Cystatins, a family of structurally related cysteine proteinase inhibitors, have proved to be useful model system to study amyloidogenesis. We have extended previous studies of the kinetics of amyloid-fibril formation by human stefin B (cystatin B) and some of its mutants, and proposed an improved model for the reaction. Overall, the observed kinetics follow the nucleation and growth behavior observed for many other amyloidogenic proteins. The minimal kinetic scheme that best fits measurements of changes in CD and thioflavin T fluorescence as a function of protein concentration and temperature includes nucleation (modeled as N(I) irreversible transitions with equivalent rates (k(I)), which fitted with N(I) = 64), fibril growth and nonproductive oligomerization, best explained by an off-pathway state with a rate-limiting escape rate. Three energies of activation were derived from global fitting to the minimal kinetic scheme, and independently through the fitting of the individual component rates. Nucleation was found to be a first-order process within an oligomeric species with an enthalpy of activation of 55 +/- 4 kcal mol(-1). Fibril growth was a second-order process with an enthalpy of activation (27 +/- 5 kcal mol(-1)), which is indistinguishable from that of tetramer formation by cystatins, which involves limited conformational changes including proline trans to cis isomerization. The highest enthalpy of activation (95 +/- 5 kcal mol(-1) at 35 degrees C), characteristic of a substantial degree of unfolding as observed prior to domain-swapping reactions, equated with the escape rate of the off-pathway oligomeric state.  相似文献

Most population genetic theories on the evolution of sex or recombination are based on fairly restrictive assumptions about the nature of the underlying fitness landscapes. Here we use computer simulations to study the evolution of sex on fitness landscapes with different degrees of complexity and epistasis. We evaluate predictors of the evolution of sex, which are derived from the conditions established in the population genetic literature for the evolution of sex on simpler fitness landscapes. These predictors are based on quantities such as the variance of Hamming distance, mean fitness, additive genetic variance, and epistasis. We show that for complex fitness landscapes all the predictors generally perform poorly. Interestingly, while the simplest predictor, ΔVarHD, also suffers from a lack of accuracy, it turns out to be the most robust across different types of fitness landscapes. ΔVarHD is based on the change in Hamming distance variance induced by recombination and thus does not require individual fitness measurements. The presence of loci that are not under selection can, however, severely diminish predictor accuracy. Our study thus highlights the difficulty of establishing reliable criteria for the evolution of sex on complex fitness landscapes and illustrates the challenge for both theoretical and experimental research on the origin and maintenance of sexual reproduction.  相似文献

Maintaining the cholesterol homeostasis is essential for normal CNS functioning. The enzyme responsible for elimination of cholesterol excess from the brain is cholesterol 24-hydroxylase (Cyp46). Since cholesterol homeostasis is disrupted following brain injury, in this study we examined the effect of right sensorimotor cortex suction ablation on cellular and temporal pattern of Cyp46 expression in the rat brain. Increased expression of Cyp46 at the lesion site at all post injury time points (2, 7, 14, 28 and 45 days post injury, dpi) was detected. Double immunofluorescence staining revealed colocalization of Cyp46 expression with different types of glial cells in time-dependent manner. In ED1+ microglia/macrophages Cyp46 expression was most prominent at 2 and 7 dpi, whereas Cyp46 immunoreactivity persisted in reactive astrocytes throughout all time points post-injury. However, during the first 2 weeks Cyp46 expression was enhanced in both GFAP+ and Vim+ astrocytes, while at 28 and 45 dpi its expression was mostly associated with GFAP+ cells. Pattern of neuronal Cyp46 expression remained unchanged after the lesion, i.e. Cyp46 immunostaining was detected in dendrites and cell body, but not in axons. The results of this study clearly demonstrate that in pathological conditions, like brain injury, Cyp46 displayed atypical expression, being expressed not only in neuronal cells, but also in microglia and astrocytes. Therefore, injury-induced expression of Cyp46 in microglial and astroglial cells may be involved in the post-injury removal of damaged cell membranes contributing to re-establishment of the brain cholesterol homeostasis.  相似文献

