Cellular immune findings in Lyme disease   总被引:3,自引:0,他引:3  
From 1981 through 1983, we did the first testing of cellular immunity in Lyme disease. Active established Lyme disease was often associated with lymphopenia, less spontaneous suppressor cell activity than normal, and a heightened response of lymphocytes to phytohemagglutinin and Lyme spirochetal antigens. Thus, a major feature of the immune response during active disease seems to be a lessening of suppression, but it is not yet known whether this response plays a role in the pathophysiology of the disease.  相似文献

Semiallogeneic somatic hybrid cells (AB2) derived from fusion of a C57B1/6 chemically induced fibrosarcoma (MCB6-1) and a fibroblastic cell (A9) of C3H origin were used to immunize C57B1/6 mice against the parental MCB6-1 tumor cells. In vitro immune lymphocytes were directly cytotoxic against AB2 hybrid cells and A9 allogeneic parental cells, but could not lyse the syngeneic MCB6-1 parental tumor cells. Nevertheless, after a 4-day culture of these immune lymphocytes, a cytotoxic activity against the syngeneic MCB6-1 tumor cells appeared; expression of such a cytotoxic activity did not require the presence of stimulator cells (mitomycin-treated MCB6-1 tumor cells) during the culture. This cytotoxicity is mediated by T cells, as it was completely abrogated by treatment with anti-Thy 1–2 antiserum and complement. These results suggest that a maturation or a differentiation of immune T lymphocytes occurs during in vitro culture, and is necessary for the expression of antitumor cytotoxicity.  相似文献

  1. Movement behaviours of adult aquatic insects can produce distinct spatial distribution patterns. Studies of adult abundance with distance away from water bodies are common and may invoke flight capability to explain species differences. In contrast, distribution patterns along river channels are poorly described, but are no less important for understanding population dynamics. Longitudinal patterns in adult abundance along short river lengths may differ between sexes and at different life stage transitions between aquatic and terrestrial environments, i.e. at emergence and oviposition. Flight capability is unlikely to influence longitudinal patterns created at emergence, but may influence local abundances of mature females seeking to lay eggs. We tested hypotheses about how local abundances of mature females might differ according to oviposition habits and flight capability.
  2. We surveyed abundances of mature female caddisflies at adjacent riffle–pool pairs along short river lengths with homogeneous riparian cover. Our survey included nine species in three families (Hydrobiosidae, Leptoceridae, Hydropsychidae), which encompassed multiple different oviposition habits and a range of wing sizes and shapes. Several of the species oviposit preferentially in riffles. Accordingly, we tested for differences in female abundance between channel units (adjacent riffle–pool pairs). We also tested whether females attained higher abundances in some places along channels than others (i.e. over larger spatial scales and regardless of channel unit) which imply movements along the channel and aggregation in some locations. Wing morphology was used as a proxy measure of flight capability and included measures of wing span, area, aspect ratio and the second moment of wing area.
  3. Three distinctly different distribution patterns of mature female caddisflies were identified. The abundance of three species varied over larger scales only (multiple channel units). Six species that oviposit preferentially in riffles had higher female abundances at riffles than pools, but for only one did abundances also vary over larger scales. There was no association between these different patterns and measures of wing morphology, after removing metrics that were correlated and that differed systematically between taxonomic families. However, we could not reject the hypothesis that some aspect of flight behaviour may have contributed to observed patterns.
  4. The diverse but distinct distributions of mature female caddisflies we observed along short channel lengths are novel and suggest that species differ in their propensity for movement along streams, which could have consequences for local densities of eggs and juveniles in the aquatic environment. The degree to which population sizes are coupled across the terrestrial-to-aquatic transition is rarely investigated in aquatic insects and may provide fresh insight into sources of spatial variation within populations. Similarly, a more nuanced approach to research on the flight of aquatic insects, including age- and sex-specific phenomena, may provide greater insight into the diverse ecological functions and consequences of movement.

