The important role of semiochemicals in the interactions between plants and insects has been extensively investigated. The volatiles produced by oak trees are thought to attract the ambrosia beetle Platypus quercivorus (Murayama), which causes wilt disease in Quercus trees, resulting in widespread damage. In the present study, we hypothesized that (a) P. quercivorus is attracted to the leaf volatiles emitted by host trees and (b) the response of P. quercivorus to leaf volatiles is affected by flight. An experiment was performed to survey the preferences of both sexes of P. quercivorus for the leaf volatiles of Quercus crispula Blume at various stages of leaf deterioration, represented by different number of days after cutting. Additionally, the effect of flight on both sexes was evaluated by testing the beetle flight on a flight mill. The results showed that P. quercivorus was attracted to the volatiles emitted from fresh leaves and was not attracted to those emitted from dry leaves. This suggests that leaf volatiles from healthy hosts are primary attractants for P. quercivorus. Further, males exhibited increased olfactory responses to leaf volatiles as their flight duration increased, whereas the opposite pattern was observed in females. These results suggest that the different ecological roles of the sexes contribute to differences in olfactory responses.  相似文献

For most multidomain proteins the thermal unfolding transitions are accompanied by an irreversible step, often related to aggregation at elevated temperatures. As a consequence the analysis of thermostabilities in terms of equilibrium thermodynamics is not applicable, at least not if the irreversible process is fast with respect the structural unfolding transition. In a comparative study we investigated aggregation effects and unfolding kinetics for five homologous alpha-amylases, all from mesophilic sources but with rather different thermostabilities. The results indicate that for all enzymes the irreversible process is fast and the precedent unfolding transition is the rate-limiting step. In this case the kinetic barrier toward unfolding, as measured by unfolding rates as function of temperature, is the key feature in thermostability. The investigated enzymes exhibit activation energies (E(a)) between 208 and 364 kJmol(-1) and pronounced differences in the corresponding unfolding rates. The most thermostable alpha-amylase from Bacillus licheniformis (apparent transition temperature, T(1/2) approximately 100 degrees C) shows an unfolding rate which is four orders of magnitude smaller as compared with the alpha-amylase from pig pancreas (T(1/2) approximately 65 degrees C). Even with respect to two other alpha-amylases from Bacillus species (T(1/2) approximately 86 degrees C) the difference in unfolding rates is still two orders of magnitude.  相似文献

Stretchable piezoelectric nanogenerators (SPENGs) for human kinematics energy harvesting have limited use due to the low stretchability or mechanical robustness and the difficulty of structural design for omnidirectional stretchability. This study reports an efficient, omnidirectionally stretchable, and robust SPENG based on a stretchable graphite electrode on a 3D micropatterned stretchable substrate and a stacked mat of piezoelectric nanofibers. The stacked mat of free‐standing nanofibers is alternatively composed of nanocomposite nanofibers of barium titanate nanoparticles embedded in polyurethane and poly(vinylidene fluoride‐trifluoroethylene) nanofibers. The nanofiber SPENG (nf‐SPENG) exhibits a high stretchability of 40% and high mechanical durability up to 9000 stretching cycles at 30% strain, which are attributed to the stress‐relieving nature of the 3D micropattern on the substrate and the free‐standing stacked hybrid nanofibers. The nf‐SPENG produces a peak open circuit voltage (Voc) and short circuit current (Isc) of 9.3 V and 189 nA, respectively. The nf‐SPENG is demonstrated to harvest the energy from human kinematics while walking when placed over the knee cap of a subject, generating a maximum Voc of 10.1 V. The omnidirectional stretchability, efficiency, facile fabrication process, mechanical durability, environmentally friendly lead‐free components, and response to multimodal straining make this device suitable for self‐powered wearable sensing systems.  相似文献

A new species of Goniothalamus (Blume) Hook.f. & Thomson (Annonaceae), G. banii B. H. Quang, R. K. Choudhary & V.T. Chinh, is described and illustrated from Thanh Hoa Province, Vietnam. This species shows morphological similarities with G. uvarioides King and G. donnaiensis Finet & Gagnep. but is differentiated by having 22–30 (or more) secondary veins in the leaf, a 1–3‐flowered cyme, a ca 2–3 cm long pedicel, lanceolate and tomentose outer petals, stipitate obovoid‐oblong monocarps, and a single‐seeded monocarp.  相似文献

