A common-garden study of Populus trichocarpa Torr. & Gray was established in spring 1986 with 128 clones collected from sites along two mesic (Hoh and Nisqually) and
two xeric (Dungeness and Yakima) river valleys in Washington. Two replicate plantations, one in Puyallup and the other in
Wenatchee, Wash., were established with this material. Over 2 years data were taken on stem growth, leaf/crown characters,
spring/autumn phenology, and the incidence of Melampsora occidentalis leaf rust. Combining clones from all four sources, correlation/regression analyses were used to examine clonal stability
of traits between test sites and trait relationships with stem growth; broad-sense heritabilities (H
2) and genetic correlations revealed the genetic strength of these traits. At Puyallup, many leaf/crown traits predicted stem
growth moderately to very well (r
2>0.50), e.g., total leaf area (TLA) to diameter gave an r
2 of 0.91 and current-terminal leaf size, of 0.79. Some regressions were quadratic, suggesting a threshold level in a trait
(e.g., leaf size) beyond which stem growth levels off. Upper-crown TLA was more closely related to height than TLA of the
lower sylleptics, but the reverse was true for diameter. A decline in r
2 values from upper to lower crown positions was sharper for correlations of TLAs with height than with diameter. Thus, leaf
area allocation seems to differentially affect stem growth. When autumn leaf fall (LF) and rust incidence (R) were regressed
with growth, r
2 values ranged from 0.58 to 0.71, but those of spring flush (SF) were only 0.10 to 0.12. Early LF and high R, both negatively
affecting growth, had a strong geographic component as it occurred mainly on lower-elevation Yakima clones. At Wenatchee,
field conditions were harsher and microsites more variable, so trait/growth relationships were weaker. Genetic correlations
with growth revealed similar trends as phenotypic analyses. Unlike leaf/crown traits, clonal scores of LF, SF, and R were
fairly stable across the two test sites (r
2: 0.58–0.80). These traits also showed strong genetic control (H
2: 0.96–0.98). The trait/growth relationships as well as trait stability within clones have implications for selecting clonal
stock in poplar culture and conservation.

The signal produced by fluorescence in situ hybridization (FISH) often is inconsistent among cells and sensitivity is low. Small DNA targets on the chromatin are difficult to detect. We report here an improved nick translation procedure for Texas red and Alexa Fluor 488 direct labeling of FISH probes. Brighter probes can be obtained by adding excess DNA polymerase I. Insect pests persist in a wide-variety of agricultural, arboreal and urban environments. Effective control with fungal entomopathogens
using inundation biocontrol requires an understanding of the ecology of the target insect, fungal pathogen, and the insect-pathogen
interaction. Historically, the development of production and formulation processes for biocontrol fungi has primarily focused
on reducing costs by maximizing the yield of infective propagules, increasing storage stability, and improving product form
for ease of application. These goals are critical for commercialization but are often in conflict with environmental and ecological
considerations. Critical parameters for selecting a fungal pathogen for use in inundation biocontrol include the cost-effective
production of a stable, infective propagule that is suited for use in the environment where the insect must be controlled.
Production processes can be manipulated nutritionally and environmentally to produce efficacious propagules or to direct fungal
differentiation to propagule forms that may be better suited for use in specific environments. Formulation development must
also consider ecological and environmental factors to maximize biocontrol efficacy. A basic understanding of the surface chemistries
of the fungal propagule and insect, the interactions between a fungal propagule and the insect cuticle that lead to infection,
and the impact of the environment on this interaction can aid in the development of effective formulations. 相似文献

