients, and hence motility is believed to serve as a method of approaching sources of food. This picture is well established in a stagnant environment. In the ocean a shear microenvironment is associated with turbulence. This shear flow prevents clustering of bacteria around local nutrient sources if they swim in the commonly assumed "run-and-tumble" strategy. Recent observations, however, indicate a "back-and-forth" swimming behavior for marine bacteria. In a theoretical study we compare the two bacterial swimming strategies in a realistic ocean environment. The "back-and-forth" strategy is found to enable the bacteria to stay close to a nutrient source even under high shear. Furthermore, rotational diffusion driven by thermal noise can significantly enhance the efficiency of this strategy. The superiority of the "back-and-forth" strategy suggests that bacterial motility has a control function rather than an approach function under turbulent conditions.  相似文献

The overall objective was to evaluate the use of porcine luteinizing hormone (pLH) for synchronization of ovulation in cyclic gilts and its effect on reproductive function. In an initial study, four littermate pairs of cyclic gilts were given altrenogest (15 mg/d for 14 d). Gilts received 500 microg cloprostenol (Day 15), 600 IU equine chorionic gonadotropin (eCG) (Day 16) and either 5mg pLH or saline (Control) 80 h after eCG. Blood samples were collected every 4h, from 8h before pLH/saline treatment to the end of estrus. Following estrus detection, transcutaneous real-time ultrasonography and AI, all gilts were slaughtered 6d after the estimated time of ovulation. Peak plasma pLH concentrations (during the LH surge), as well as the amplitude of the LH surge, were greater in pLH-treated gilts than in the control (P=0.01). However, there were no significant differences between treatments in the timing and duration of estrus, or the timing of ovulation within the estrous period. In a second study, 45 cyclic gilts received altrenogest for 14-18d, 600 IU eCG (24h after last altrenogest), and 5mg pLH, 750 IU human chorionic gonadotropin (hCG), or saline, 80 h after eCG. For gilts given pLH or hCG, the diameter of the largest follicle before the onset of ovulation (mean+/-S.E.M.; 8.1+/-0.2 and 8.1+/-0.2mm, respectively) was smaller than in control gilts (8.6+/-0.2mm, P=0.05). The pLH and hCG groups ovulated sooner after treatment compared to the saline-treated group (43.2+/-2.5, 47.6+/-2.5 and 59.5+/-2.5h, respectively; P<0.01), with the most synchronous ovulation (P<0.01) in pLH-treated gilts. Embryo quality (total cell counts and embryo diameter) was not significantly different among groups. In conclusion, pLH reliably synchronized ovulation in cyclic gilts without significantly affecting embryo quality.  相似文献

Insight into demographic processes that operate at larger spatial scales can be achieved through studying local populations when a particular species of interest is examined over time, by many investigators, in a variety of locations. On the west coast of North America, Mazzaella splendens (Setchell et Gardner) Fredericq is such a species of interest. A synthesis of local demographic studies of M. splendens from the late 1960s to the present reveals a pattern that is potentially common to the larger natural populations. This is the pattern: population density is high in summer and low in winter for both alternate free‐living life history phases of M. splendens. The magnitude of this seasonal change decreases in increasingly wave‐exposed habitats. In wave‐sheltered habitats there is a seasonal alternation from summer haploid to winter diploid dominance. This alternation gradually changes to constant diploid dominance as wave exposure in the habitat increases. Changes in population density are primarily a function of appearances and disappearances of perennating basal crusts (genets), as modules are produced or lost, rather than differential module production by genets of one phase over those of the other. To test the generality of this pattern, we examined seasonal changes in density, in local populations of M. splendens, in both a wave‐sheltered and a wave‐exposed habitat at Second Beach, Barkley Sound. Greater seasonal fluctuation in population density at wave‐sheltered, compared to wave‐exposed habitats is supported as a pattern potentially common to the natural populations of M. splendens. A change from summer haploid dominance in wave‐sheltered areas to summer diploid dominance in wave‐exposed areas is similarly supported. All changes in population density were the result of appearances and disappearances of genets rather than differential module production by haploid versus diploid basal crusts, also consistent with previous observations. A seasonal alternation in phase dominance, however, was absent from the wave‐sheltered site at Second Beach, Barkley Sound for 3 consecutive years. Seasonal alternation in phase dominance of M. splendens appears dependent on local conditions and is not common to all natural populations.  相似文献
