Concentrations of (E,E)-farnesol needed to inhibit germ tube formation were determined for Candida albicans strains A72 and SC5314 by using six different conditions known to trigger germination. For defined media, 1 to 2 μM farnesol was sufficient. However, with serum at 2 to 20%, up to 250 μM farnesol was required. Farnesol blocked germ tube formation but did not block elongation of existing germ tubes. 相似文献

Neurodegenerative diseases are characterized by progressive degeneration of selective neurones in the nervous system, but the underlying mechanisms involved in neuroprotection and neurodegeneration remain unclear. Dysfunction of the ubiquitin proteasome system is one of the proposed hypotheses for the cause and progression of neuronal loss. We have performed quantitative two-dimensional fluorescence difference in-gel electrophoresis combined with peptide mass fingerprinting to reveal proteome changes associated with neurodegeneration following 26S proteasomal depletion in mouse forebrain neurones. Differentially expressed proteins were validated by Western blotting, biochemical assays and immunohistochemistry. Of significance was increased expression of the antioxidant enzyme peroxiredoxin 6 (PRDX6) in astrocytes, associated with oxidative stress. Interestingly, PRDX6 is a bifunctional enzyme with antioxidant peroxidase and phospholipase A2 (PLA2) activities. The PLA2 activity of PRDX6 was also increased following 26S proteasomal depletion and may be involved in neuroprotective or neurodegenerative mechanisms. This is the first in vivo report of oxidative stress caused directly by neuronal proteasome dysfunction in the mammalian brain. The results contribute to understanding neuronal–glial interactions in disease pathogenesis, provide an in vivo link between prominent disease hypotheses and importantly, are of relevance to a heterogeneous spectrum of neurodegenerative diseases. 相似文献

The paper analyses migration flows from the Philippines in two gendered occupations: domestic helpers and computer programmers. The international division of labour theory claims that foreign investment determines migration from developing countries, especially of women, towards low-skilled gendered occupations in developed countries. This paper shows that the division of labour is neither gendered nor international in the predicted sense. For instance, data from Philippines Overseas Employment Agency shows that the theory is Eurocentric as Northern America and Europe are destinations for only 3 per cent of domestic workers’ flows. The paper argues that neo-Marxism creates bias in gender and migration research and hinders understanding of important gendered effects concerning migrants. Two examples of such gendered effects are highlighted here: the higher vulnerability to legislative change of migrant men employed as domestic workers in Italy and the higher penetration of women into computer programming in the migrant flows to the U.S. 相似文献

Sequence variation among 10 alleles of the alcohol dehydrogenase (Adh) gene
of the Hawaiian drosophilid D. mimica was analyzed with reference to the
evolutionary history of the Hawaiian subgroup as well as to levels and
patterns of polymorphism of the Adh gene in continental drosophilid
species. The Adh gene of D. mimica is less polymorphic than that of other
drosophilid species, and no replacement substitutions were found.
Statistical analyses of the Adh alleles suggested the action of balancing
selection and revealed significant linkage disequilibrium among three of
the variable sites. The effective population size was estimated to be only
slightly smaller than that of continental species and, surprisingly, on the
same order of magnitude as the actual size.

Previous studies indicated that signalling via CD95 and DR5 is greatly enhanced by the formation of ceramide-enriched membrane
platforms. Here, we employed this concept to convert doses of subtherapeutic TRAIL that were unable to release ceramide and
kill leukemic B-cells or ex vivo T lymphocytes, into a very effective apoptotic stimulus. Ceramide production was induced by application of sub-toxic doses
of doxorubicin that resulted in an activation of the acid sphingomyelinase (ASM), release of ceramide and formation of ceramide-enriched
membrane platforms. The latter served DR5 to cluster after application of very low doses of TRAIL in combination with doxorubicin.
Genetic deficiency of the ASM abrogated doxorubicin-induced ceramide release, as well as clustering of DR5 and apoptosis induced
by the combined treatment of doxorubicin and TRAIL. These data show that local release of ceramide potentiates very low, otherwise
inactive doses of TRAIL that may represent a novel therapeutic concept to treat tumors. 相似文献

Circular dichroism measurements in solution were used to confirm the enantiomeric nature of all twelve complexes. For crystal structures of 1 ‐ Λ‐ (S,S,S,S)‐M2+ or 1 ‐?‐ (R,R,R,R)‐M2+ complexes, absolute configurations {? (or P), Λ (or M)} were confirmed by refinement of the Flack parameter x: ?0.007 ≤ x ≤ 0.11 for the single crystals of 1 ‐Λ‐(S,S,S,S)‐M2+/ 1 ‐?‐ (R,R,R,R)‐M2+, 2 ‐ Λ‐ (S,S,S,S)‐Fe2+, and 2 ‐?‐ (R,R,R,R)‐Co2+. 相似文献

Dichlorvos (DDVP) is an organophosphorous pesticide with a high degree of dangerous effect towards the environment. We have
investigated the growth and susceptibility to DDVP of halophilic bacteria isolated from Romanian salt lakes. The growth of
four strains was affected by DDVP, which may be correlated with the rate constant values of DDVP disappearance from the saline
solutions. This is due not to a chemical degradation in solution but to the diffusion process and namely DDVP penetration