Herein, we present a novel imaging platform to study the biological effects of non-invasive radiofrequency (RF) electric field cancer hyperthermia. This system allows for real-time in vivo intravital microscopy (IVM) imaging of radiofrequency-induced biological alterations such as changes in vessel structure and drug perfusion. Our results indicate that the IVM system is able to handle exposure to high-power electric-fields without inducing significant hardware damage or imaging artifacts. Furthermore, short durations of low-power (< 200 W) radiofrequency exposure increased transport and perfusion of fluorescent tracers into the tumors at temperatures below 41°C. Vessel deformations and blood coagulation were seen for tumor temperatures around 44°C. These results highlight the use of our integrated IVM-RF imaging platform as a powerful new tool to visualize the dynamics and interplay between radiofrequency energy and biological tissues, organs, and tumors.  相似文献

Yellow fever virus (YFV) can induce acute, life-threatening disease that is a significant health burden in areas where yellow fever is endemic, but it is preventable through vaccination. The live attenuated 17D YFV strain induces responses characterized by neutralizing antibodies and strong T cell responses. This vaccine provides an excellent model for studying human immunity. While several studies have characterized YFV-specific antibody and CD8+ T cell responses, less is known about YFV-specific CD4+ T cells. Here we characterize the epitope specificity, functional attributes, and dynamics of YFV-specific T cell responses in vaccinated subjects by investigating peripheral blood mononuclear cells by using HLA-DR tetramers. A total of 112 epitopes restricted by seven common HLA-DRB1 alleles were identified. Epitopes were present within all YFV proteins, but the capsid, envelope, NS2a, and NS3 proteins had the highest epitope density. Antibody blocking demonstrated that the majority of YFV-specific T cells were HLA-DR restricted. Therefore, CD4+ T cell responses could be effectively characterized with HLA-DR tetramers. Ex vivo tetramer analysis revealed that YFV-specific T cells persisted at frequencies ranging from 0 to 100 cells per million that are detectable years after vaccination. Longitudinal analysis indicated that YFV-specific CD4+ T cells reached peak frequencies, often exceeding 250 cells per million, approximately 2 weeks after vaccination. As frequencies subsequently declined, YFV-specific cells regained CCR7 expression, indicating a shift from effector to central memory. Cells were typically CXCR3 positive, suggesting Th1 polarization, and produced gamma interferon and other cytokines after reactivation in vitro. Therefore, YFV elicits robust early effector CD4+ T cell responses that contract, forming a detectable memory population.  相似文献

Clonorchiasis is a parasitic disease of high public health importance in many countries in southeastern Asia and is caused by the Chinese liver fluke Clonorchis sinensis. However, the genetic structure and demographic history of its populations has not been sufficiently studied throughout the geographic range of the species and available data are based mainly on partial gene sequencing. In this study, we explored the genetic diversity of the complete 1560 bp cytochrome c oxidase subunit 1 (cox1) gene sequence for geographically isolated C. sinensis populations in Russia and Vietnam, to our knowledge for the first time. The results demonstrated low nucleotide and high haplotype differentiation within and between the two compared regions and a clear geographical vector for the distribution of genetic diversity patterns among the studied populations. These results suggest a deep local adaptation of the parasite to its environment including intermediate hosts and the existence of gene flow across the species’ range. Additionally, we have predicted an amino acid substitution in the functional site of the COX1 protein among the Vietnamese populations, which were reported to be difficult to treat with praziquantel. The haplotype networks consisted of several region-specific phylogenetic lineages, the formation of which could have occurred during the most extensive penultimate glaciations in the Pleistocene Epoch. The patterns of genetic diversity and demographics are consistent with population growth of the liver fluke in the late Pleistocene following the Last Glacial Maximum, indicating the lack of a population bottleneck during the recent past in the species’ history. The data obtained have important implications for understanding the phylogeography of C. sinensis, its host-parasite interactions, the ability of this parasite to evolve drug resistance, and the epidemiology of clonorchiasis under global climate change.  相似文献
