The combination of NaCl and low temperature stress makes winter a critical time for evergreen halophytes at temperate latitudes. As part of a study of ecotypic differentiation of the evergreen Armeria maritima (Mill.) Willd., inland and salt-marsh populations were compared with respect to their growth and survival, osmotic adjustment and NaCl concentrations (based on dry weight) under salt stress in pot experiments in winter. Increased leaf necrosis in all populations under salt stress indicated a higher NaCl sensitivity in winter than in summer. Plants from inland populations were more sensitive than salt-marsh populations. Inland and salt-marsh populations showed similar capacities for seasonal osmotic adjustment, high seasonal increase of praline concentration and long-term betaine accumulation. Inland and salt-marsh populations allocated Na and Cl preferentially to the shoot. In inland populations, Na and Cl accumulated to high concentrations in leaves, whereas the much lower NaCl concentration in salt-marsh populations suggested that in these plants, Na uptake was regulated to match the growth-dependent ion demand of the shoot. The prevention of NaCl accumulation in times of slow growth by a NaCl homoeostasis system seemed to be an important adaptation with respect to the survival of evergreen plants in salt marshes.  相似文献

 An effective method has been developed for the stable transformation and regeneration of Cavendish banana (Musa spp. AAA group) cv 'Grand Nain' by microprojectile bombardment. Embryogenic cell suspensions were initiated using immature male flowers as the explant. Cells were co-bombarded with the neomycin phosphotransferase (nptII) selectable marker gene under the control of a banana bunchy top virus (BBTV) promoter or the CaMV 35S promoter, and either the β-glucuronidase (uidA) reporter gene or BBTV genes under the control of the maize polyubiquitin promoter. Plants were regenerated, under selection with kanamycin, that were co-transformed with nptII and either the uidA or BBTV genes. Molecular characterisation of transformants demonstrated that the transgenes had been stably integrated into the banana genome. BACKGROUND: Human MCF-7 cells have been studied extensively as a model for breast cancer cell growth. Many reports have established that serum-starved MCF-7 cells can be induced to proliferate upon the sole addition of 17beta-estradiol (E2). However, the extent of the mitogenic response to E2 varies in different MCF-7 strains and may even be absent. In this study we compared the E2-sensitivity of three MCF-7 laboratory strains. RESULTS: The MCF-7S line is non-responsive to E2, the MCF-7 ATCC has an intermediate response to E2, while the MCF-7 NKI is highly E2-sensitive, although the levels and activities of the estrogen receptor (ER) are not significantly different. Both suramin and IGF type I receptor blocking antibodies are able to inhibit the mitogenic response to E2-treatment in MCF-7 ATCC and MCF-7 NKI cells. From this we conclude that E2-induced proliferation is dependent on IGF type I receptor activation in all three MCF-7 strains. CONCLUSIONS: The results presented in this article suggest that E2-responsiveness of MCF-7 cells is dependent on the secretion of an autocrine factor activating the IGF-IR. All three strains of MCF-7 breast cancer cells investigated do not respond to E2 if the IGF-RI-pathway is blocked. Generally, breast cancer therapy is targeted at inhibiting estrogen action. This study suggests that inhibition of IGF-action in combination with anti-estrogen-treatment may provide a more effective way in treatment or even prevention of breast cancer.  相似文献

A series of 3-carbamoylpiperidines (nipecotamides) are designed, synthesized and tested for their inhibitory action against adenosine diphosphate (ADP)-induced aggregation of human platelets. A structure-activity analysis of the bis(nipecotamido)aralkane type showed that a substituent on the piperidine ring should preferably be an amide and that the electronegativity of the carbonyl oxygen and the orientation of the amide group affected activities. Based on the knowledge of factors influencing platelet activation and aggregation, a nitric ester moiety which could release nitric oxide (NO) in situ, is incorporated into the nipecotamide structure. These compounds exhibit increased activity compared to those having no -ONO2 function. They also show stereoselectivity, with the meso isomer being approximately twice as potent as the synthetic diastereomeric mixture. Replacement of the -ONO2 function with hydroxyl, ester or alkyl groups considerably diminishes aggregation-inhibitory potential. Nipecotamides are shown here to inhibit the basal and collagen-induced rise in platelet inositol trisphosphate (IP3) levels, as well as phosphoinositide turnover. A comprehensive mechanism of action is proposed taking earlier results into consideration.  相似文献

Contemporary genetic structure of Atlantic salmon (Salmo salar L.) in the River Moy in Ireland is shown here to be strongly related to landscape features and population demographics, with populations being defined largely by their degree of physical isolation and their size. Samples of juvenile salmon were collected from the 17 major spawning areas on the river Moy and from one spawning area in each of five smaller nearby rivers. No temporal allele frequency differences were observed within locations for 12 microsatellite loci, whereas nearly all spatial samples differed significantly, suggesting that each was a separate population. Bayesian clustering and landscape genetic analyses suggest that these populations can be combined hierarchically into five genetically informative larger groupings. Lakes were found to be the single most important determinant of the observed population structure. Spawning area size was also an important factor. The salmon population of the closest nearby river resembled genetically the largest Moy population grouping. In addition, we showed that anthropogenic influences on spawning habitats, in this case arterial drainage, can affect relationships between populations. Our results show that Atlantic salmon biodiversity can be largely defined by geography, and thus, knowledge of landscape features (for example, as characterized within Geographical Information Systems) has the potential to predict population structure in other rivers without an intensive genetic survey, or at least to help direct sampling. This approach of combining genetics and geography, for sampling and in subsequent statistical analyses, has wider application to the investigation of population structure in other freshwater/anadromous fish species and possibly in marine fish and other organisms.  相似文献