Focal adhesion kinase (FAK) is a major signaling molecule which functions downstream of integrins or in conjunction with mitogenic signaling pathways. FAK is overexpressed and/or activated in many types of human tumors, in which it promotes cell adhesion, survival, migration and invasion. In addition to FAK''s ability to regulate signaling through its scaffolding activities, FAK encodes an intrinsic kinase activity. Although some FAK substrates have been identified, a more comprehensive analysis of substrates is lacking. In this study, we use a protein microarray to screen the human proteome for FAK substrates. We confirm that several of the proteins identified are bona fide in vitro FAK substrates, including several factors which are known to regulate the NFκB pathway. Finally, we identify a role for FAK''s kinase activity in both canonical and non-canonical NFκB signaling. Our screen therefore represents the first high throughput screen for FAK substrates and provides the basis for future in-depth analysis of the role of FAK''s kinase activity in the processes of tumorigenesis.  相似文献

Premise

Seed production is frequently limited by the receipt of insufficient or low-quality pollen, collectively termed “pollen limitation” (PL). In taxa with gametophytic self-incompatibility (GSI), incompatible pollen can germinate on stigmas but pollen tubes are arrested in styles. This allows for estimates of pollen performance before, during, and after self-recognition, as well as insight into the factors underlying pollen quality limitation in GSI taxa.

Methods

We scored pollen performance following self and outcross pollinations in Argentina anserina to identify the location of self-recognition and establish the relationship between pollen tubes and seed production. We then estimated quantity and quality components of PL from >3300 field-collected styles. We combined our results with other studies to test the prediction that low pollen quality, but not quantity, drives higher PL in self-incompatible (SI) taxa than in self-compatible taxa (SC).

Results

Self and outcross pollen germinated readily on stigmas, but 96% of germinated self-pollen was arrested during early tube elongation. Reproduction in the field was more limited by pollen quality than by quantity, and pollen failure near the location of self-recognition was a stronger barrier to fertilization than pollen germination. Across 26 taxa, SI species experienced stronger pollen quality, but not quantity, limitation than SC species.

Conclusions

Evaluating pollen performance at multiple points within pistils can elucidate potential causes of pollen quality limitation. The receipt of incompatible pollen inhibits fertilization success more than insufficient pollen receipt or poor pollen germination in A. anserina. Likewise, pollen quality limitation drives high overall PL in other SI taxa.  相似文献

Catechol 2,3-dioxygenase from the meta-cleavage pathway encoded on the TOL plasmid of Pseudomonas putida (pWWO) was investigated by electron microscopy. Negatively stained samples of the purified catechol 2,3-dioxygenase revealed that the enzyme consists of four subunits arranged in a tetrahedral conformation. Monoclonal antibodies raised against catechol 2,3-dioxygenase showed highly specific reactions and were used to localize the enzyme in Escherichia coli (pAW31) and P. putida (pWWO), using the protein A-gold technique carried out as a post-embedding immunoelectron microscopy procedure. Olanzapine, an atypical antipsychotic drug, was previously shown to protect neuronal cells against nutrient deprivation and to enhance neurite outgrowth. In an effort to identify small molecules with greater potency, the structure of olanzapine was used as a template to search commercially available chemical inventories for compounds with similar features. These compounds were evaluated for their ability to protect cells against glutamine deprivation and low-serum conditions. Positive compounds, 'hits' from initial screening, were then tested for stimulation of neurite outgrowth, alone and in combination with suboptimum concentrations of nerve growth factor (NGF). Numerous neuroprotective compounds (mw < 550 Da) were identified that significantly stimulated neurite outgrowth in PC12 cells. These included 4', 6'-diamidino-2-phenylindole, a nuclear stain; staurosporine, an antibiotic and kinase inhibitor; and 2-phenylamino-adenosine, an adenosine analog. The small molecules were comparable with NGF, and in fact, replaced NGF in outgrowth assays. Pharmacophore analysis of the hits led to the design and synthesis of an active compound, LSU-D84, which represented an initial lead for drug discovery efforts.  相似文献
