The Ecology of Temporary Streams I. The Faunas of Two Canadian Streams

This, the first of two papers on the ecology of temporary streams, describes, in detail, the faunas of two such habitats in southeastern Canada. The adaptive mechanisms for coping with the summer drought are documented for a variety of species and faunal succession is shown to occur from the stream stage (fall-winter) through the pool stage (spring) to the terrestrial (summer).     

Three-dimensional structures of the human α2-macroglobulin-methylamine and chymotrypsin complexes

The three-dimensional structures of chymotrypsin- and methylamine-treated negatively stained human α₂-macroglobulin have been determined by weighted back projection from electron microscope data. Projections of the reconstructions show good concordance with two-dimensional averages of both stained and frozen-hydrated molecules. The reconstructions reveal that the H-shaped front projection of the molecule is related to the smaller ellipsoidal end view by a rotation of 90° about the crossbar (minor axis) of the H. This finding is in agreement with tilt studies. The reconstruction of the α₂-macroglobulin-methylamine reveals an hour-glass shaped void which is filled by the two proteinase molecules in the reconstruction of α₂-macroglobulin-chymotrypsin. Protein plugs which appear to block the exterior entrances to the cavity may function to prevent access of proteins to the encapsulated proteinase and serve to block its escape. Extensive thresholding of each reconstruction leaves a “backbone” consisting of two side-by-side rod-like structures, suggesting that this is the arrangement of the two protomeric units which form the molecule. Both structures show some departure from the expected symmetry. The asymmetries are robust features of the reconstructions and may reflect structurally asymmetric features of the transformation from the native to the chymotrypsin-treated form of the molecule.     

An evaluation of methods for sampling macrophyte maximum colonisation depth in Loch Leven, Scotland

The maximum colonisation depth (MCD) of macrophytes in lakes is considered a robust ecological indicator of system health. The accuracy of five boat-based survey methods (double-headed rake, Ekman grab, bathyscope, hydroacoustics, and underwater photography) was evaluated along depth-transects within a shallow eutrophic loch (Loch Leven, Scotland, UK) in 2006. Data from an earlier field campaign conducted in 1993 (33 transects) were used to assess sample frequency and error associated with the use of the double-headed rake method. The double-headed rake consistently sampled more species than the Ekman grab or bathyscope, probably as a result of its ability to sample relatively larger areas of the lake bed in comparison to the other methods. The bathyscope method underestimated the MCD in comparison to all other methods. This appeared to be because the turbidity of the water-column restricted the depth of view when using the bathyscope. The double-headed rake and underwater photography methods were in good agreement, with maximum observed MCD estimates of 5.0 m and 4.5 m water depth, respectively (2006). Using the 1993 data (MCD observations taken from 33 selected transects), we estimated that over 5 transects were required to ensure a MCD estimate greater than the 75th percentile from the distribution of all 33 colonisation depths, more than 15 replicate transects were required to ensure an estimate greater than the 90th percentile, and more than 20 replicate transects were required to ensure an estimate greater than the 95th percentile.