This Letter reports on adhesive modular proteins recorded by atomic force microscopy on live cells from the extracellular mucilage secreted from, and deposited around, the motile form of the pennate diatom Phaeodactylum tricornutum. This is the first report of modular proteins and their supramolecular assemblies, called adhesive nanofibers (ANFs), to be found on diatoms that use adhesives not only for substratum adhesion, but as a conduit for cell motility. The permanent adhesive pads secreted by Toxarium undulatum, a sessile centric diatom, were previously shown to possess ANFs with a modular protein backbone. Our results reported here suggest that modular proteins may be an important component of diatom adhesives in general, and that diatoms utilize the tensile strength, toughness, and flexibility of ANFs for multiple functions. Significantly, the genome of P. tricornutum has recently been sequenced; this will allow directed searches of the genome to be made for genes with modular protein homologs, and subsequent detailed studies of their molecular structure and function.  相似文献

We determined the effects of temperature and nutrients on thechain length of a diatom, Skeletonema costatum, in batch cultureand enclosure experiments with estuarine water from San FranciscoBay, USA, using the recently developed CytoBuoy flow cytometer.Determination of the number of cells per diatom chain by CytoBuoyflow cytometer and associated software correlated well withbut was much more precise and time efficient than microscopicquantification. Increasing temperatures (from 6, 8 to 17°C)and nutrient concentrations induced high growth rates and dominanceby longer chains in a cultured S. costatum strain that was originallyacclimatized to a temperature range of 11–30°C. Similarly,a positive correlation between growth rate and chain lengthwas observed in S. costatum in batch culture and natural communitiesin enclosure experiments. Maximal chain lengths of S. costatumwere greater in natural populations than in the batch culture.Longer chains affect sinking rates and thus likely help thediatom remain suspended in the upper part of the water columnwhere physical and chemical parameters are more favorable forgrowth.  相似文献

Bananas are a staple food source and a major export commodity worldwide. The Cavendish dessert banana is a triploid AAA genome type and accounts for around 47% of global production. Being essentially sterile, genetic modification is perhaps the only pathway available to improve this cultivar. In this study, we used the CRISPR/Cas9 gene editing system to deliver a self-cleaving polycistronic guide RNA (gRNA) designed to target exon 1 of the Phytoene desaturase (PDS) gene in the Cavendish cultivar “Williams”. Genotyping of 19 independent events showed a 100% PDS modification rate primarily in the form of insertions (1–105 nt) or deletions (1–55 nt) (indels) at the predicted cleavage site. Tri-allelic disruptive modifications were observed in 63% of plants and resulted in both albinism and dwarfing. Pale green (16%) and wildtype green (21%) phenotypes generally correlated with in-frame indels in at least one of the three PDS alleles. Editing efficiency was dependent on both target site selection and Cas9 abundance. This is the first report of a highly effective CRISPR/Cas9 modification system using a polycistronic gRNA in Cavendish banana. Such an editing platform will be of considerable utility for the development of disease resistance and novel agro-traits in this commercially important cultivar into the future.  相似文献

In Coleoptera, Neuroptera, and Megaloptera, and the panorpoid orders Diptera, Trichoptera, and Mecoptera the common oviduct is ventral to, or opens into, the vagina or genital chamber. In Lepidoptera, in the superfamilies Micropterigoidea, Eriocranioidea, Incurvarioidea, and Nepticuloidea, the common oviduct enters the copulatory chamber ventrally; in Mnesarchaeidae, Hepialoidea, and all Ditrysia auct. the common oviduct is dorsal to the copulatory chamber, and the vagina (that region posterior to the entry of the spermatheca) opens separately from the genital ostium. Lepidoptera are unique in the complex and various arrangements of the ectodermal elements in the female genitalia.

The arrangements of, and connections between rectal and genital structures in representatives of Megaloptera, Neuroptera, and the panorpoid orders are re‐examined for comparison with the systems found in Zeugloptera, Dacnonypha, Monotrysia, and Ditrysia auct.

The Zeugloptera are here included in Lepidoptera because they have a circumcloacal chamber. Other female zeuglopteran genital structures intergrade with those of Dacnonypha, here restricted to Eriocraniidae, Agathiphagidae, Lophocoronidae, and the divergent Acanthopteroctetes. Dacnonypha have a less specialised spermathecal and vaginal structure than do most of the Monotrysia, here restricted to Incurvarioidea and Nepticuloidea (including Tischeriidae); many Monotrysia lack a cloaca, whereas it is always present in Dacnonypha.

There is no basis for retaining either Mnesarchaeidae or Hepialoidea in Monotrysia auct., as the dorsal common oviduct and the two genital openings indicate that these groups are ditrysian. They are here regarded as exoporian Ditrysia, a group characterised by the lack of a free, tubular ductus seminalis. A fixed gutter or channel between ostium and ovipore characterises the Hepialoidea, and the absence of this channel (ostium and ovipore opposable within an external genital pouch) characterises Mnesarchaeidae.

The endoporian Ditrysia all have a free, tubular ductus seminalis; where a cloaca is present it is incomplete, i.e., combines ovipore and rectum but never copulatory structures, in contrast to the complete cloaca found in Zeugloptera, Dacnonypha, and many Monotrysia. The endoporian Ditrysia comprise all other superfamilies, i.e., about 97% of species of Lepidoptera.  相似文献