The purpose of the present longitudinal study was to explore distinctive anthropometric and physical performance characteristics of young soccer players between the age of 11 and 14 and to reveal the performance at the age of 11, which contributes to the later success. Male players of the best national male squads of the 'cadet league' (14 years of age; n = 26) were annually tested starting from the age of 11 for body size and composition, flexibility, power, coordination, and agility. Randomly selected untrained but physically active age-matched boys (n = 63) were also tested over 4 consecutive years. The results revealed no difference between 2 groups regarding the body size and composition (p > 0.05). The differences in flexibility emerged only at the later age, whereas the differences regarding the explosive power (as assessed by various jumps) were moderate and partly inconsistent. The most prominent advantage of the soccer players over the control subjects during the entire tested age period appeared to be movement agility and coordination (p < 0.01). Therefore, the explosive muscle power and, in particular, the agility and coordination characterize elite soccer players of 11-14 years of age but not the body size and body composition. In addition, the agility and coordination could be among the crucial factors of future success in 11-year-old players and, therefore, should be used for early selection.  相似文献

1. The biological effects of carnosine, a natural hydrophilic neuropeptide, on the reactive oxygen species (ROS) pathological generation are reviewed.2. We describe direct antioxidant action observed in the in vitro experiments.3. Carnosine was found to effect metabolism indirectly. These effects are reflected in ROS turnover regulation and lipid peroxidation (LPO) processes.4. During brain ischemia carnosine acts as a neuroprotector, contributing to better cerebral blood flow restoration, electroencephalography (EEG) normalization, decreased lactate accumulation, and enyzmatic protection against ROS.5. The data presented demonstrate that carnosine is a specific regulator of essential metabolic pathways in neurons supporting brain homeostasis under unfavorable conditions.  相似文献

The occurrence of apoptosis in mouse preimplantation embryos was analyzed using DNA staining (Hoechst 33342, PI) for the visualization of nuclear changes and by the comet assay, a single-cell gel electrophoresis assay, modified for the analysis of blastocysts. Mouse preimplantation embryos isolated 56 h after superovulation were cultured in vitro for 64 h. Apoptosis was induced by treatment with camptothecin and actinomycin D during the first 15 h of culture. After culture in vitro, a number of embryos were stained and analyzed using morphological criteria. The remaining embryos were examined using the comet assay for the detection of DNA fragmentation. The proportion of damaged embryos in experimental groups, in comparison to controls, was dependent on the dose of apoptosis inductor. At high doses (camptothecin, microg/ml and actinomycin D, 0.05 microg/ml) over 90% (chi-square test, P<0.001) of embryos had apoptotic comets, at medium doses (camptothecin, 0.01 microg/ml and actinomycin D, 0.005 microg/ml) comets appeared only in 30-70% of embryos (camptothecin, P<0.01 and actinomycin D, P<0.001). At low doses (camptothecin, 0.001 microg/ml and actinomycin D, 0.0005 microg/ml) the increase in damaged embryos was not statistically significant. Hoechst/PI staining showed a higher percentage of damaged blastomeres at high doses. Morphological changes correlated with the outcome of the comet assay. Our results show that comet assay is an appropriate method for studying apoptosis in preimplantation embryos, and it appears to be more sensitive than the classically used morphological analyses.  相似文献

This study compared physiological responses to 2 high-speed resistance training (RT) protocols in untrained adults. Both RT protocols included 12 repetitions for the same 6 exercises, only differing in continuous (1 x 12) or discontinuous (2 x 6) mode. For discontinuous mode, there was a 15-second rest interval between sets. We hypothesized that the 2 x 6 protocol was less physiologically demanding than the 1 x 12 protocol. Fifteen untrained adults randomly performed the protocols on 2 different days while heart rate (HR), blood lactate (BL), rate of perceived exertion (RPE), and concentric phase mean power (CPMP) were measured. Significantly lower values (mean +/- SE) were seen with the discontinuous protocol for exercise HR (119 +/- 5 vs. 124 +/- 5 b x min(-1)), BL (5.7 +/- 0.5 vs. 6.7 +/- 0.3 mMol/L), and RPE (5.4 +/- 0.3 vs. 5.8 +/- 0.4) (p < 0.05). CPMP tended to be higher in the discontinuous protocol, especially for the 2 last repetitions. The discontinuous protocol was significantly less physiologically demanding, although similar or higher CPMP values were obtained. These findings may help foster long-term adherence to RT in untrained individuals. However, future studies are needed to compare physiological adaptations induced by these 2 RT protocols.  相似文献

