We have recently established that the facultative phototrophic bacterium Rhodobacter sphaeroides, like the closely related Rhodobacter capsulatus species, contains both the previously characterized mobile electron carrier cytochrome c2 (cyt c2) and the more recently discovered membrane-anchored cyt cy. However, R. sphaeroides cyt cy, unlike that of R. capsulatus, is unable to function as an efficient electron carrier between the photochemical reaction center and the cyt bc1 complex during photosynthetic growth. Nonetheless, R. sphaeroides cyt cy can act at least in R. capsulatus as an electron carrier between the cyt bc1 complex and the cbb3-type cyt c oxidase (cbb3-Cox) to support respiratory growth. Since R. sphaeroides harbors both a cbb3-Cox and an aa3-type cyt c oxidase (aa3-Cox), we examined whether R. sphaeroides cyt cy can act as an electron carrier to either or both of these respiratory terminal oxidases. R. sphaeroides mutants which lacked either cyt c2 or cyt cy and either the aa3-Cox or the cbb3-Cox were obtained. These double mutants contained linear respiratory electron transport pathways between the cyt bc1 complex and the cyt c oxidases. They were characterized with respect to growth phenotypes, contents of a-, b-, and c-type cytochromes, cyt c oxidase activities, and kinetics of electron transfer mediated by cyt c2 or cyt cy. The findings demonstrated that both cyt c2 and cyt cy are able to carry electrons efficiently from the cyt bc1 complex to either the cbb3-Cox or the aa3-Cox. Thus, no dedicated electron carrier for either of the cyt c oxidases is present in R. sphaeroides. However, under semiaerobic growth conditions, a larger portion of the electron flow out of the cyt bc1 complex appears to be mediated via the cyt c2-to-cbb3-Cox and cyt cy-to-cbb3-Cox subbranches. The presence of multiple electron carriers and cyt c oxidases with different properties that can operate concurrently reveals that the respiratory electron transport pathways of R. sphaeroides are more complex than those of R. capsulatus. 相似文献

The invasion of an established community by new species can trigger changes in community structure. Invasions often occur in phytophagous insect communities, the dynamics of which are driven by the structure of the host assemblage and the presence of competitors. In this study, we investigated how a community established through successive invasions changed over time, taking the last invasion as the reference. The community included four generalist and four specialist species of Tephritidae fruit flies. We analyzed a long‐term database recording observed numbers of flies per fruit for each species on 36 host plants, over 18 years, from 1991 to 2009. Community structure before the last invasion by Bactrocera zonata in 2000 was described in relation to host plant phylogeny and resource availability. Changes in the host range of each species after the arrival of B. zonata were then documented by calculating diversity indices. The flies in the community occupied three types of niches defined on the basis of plant phylogeny (generalists, Solanaceae specialist, and Cucurbitaceae specialists). After the arrival of B. zonata, no change in the host range of specialist species was observed. However, the host ranges of two generalist species, Ceratitis quilicii and Ceratitis capitata, tended to shrink, as shown by the decreases in species richness and host plant α‐diversity. Our study shows increased host specialization by generalist phytophagous insects in the field following the arrival of an invasive species sharing part of their resources. These findings could be used to improve predictions of new interactions between invaders and recipient communities. 相似文献

Several mechanoregulatory tissue differentiation models have been proposed over the last decade. Corroboration of these models by comparison with experimental data is necessary to determine their predictive power. So far, models have been applied with various success rates to different experimental set-ups investigating mainly secondary fracture healing. In this study, the mechanoregulatory models are applied to simulate the implant osseointegration process in a repeated sampling in vivo bone chamber, placed in a rabbit tibia. This bone chamber provides a mechanically isolated environment to study tissue differentiation around titanium implants loaded in a controlled manner. For the purpose of this study, bone formation around loaded cylindrical and screw-shaped implants was investigated. Histologically, no differences were found between the two implant geometries for the global amount of bone formation in the entire chamber. However, a significantly larger amount of bone-to-implant contact was observed for the screw-shaped implant compared to the cylindrical implant. In the simulations, a larger amount of bone was also predicted to be in contact with the screw-shaped implant. However, other experimental observations could not be predicted. The simulation results showed a distribution of cartilage, fibrous tissue and (im)mature bone, depending on the mechanoregulatory model that was applied. In reality, no cartilage was observed. Adaptations to the differentiation models did not lead to a better correlation between experimentally observed and numerically predicted tissue distribution patterns. The hypothesis that the existing mechanoregulatory models were able to predict the patterns of tissue formation in the in vivo bone chamber could not be fully sustained. 相似文献

