Summary Primary cultivated rabbit articular chondrocytes were immobilized in calcium alginate beads. Both free and entrapped cells were allowed to grow under normal conditions. After long-term immobilization, the cells still exhibited metabolic activities, patterns of division, synthesis and secretion of extracellular matrix macromolecules such as type II collagen and proteoglycans. After 38 days, immobilized rabbit articular chondrocytes predominantly expressed type II but not type I collagen. Thus, they maintained their cartilage phenotype. After bead lysis, harvested cells showed normal growth patterns when resuspended in culture medium. Vascular endothelial growth factor (VEGF) is an endothelial cell mitogen which stimulates angiogenesis. VEGF is regulated by multiple factors such as hypoxia, phorbol esters, and growth factors. However, data concerning the expression of VEGF in the different vascular cell types and its regulation by cAMP are not available. In the present study, we have investigated the effect of adenylate cyclase activation on VEGF mRNA expression in rat vascular cells in primary culture. Basal VEGF expression is greater in smooth muscle cells than in endothelial cells and fibroblasts. A 4-h treatment with forskolin (10−5M) induced a 2-fold stimulation of VEGF mRNA expression in smooth muscle cells and fibroblasts, but, in contrast, did not affect VEGF expression in endothelial cells. In smooth muscle cells, a pharmacologically induced increase in intracellular cAMP levels using iloprost or isoprenaline led to a rise in VEGF mRNA expression comparable to that induced by forskolin. Adenosine, which increases cAMP levels in smooth muscle cells, also increases VEGF expression. Moreover, the 2.2-fold stimulation of VEGF expression by adenosine was enhanced following a cotreatment with cobalt chloride (a hypoxia miming agent). The observed additive effect (4.3-fold increase) suggests that these two factors, hypoxia and adenosine, regulate VEGF mRNA expression in smooth muscle cells by independent mechanisms.  相似文献

Aim

Intraspecific genetic diversity is one of the pillars of biodiversity, supporting the resilience and evolutionary potential of populations. Yet, our knowledge regarding the patterns of genetic diversity at macroecological scales, so-called macrogenetic patterns, remains scarce, particularly in marine species. Marine habitat-forming (MHF) species are key species in some of the most diverse but also most impacted marine ecosystems, such as coral reefs and marine forests. We characterize the patterns and drivers of genetic diversity in MHF species and provide a macrogenetic baseline, which can be used for conservation planning and for future genetic monitoring programmes.

Methods

We analysed a database including genetic diversity estimates based on microsatellites in more than 9,000 georeferenced populations from 140 species, which belong to seven animal and plant taxa. Focusing on expected heterozygosity, we used generalized additive models to test the effect of latitude, taxon, and conservation status. We tested the correlation between the species richness and the genetic diversity.

Results

We reveal a significant but complex biogeographic pattern characterized by a bimodal latitudinal trend influenced by taxonomy. We also report a positive species genetic diversity correlation at the scale of the ecoregions. The difference in genetic diversity between protected and unprotected areas was not significant.

Main conclusions

The contrasting results between MHF animals and plants suggest that the latitudinal genetic diversity patterns observed in MHF species are idiosyncratic, as reported in terrestrial species. Our results support the existence of shared drivers between genetic and species diversities, which remain to be formally identified. Concerning, these macrogenetic patterns are not aligned from the existing network of marine protected areas. Providing the first macrogenetic baseline in MHF species, this study echoes the call regarding the need to consider genetic diversity in biodiversity assessments and management.  相似文献

Summary CoSTIC, the French HVAC Contractors' Technical Institute, investigated the rules and difficulties in designing, fitting and maintaining healthy air conditioning systems. Precautions should be taken in design, notably regarding air intakes, filters, humidifiers and especially ease of access to equipment, yet a thorough commissioning and good maintenance are equally important. However, technicians have to live with various difficulties, particularly the tendency of the owner to save on air conditioning and the tendering procedure for the works. Potassium translocation in plants is accomplished by a large variety of transport systems. Most of the available molecular information on these proteins concerns voltage-gated potassium channels (Kv channels). The Arabidopsis genome comprises nine genes encoding α-subunits of Kv channels. Based on knowledge of their animal homologues, and on biochemical investigations, it is broadly admitted that four such polypeptides must assemble to yield a functional Kv channel. The intrinsic functional properties of Kv channel α-subunits have been described by expressing them in suitable heterologous contexts where homo-tetrameric channels could be characterized. However, due to the high similarity of both the polypeptidic sequence and the structural scheme of Kv channel α-subunits, formation of heteromeric Kv channels by at least two types of α-subunits is conceivable. Several examples of such heteromeric plant Kv channels have been studied in heterologous expression systems and evidence that heteromerization actually occurs in planta has now been published. It is therefore challenging to uncover the physiological role of this heteromerization. Fine tuning of Kv channels by heteromerisation could be relevant not only to potassium transport but also to electrical signaling within the plant.Key words: heteromerization, voltage-gated channels, membrane potential  相似文献

Therapeutic angiogenesis and myogenesis restore perfusion of ischemic myocardium and improve left ventricular contractility. These therapeutic modalities must be considered as complementary rather than competing to exploit their advantages for optimal beneficial effects. The resistant nature of cardiomyocytes to gene transfection can be overcome by ex vivo delivery of therapeutic genes to the heart using genetically modified stem cells. This review article gives an overview of different vectors and delivery systems in general used for therapeutic gene delivery to the heart and provides a critical appreciation of the ex vivo gene delivery approach using genetically modified stem cells to achieve angiomyogenesis for the treatment of infarcted heart.  相似文献