The biosynthetic replacement of Met residues by selenomethionine (SeMet) facilitates the determination of three-dimensional structure by multiwavelength anomalous diffraction (Yang, W., Hendrickson, W. A., Crouch, R.J., and Satow, Y. (1990) Science 249, 1398-1405). In an effort to examine any biochemical effects due to the replacement of Met residues by SeMet, we chose to compare the kinetic and binding properties of selenomethionyl dihydrofolate reductase with those of the wt enzyme. There are 5 Met residues in Escherichia coli dihydrofolate reductase with 2 located in the Met-20 loop, which is a sequence of residues forming a lid over the active site. Utilizing plasmid pWT8, which affords 10-15% soluble protein as E. coli dihydrofolate reductase, we readily isolated both the SeMet and wt enzymes from E. coli DL41 utilizing a novel purification protocol. Both enzymes exhibited essentially the same kinetic and binding properties, including specific activities (45 mumol/min/mg), Km (7,8-dihydrofolate = 0.39 microM; NADPH = 2.0 microM), kcat (13.5/s), and 1:1 noncovalent inhibitory binding ratios with methotrexate. The inhibitory effects of divalent and monovalent cations on activity were also assessed, with the SeMet-containing enzyme exhibiting a uniformly greater sensitivity than the wt enzyme. We conclude that the biochemical properties of dihydrofolate reductase are virtually unperturbed by SeMet inclusion. Analysis of SeMet dihydrofolate reductase by 77Se nuclear magnetic resonance spectroscopy revealed five distinct resonances, thus indicating the potential value of this technique in employing selenium as a nonperturbing NMR probe of protein structure and function. 相似文献

Experimental animals often prefer small but immediate rewardseven when larger-delayed rewards provide a higher rate of intake.This impulsivity has important implications for models of foragingand cooperation. Behavioral ecologists have hypothesized thatanimals discount delayed rewards because delay imposes a collectionrisk. According to this long-standing hypothesis, delay reducesvalue because an interruption that occurs while an animal iswaiting may prevent it from collecting the delayed reward. Althoughthere have been many experimental demonstrations of animal preferencesfor immediacy, none have included any interruptions. This paperdevelops a simple model of discounting by interruptions andthen tests this model experimentally. The model considers theeffects of interruption rate and duration on choice behavior.The experiment tests the effects of interruptions on the choicebehavior of captive blue jays (Cyanocitta cristata) using afactorial design that manipulates the rate and duration of interruptions.The results do not support the discounting-by-interruptionshypothesis. Aldose reductase activity is increased in neuroblastoma cells grown in media containing 30 mM fructose and/or 30 mM glucose. Neuroblastoma cells cultured in media supplemented with increased concentrations of glucose and fructose amass greater amounts of sorbitol than do cells exposed to media containing only high glucose concentrations. The increase in sorbitol content is dependent on the fructose and glucose concentration in the media. The increase in sorbitol content caused by exposing neuroblastoma cells to media containing 30 mM glucose/30 mM fructose is due to a protein synthesis sensitive mechanism and not to an alteration in the redox state. The addition of sorbinil to media containing 30 mM glucose blocks the increase in sorbitol content. In contrast, sorbinil treatment of media containing 30 mM glucose/30 mM fructose does not totally block the increase in sorbitol levels. myo-Inositol accumulation and incorporation into inositol phospholipids and intracellular myo-inositol content are decreased in cells chronically exposed to media containing 30 mM glucose or 30 mM glucose/30 mM fructose compared to cells cultured in unsupplemented media or media containing 30 mM fructose. However, maximal depletion of myo-inositol accumulation and intracellular content occurs earlier in cells exposed to media containing 30 mM glucose/30 mM fructose than in cells exposed to media supplemented with 30 mM glucose. Sorbinil treatment of media containing 30 mM glucose/30 mM fructose maintains cellular myo-inositol accumulation and incorporation into phospholipids at near normal levels. myo-Inositol content in neuroblastoma cells chronically exposed to media containing 30 mM glucose or 30 mM glucose/30 mM fructose recovers within 72 h when the cells are transferred to unsupplemented media or media containing 30 mM fructose. In contrast, the sorbitol content of cells previously exposed to media containing 30 mM glucose or 30 mM glucose/30 mM fructose then transferred into media containing 30 mM fructose remains elevated compared to the sorbitol content of cells transferred into unsupplemented media. These data suggest that fructose may be activating or increasing sorbinil-resistant aldose reductase activity as well as partially blocking sorbitol dehydrogenase activity. The presence of increased concentrations of fructose in combination with increased glucose levels may enhance alterations in cell metabolism and properties due to increased sorbitol levels. 相似文献