Background
Application of plasmid DNA for immunization of food-producing animals established new standards of food safety. The addition of foreign products e.g. pDNA into the food chain should be carefully examined to ensure that neither livestock animals nor consumers develop unpredicted or undesirable side-effects.Methods
A quantitative real-time PCR (QRTPCR) methodology was developed to study the biodistribution and persistence of plasmid DNA vaccine pDNAX (pVAX-Hsp60 TM814) in mice and beef cattle. The linear quantification range and the sensitivity of the method was found to be 10 – 109 copies per reaction (500 ng/gDNA) and 3 copies per reaction, respectively.Results
Persistence of pDNAX in mice muscle tissue was restricted to injection site and the amount of pDNAX showed delivery formulation dependent (naked pDNA, electroporation, cationic liposome complexes) and mouse age-dependent clearance form injection site but pDNAX was still detectable even after 365 days. The QRTPCR analysis of various muscle tissue samples of vaccinated beef bulls performed 242–292 days after the last revaccination proved that residual pDNAX was found only in the injection site. The highest plasmid levels (up to 290 copies per reaction) were detected in the pDNAX:CDAN/DOPE group similarly to mice model. No pDNA was detected in the samples from distant muscles and draining lymph nodes.Conclusion
Quantitative real-time PCR (QRTPCR) assay was developed to assess the residual pDNA vaccine pVAX-Hsp60 TM814 in mice and beef cattle. In beef cattle, ultra low residual level of pDNA vaccine was only found at the injection site. According to rough estimation, consumption of muscles from the injection site represents almost an undetectable intake of pDNA (400 fg/g muscle tissue) for consumers. Residual plasmid in native state will hardly be found at measurable level following further meat processing. This study brings supportive data for animal and food safety and hence for further approval of pDNA vaccine field trials. 相似文献55.
In an appropriate ionic environment, the resting potential of canine cardiac purkinje fibers may have either of two value. By changing the external K concentration, [K](0), in small steps, it was shown that, in the low (1 mM) Cl, Na-containing solutions used in this study, the two levels of resting potential could be obtained only within a narrow range of [K](0) values; that range was usually found between 1 and 4 mM. Within the critical [K](0) range the resting potential could be shifted from either level to the other by the application of small current pulses. It was shown that under these conditions the steady-state current- voltage relationship was “N-shaped,” and that a region of both negative slope, and negative chord conductance lay between the two stable zero-current potentials. The negative chord conductance was largely due to inward sodium current, only part of which was sensitive to tetrodotoxin (TTX). Under appropriate conditions, the negative chord conductance could be abolished by several experimental interventions and the membrane potential thereby shifted from the lower to the higher resting level: those interventions which were effective by presumably diminishing the steady-state inward current included reducing the external sodium concentration, adding TTX, or adding lidocaine; those which presumably increased the steady-state outward current included small increases in [K](0), brief depolarizations to around -20 mV, or the addition of acetylcholine chloride. 相似文献

- 1 Primary resources may affect the structure of species assemblages in upper trophic levels of food webs. These bottom‐up effects may involve important ecological processes that affect pest control. For example, population densities of natural enemies may increase when alternative preys are favoured by a new resource.
- 2 Ants, particularly Solenopsis geminata Forel (Myrmicinae), are recognized as effective predators of herbivores eggs. In the present study, we tested the hypothesis that the addition of a primary resource in a banana agroecosystem will increase pest control of the banana weevil Cosmopolites sordidus Germar (Coleoptera, Curculionidae) by ants.
- 3 Using stable isotope analyses, we showed that S. geminata used the new resource pathway provided by the addition of a cover crop. This change was revealed by increased δ13C values (13C/12C) in a cover crop treatment (C4 plant) compared with a banana (C3 plant) in bare soil treatment.
- 4 We also showed that the abundance of S. geminata, as measured with pitfall traps, was 4.8–5.4‐fold higher in the cover crop treatment than in the bare soil treatment. Estimation of egg predation rates was in the range 7.8–70.3%, which occurred in plots with the lowest and highest S. geminata numbers, respectively.
- 5 The addition of a new primary resource in this system changed the origin of the carbon consumed by predators and increased their densities. The increased ant abundance resulted in increased predation of weevil eggs, which was consistent with our hypothesis. These results will be useful for the design of cropping systems that enhance the biological regulation of pests.

Invasive species are often said to be r -selected. However, invaders must sometimes compete with related resident species. In this case invaders should present combinations of life-history traits that give them higher competitive ability than residents, even at the expense of lower colonization ability. We test this prediction by comparing life-history traits among four fruit fly species, one endemic and three successive invaders, in La Réunion Island. Recent invaders tend to produce fewer, but larger, juveniles, delay the onset but increase the duration of reproduction, survive longer, and senesce more slowly than earlier ones. These traits are associated with higher ranks in a competitive hierarchy established in a previous study. However, the endemic species, now nearly extinct in the island, is inferior to the other three with respect to both competition and colonization traits, violating the trade-off assumption. Our results overall suggest that the key traits for invasion in this system were those that favoured competition rather than colonization. 相似文献

1. Noxious larval secretions of leaf beetles, which repel generalist predators, do not deter specialist syrphid fly predators (genus Parasyrphus ). These flies cause considerable mortality to the beetles, but little is known about their foraging behaviour.
2. Larvae of Parasyrphus nigritarsis were attracted to the volatile larval secretions produced by two prey species Phratora vitellinae and Linaeidea aenea. Parasyrphus nigritarsis feeds on both beetles in nature. Phratora vitellinae feeds on willows and utilizes host plant compounds for secretion production, while the alder-feeding L. aenea produces an autogenous secretion.
3. Fly larvae were strongly attracted to pieces of filter paper treated with larval secretion of the beetles. They attempted to feed on them for up to 7 min, and were equally attracted to the secretions of Ph. vitellinae and L. aenea . Fly larvae were also attracted to pure salicyl aldehyde, the main component of the secretion of Ph. vitellinae .
4. Fly larvae searched extensively for prey on leaves that had been damaged by beetle larvae. They also followed trails made with solutions containing faecal matter of prey larvae. They showed no differential preference for Ph. vitellinae or L. aenea , but always rejected larvae of the non-prey leaf beetle Agelastica alni .
5. Beetle secretions thus play an important, but unexpected, role in the feeding behaviour of P. nigritarsis . This predator uses the beetle secretion to locate its prey. The implications of these results for three trophic level interactions are discussed. 相似文献