Equinatoxin II (EqtII) belongs to a unique family of 20-kDa pore-forming toxins from sea anemones. These toxins preferentially bind to membranes containing sphingomyelin and create cation-selective pores by oligomerization of 3-4 monomers. In this work we have studied the binding of EqtII to lipid membranes by the use of lipid monolayers and surface plasmon resonance (SPR). The binding is a two-step process, separately mediated by two regions of the molecule. An exposed aromatic cluster involving tryptophans 112 and 116 mediates the initial attachment that is prerequisite for the next step. Steric shielding of the aromatic cluster or mutation of Trp-112 and -116 to phenylalanine significantly reduces the toxin-lipid interaction. The second step is promoted by the N-terminal amphiphilic helix, which translocates into the lipid phase. The two steps were distinguished by the use of a double cysteine mutant having the N-terminal helix fixed to the protein core by a disulfide bond. The kinetics of membrane binding derived from the SPR experiments could be fitted to a two-stage binding model. Finally, by using membrane-embedded quenchers, we showed that EqtII does not insert deeply in the membrane. The first step of the EqtII binding is reminiscent of the binding of the evolutionarily distant cholesterol-dependant cytolysins, which share a similar structural motif in the membrane attachment domain.  相似文献

The aim of the study was to investigate whether variables routinely assessed while testing athletes can also predict movement performance. The relation between jumping performance and standard strength, anthropometric, and body composition variables was examined in elite junior basketball players. The 33 males were tested for maximal vertical jump, as well as for maximal isometric voluntary force and rate of force development of hip and knee extensors. Standard anthropometric and body composition measures (body height, lean body mass, as well as the percentage of fat and muscle tissue) were also taken. Except for maximal isometric forces (0.38 and 0.52 N.kg(-1) for hip and knee extensors, respectively), all correlation coefficients between the selected variables and jump height were insignificant. As a consequence, the corresponding multiple correlation coefficient, R = 0.71, also suggested a moderate predictability of jumping performance by the standard strength tests and anthropometric and body composition variables. The results obtained dispute the use of the examined tests in sport performance assessment, and also question applying the tests for other purposes such as evaluation of training procedures or selection of young athletes. Therefore, the results are in line with the concept that a reliable performance assessment in homogeneous groups of athletes requires predominantly movement-specific testing.  相似文献

The obese strain (OS) of chickens, which suffers from spontaneous autoimmune thyroiditis, is an excellent animal model for Hashimoto's thyroiditis and provides a unique opportunity to investigate the mechanisms underlying and driving the onset of the disease. Following recent advances in cloning chicken cytokines, we can now begin to investigate the role of cytokines in driving the lymphoid infiltration of the thyroid seen in these birds from day 7 posthatch. Using real-time quantitative RT-PCR, we characterized the expression of IFN-gamma, IL-1beta, IL-2, IL-6, IL-8, IL-15, and IL-18 in thyroids from OS birds and control CB line birds, both in the embryo just before hatch (embryonic day 20) and at 3 and 5 days posthatch. All of these cytokines were up-regulated compared with levels in thyroids from CB birds, at least at some time points, with some evidence for coordination of regulation, e.g., for the proinflammatory cytokines IL-1beta and IL-8. Only IL-15 was up-regulated at all time points. IL-15 was also shown to be up-regulated in spleens of OS birds at embryonic day 20 and 5 days posthatch, suggesting that IL-15 is constitutively up-regulated in this line of birds. This could explain the general immune system hyperreactivity exhibited by OS chickens and may be a factor driving the lymphoid infiltration of the thyroid.  相似文献
