We show that matrices carrying the tethered homologs of natural phosphoinositides can be used to capture and display multiple phosphoinositide binding proteins in cell and tissue extracts. We present the mass spectrometric identification of over 20 proteins isolated by this method, mostly from leukocyte extracts: they include known and novel proteins with established phosphoinositide binding domains and also known proteins with surprising and unusual phosphoinositide binding properties. One of the novel PtdIns(3,4,5)P3 binding proteins, ARAP3, has an unusual domain structure, including five predicted PH domains. We show that it is a specific PtdIns(3,4,5)P3/PtdIns(3,4)P2-stimulated Arf6 GAP both in vitro and in vivo, and both its Arf GAP and Rho GAP domains cooperate in mediating PI3K-dependent rearrangements in the cell cytoskeleton and cell shape.  相似文献

A study of laboratory-reared larvae of Solea solea (Soleidae) and Scophthalmus maximus (Scophthalmidae) indicated that the epicranial portion of the dorsal fin results from the anterior displacement of proximal pterygiophores during ontogeny. The adult epicranial formula is attained early during ontogeny, and the anterior displacement is finalized after the passage of the migrating eye. In both species, the first two proximal pterygiophores fuse to form an erisma that is particularly long and well-developed in Solea. Moreover, in Solea, the neural spine of the second abdominal vertebra curves over the otic region, and the neural arch of the first vertebra remains incomplete. Endocrine disruptors are characterized by their influence on animal endocrine systems resulting in reproductive, developmental, neurological, and immune dysfunction. The purpose of this overview is to provide the reader with a sense of the activities within the U.S. Environmental Protection Agency (USEPA), in particular NHEERL, that address the many facets of research on endocrine disrupting chemicals (EDCs) and to highlight the approach being taken at the different organizational levels within the USEPA, including screening, testing and evaluating endocrine disrupting chemicals. As a part of this endeavor, the USEPA continues to evaluate the current research activities in order to better understand and refine the process of risk characterization of EDCs. Thus, the participants in this session were asked to review their research within the framework of a better identification of EDC effects, better characterization of those compounds that have endocrine disrupting activity and how to incorporate this information into the risk assessment paradigm. Specifically, the goals of the ensuing papers were to compare individual vs. population indicators of endocrine disrupting effects, examine comparable and multiple mechanisms of toxicity, and describe the use of effects as indicators to identify toxicants and their sources. Mammalian and fish reproductive endpoints served as models to emphasize commonalities between human and wildlife risks.  相似文献

The identification of mutations in the NF1 gene causing type 1 neurofibromatosis (NF1) has presented a considerable challenge because of the large size of the gene, the lack of significant mutational clustering, the diversity of the underlying pathological lesions and the presence of NF1 pseudogenes. Denaturing high performance liquid chromatography (DHPLC), a high throughput, non-hazardous and largely automated heteroduplex-based technique, is in many ways ideally suited to mutation detection in this condition. DHPLC was therefore optimised for the rapid screening of the 60 exons and splice junctions of the NF1 gene in patients with NF1. The sensitivity of DHPLC was evaluated in a retrospective study of a cohort of 111 unrelated NF1 patients with known germline mutations; 97% of mutations were detected. In a subsequent prospective analysis of 50 unrelated NF1 patients, germline mutations were identified in 34 individuals (68%), 22 of these alterations being novel. This represents the highest rate of mutation detection so far reported for the NF1 gene with a single screening technique and genomic DNA as a target.  相似文献

Soybean genomic survey: BAC-end sequences near RFLP and SSR markers.   总被引:8,自引:0,他引:8  
We are building a framework physical infrastructure across the soybean genome by using SSR (simple sequence repeat) and RFLP (restriction fragment length polymorphism) markers to identify BACs (bacterial artificial chromosomes) from two soybean BAC libraries. The libraries were prepared from two genotypes, each digested with a different restriction enzyme. The BACs identified by each marker were grouped into contigs. We have obtained BAC- end sequence from BACs within each contig. The sequences were analyzed by the University of Minnesota Center for Computational Genomics and Bioinformatics using BLAST algorithms to search nucleotide and protein databases. The SSR-identified BACs had a higher percentage of significant BLAST hits than did the RFLP-identified BACs. This difference was due to a higher percentage of hits to repetitive-type sequences for the SSR-identified BACs that was offset in part, however, by a somewhat larger proportion of RFLP-identified significant hits with similarity to experimentally defined genes and soybean ESTs (expressed sequence tags). These genes represented a wide range of metabolic functions. In these analyses, only repetitive sequences from SSR-identified contigs appeared to be clustered. The BAC-end sequences also allowed us to identify microsynteny between soybean and the model plants Arabidopsis thaliana and Medicago truncatula. This map-based approach to genome sampling provides a means of assaying soybean genome structure and organization.  相似文献

Relationship between postextrasystolic potentiation and slow-phase force-frequency response in guinea-pig ventricular myocardium. Acta physiol. pol. 1985, 36 (2): 175-184. We investigated relationship between postextrasystolic potentiation (PESP) and a form of slow-phase force-frequency relation, namely the post-rest recovery of contractile force (CF). We found that although the degree of post-rest recovery of CF determines CF of the potentiated beats, PESP does not affect the time-course of post-rest recovery, even if it is sustained by means of paired stimulation. Moreover, PESP may be completely or almost completely inhibited by caffeine in 10 mmolar concentration, whereas post-rest recovery is not significantly affected by this drug. We conclude, that PESP and slow force-frequency relations are based on separate cellular mechanisms.  相似文献

The decatenation activity of topoisomerase II (Top2), which is widely conserved within the eukaryotic domain, is essential for chromosomal segregation in mitosis. It is less clear, however, whether Top2 performs the same function uniformly across the whole genome, and whether all its functions rely on decatenation. In the fission yeast, Schizosaccharomyces pombe, telomeres are bound by Taz1, which promotes smooth replication fork progression through the repetitive telomeric sequences. Hence, replication forks stall at taz1Δ telomeres. This leads to telomeric entanglements at low temperatures (⩽20°C) that cause chromosomal segregation defects and loss of viability. Here, we show that the appearance of entanglements, and the resulting cold sensitivity of taz1Δ cells, is suppressed by mutated alleles of Top2 that confer slower catalytic turnover. This suppression does not rely on the decatenation activity of Top2. Rather, the enhanced presence of reaction intermediates in which Top2 is clamped around DNA, promotes the removal of telomeric entanglements in vivo, independently of catalytic cycle completion. We propose a model for how the clamped enzyme–DNA complex promotes proper chromosomal segregation.  相似文献
