Single crystals of the free base of guanosine 5'-monophosphate were X-irradiated at 10 and at 65 K and investigated between these temperatures and room temperature using K-band ESR and ENDOR spectroscopy. Three free radicals were detected in this temperature range. Two of these were identified as the O6-protonated anion radical and the C8 H-addition radical. Both of these species were present immediately after irradiation at 10 K. The anion radical was formed in two slightly different conformations, of which one decayed at about 150 K and the second at about 250 K. No successor radicals could be detected following the decay of the anion radical. The C8 H-adduct was stable at all temperatures used. The use of partially deuterated crystals confirmed the assignments made and showed that the main pathway for the formation of the C8 H-adduct consisted of addition of a proton from an easily exchangeable site. It is suggested that the C8 H-adduct is formed subsequent to a primary oxidation event localized either at the guanine base or at a nearby water of crystallization. Possible mechanisms for the formation of this product are discussed. 相似文献

A prominent hypothesis in the diversification of placental mammals after the Cretaceous–Palaeogene (K/Pg) boundary suggests that the extinction of non-avian dinosaurs resulted in the ecological release of mammals, which were previously constrained to small body sizes and limited species richness. This ‘dinosaur incumbency hypothesis’ may therefore explain increases in mammalian diversity via expansion into larger body size niches, that were previously occupied by dinosaurs, but does not directly predict increases in other body size classes. To evaluate this, we estimate sampling-standardized diversity patterns of terrestrial North American fossil mammals within body size classes, during the Cretaceous and Palaeogene. We find strong evidence for post-extinction diversity increases in all size classes. Increases in the diversity of small-bodied species (less than 100 g, the common body size class of Cretaceous mammals, and much smaller than the smallest non-avialan dinosaurs (c. 400 g)) were similar to those of larger species. We propose that small-bodied mammals had access to greater energetic resources or were able to partition resources more finely after the K/Pg mass extinction. This is likely to be the result of a combination of widespread niche clearing due to the K/Pg mass extinctions, alongside a suite of biotic and abiotic changes that occurred during the Late Cretaceous and across the K/Pg boundary, such as shifting floral composition, and novel key innovations among eutherian mammals. 相似文献

Summary The subcellular location of dehydrins was determined by immunomicroscopy in maize kernels imbibed in the presence of ABA. Antibodies were specific to the consensus sequence that exists near the carboxy terminus of virtually all dehydrins. Aleurone, scutellar epithelium, scutellar provascular strands, and the outermost embryonic leaves contained the highest concentration of dehydrins. In these tissues, and in scutellar parenchyma cells and inner embryonic leaves, dehydrins were present in the cytoplasm and associated with nuclei. In shoot and root apex cells, dehydrins appeared to be present principally in the cytoplasm. This localization pattern of dehydrins, together with the known amino acid sequences and phosphorylation patterns, leads to analogies with other proteins that are present in both the nucleus and cytoplasm of eukaryotic cells.Abbreviations ABA

Species of the Heterodoxus octoseriatus group infest five species, comprising eight chromosome races, of Petrogale in Queensland and northern New South Wales, Australia. The precise host and geographic ranges of the 11 species of the H. octoseriatus group were determined. Individual hosts and host populations were infested by single species of lice only. The geographic ranges of lice were discrete. In some cases the geographic ranges of louse species and their hosts were approximately congruent. In others they were completely incongruent and it was unclear which species originally infested which hosts or even which was the most recent colonizer. In at least one case, expansion of the geographic range of one species of louse apparently led to fragmentation of the range of another and subsequently, to allopatric speciation. 相似文献

Wolbachia are widespread endosymbionts that affect arthropod reproduction and fitness. Mostly maternally inherited, Wolbachia are occasionally transferred horizontally. Previously, two Wolbachia strains were reported at low prevalence and titres across seven Australian tephritid species, possibly indicative of frequent horizontal transfer. Here, we performed whole-genome sequencing of field-caught Wolbachia-positive flies. Unexpectedly, we found complete mitogenomes of an endoparasitic strepsipteran, Dipterophagus daci, suggesting that Wolbachia in the flies are linked to concealed parasitization. We performed the first genetic characterization of D. daci and detected D. daci in Wolbachia-positive flies not visibly parasitized, and most but not all Wolbachia-negative flies were D. daci-negative, presumably reflecting polymorphism for the Wolbachia infections in D. daci. We dissected D. daci from stylopized flies and confirmed that Wolbachia infects D. daci, but also found Wolbachia in stylopized fly tissues, likely somatic, horizontally transferred, non-heritable infections. Furthermore, no Wolbachia cif and wmk genes were detected and very low mitogenomic variation in D. daci across its distribution. Therefore, Wolbachia may influence host fitness without reproductive manipulation. Our study of 13 tephritid species highlights that concealed early stages of strepsipteran parasitization led to the previous incorrect assignment of Wolbachia co-infections to tephritid species, obscuring ecological studies of this common endosymbiont and its horizontal transmission by parasitoids. 相似文献

Immunolocalization using polyclonal antibodies raised against a conserved dehydrin amino acid sequence was used to establish the temporal and spatial patterns of dehydrin accumulation in embryo tissue of Zea mays L. (var. Ohio 43) kernels imbibed in the presence of abscisic acid. The temporal pattern of accumulation indicated an increase in dehydrins over time (particularly between 15 and 30 h) and with maximum levels detected 48 h after the onset of imbibition. Dehydrins were first evident, and also the most concentrated, in the cytosol throughout the accumulation period suggesting that the primary function of dehydrins involves the cytosol and the structures contained therein. Only after an accumulation of dehydrins in the cytosol was there an increase in the abundance of nuclear dehydrins. In addition, dehydrins were also observed in association with the proteinaceous matrix of protein bodies and membranes of protein and lipid bodies; these findings have not been reported previously. The observed localization at a number of sites indicates that the specific biochemical roles of dehydrins are likely to be diverse. 相似文献
