Lesser prairie-chickens (Tympanuchus pallidicinctus) have been declining range wide since the early 1900''s despite efforts to establish conservation and improve their habitat. In early 2014, the lesser prairie-chicken was listed as a threatened species under the U.S Endangered Species Act and the need to find out why they are declining is more important than ever. Nine hunter shot lesser prairie-chickens were donated and sampled for the presence or absence of the eyeworm Oxyspirura petrowi, a known parasite that can cause damage to the eye of its host, and common environmental contaminants. Eyeworm infection was found in 7 of 9 birds (78% infection rate) with an infection range between 0–16 O. petrowi per bird. Breast, liver, and fat tissue samples from the lesser prairie-chickens were analyzed for the frequency of 20 organochlorine pesticides. Femurs and livers were also tested on these birds for metal contaminants. Pesticides were found in several samples above the detection limits but were still in the low ng/g range. Notable was the ubiquitous presence of endrin aldehyde across all tissues. One femur showed 5.66 µg/g of lead (Pb) but this is still relatively low. No liver samples had elevated mercury (Hg) above detection limits. The presence of these organochlorines is consistent with the historic use of pesticides in this region. With pesticide and metals found in such low levels and parasitic nematode infections at rather high levels, it is recommended that these parasites be further evaluated as a contributing factor to the decline of the lesser prairie-chicken.  相似文献

The immunodeficiency that follows HIV infection is related to the virus-mediated killing of infected CD4(+) T cells, the chronic activation of the immune system, and the impairment of T cell production. In this study we show that in HIV-infected individuals the loss of IL-7R (CD127) expression defines the expansion of a subset of CD8(+) T cells, specific for HIV as well as other Ags, that show phenotypic (i.e., loss of CCR7 and CD62 ligand expression with enrichment in activated and/or proliferating cells) as well as functional (i.e., production of IFN-gamma, but not IL-2, decreased ex vivo proliferative potential and increased susceptibility to apoptosis) features of effector T cells. Importantly, in HIV-infected individuals the levels of CD8(+)CD127(-) T cells are directly correlated with the main markers of disease progression (i.e., plasma viremia and CD4(+) T cell depletion) as well as with the indices of overall T cell activation. In all, these results identify the expansion of CD8(+)CD127(-) effector-like T cells as a novel feature of the HIV-associated immune perturbation. Further studies are thus warranted to determine whether measurements of CD127 expression on CD8(+) T cells may be useful in the clinical management of HIV-infected individuals.  相似文献

Two morphine prodrugs (‘PDA’ and ‘PDB’) were synthesized and the kinetics of esterase-mediated morphine release from these prodrugs were determined when incubated with plasma from different animal species. Morphine was rapidly released from PDA by all species plasma with the maximum reached within 5–10 min; the released morphine was biologically active as determined by an in vitro cAMP assay. The morphine was released from PDB at a slower and species-dependent rate (mouse > rat > guinea pig > human). Morphine’s release from PDB appeared to be mediated by carboxyl esterases as the release was inhibited by the carboxyl esterase inhibitor benzil. PDA nor PDB induce cytotoxicity in the neuronal cell lines SK-NSH and SH-SY5Y. The carboxyl and amino functional moieties present on the linker portions of PDA and PDB, respectively, may facilitate their conjugation to nanoparticles to tailor morphine pharmacokinetics and specific targeting. These studies suggest the potential clinical utility of these prodrugs for morphine release at desired rates by administration of their mixture at selected ratios.  相似文献

Understanding the structural biology of type IV pili, fibres responsible for the virulent attachment and motility of numerous bacterial pathogens, requires a detailed understanding of the three-dimensional structure and chemistry of the constituent pilin subunit. X-ray crystallographic refinement of Neisseria gonorrhoeae pilin against diffraction data to 2.6 A resolution, coupled with mass spectrometry of peptide fragments, reveals phosphoserine at residue 68. Phosphoserine is exposed on the surface of the modelled type IV pilus at the interface of neighbouring pilin molecules. The site-specific mutation of serine 68 to alanine showed that the loss of the phosphorylation alters the morphology of fibres examined by electron microscopy without a notable effect on adhesion, transformation, piliation or twitching motility. The structural and chemical characterization of protein phosphoserine in type IV pilin subunits is an important indication that this modification, key to numerous regulatory aspects of eukaryotic cell biology, exists in the virulence factor proteins of bacterial pathogens. These O-linked phosphate modifications, unusual in prokaryotes, thus merit study for possible roles in pilus biogenesis and modulation of pilin chemistry for optimal in vivo function.  相似文献

In addition to their g = 1.94 EPR signal, nitrogenase Fe-proteins from Azotobacter vinelandii, Azotobacter chroococcum and Klebsiella pneumoniae exhibit a weak EPR signal with g approximately equal to 5. Temperature dependence of the signal was consistent with an S = 3/2 system with negative zero-field splitting, D = -5 +/- 0.7 cm-1. The ms = +/- 3/2 ground state doublet gives rise to a transition with geff = 5.90 and the transition within the excited ms = +/- 1/2 doublet has a split geff = 4.8, 3.4. Quantitation gave 0.6 to 0.8 spin . mol-1 which summed with the spin intensity of the S = 1/2 g = 1.94 line to roughly 1 spin/mol. MgATP and MgADP decreased the intensity of the S = 3/2 signal with no concomitant changes in intensity of the S = 1/2 signal.  相似文献

Evidence from the kinetics of transport supports the hypothesis that cellular Li, Lic, interacts with the internal aspect of the (Na,K)-pump as a congener of Na, Lic and Nac compete for the same sites on the internal aspect of the pump. Lic promotes Na-activated K influx and Nac promotes Li-activated K influx. Cellular K inhibits Li-activated K influx, indicating that the interaction of Kc with the internal aspect of the pump is qualitatively different from the interaction with either Nac or Lic. The Hill coefficients for Na-promoted and Li-promoted K influx are similar and are both greater than unity, indicating the same number of multiple intracellular sites per pump for the two cations. The stoichiometry of coupling between efflux and K influx is also similar for Na and Li, and is close to 3 Na or 3 Li to 2 K. The Li-activated K influx appears to be independent of the residual Na which remains in cells prepared in Na-free solutions.  相似文献

Understanding the impact of losing trophic diversity has global significance for managing ecosystems as well as important theoretical implications for community and ecosystem ecology. In several tropical forest ecosystems, habitat fragmentation has resulted in declines and local extinctions of mammalian and avian terrestrial insectivores. To assess the ability of a tropical rainforest community in Ivory Coast to resist perturbation from such loss of trophic diversity, I traced feedbacks in above and below ground communities and measured changes in nutrient levels and herbivory rates in response to an experimental exclosure of avian and mammalian terrestrial insectivores. I present evidence that loss of this functional group may result in increased tree seedling herbivory and altered nutrient regimes through changes in the abundance and guild structure of invertebrates. Exclusion of top predators of the forest floor resulted in increased seedling herbivory rates and macro-invertebrate (>5 mm) densities with strongest effects on herbivorous taxa, spiders and earthworms. Densities of microbivores including Collembola, Acarina and Sciaridae showed the opposite trend as did levels of inorganic phosphorus in the soil. Results were evaluated using path analysis which supported the presence of a top down trophic cascade in the detrital web which ultimately affected turnover of phosphorus, a limiting nutrient in tropical soils. Results illustrate the potential importance of vertebrate predators in both above and belowground food webs despite the biotic diversity and structural heterogeneity of the rainforest floor.  相似文献
