Immobilization stress applied for 6 h induced, in adult male rats, a rise of epinephrine (E) and norepinephrine (NE) plasma levels and a decrease of baseline plasma testosterone (T) values and of human chorionic gonadotropin (hCG)-induced T response. Treatment of the animals for 5 weeks with guanethidine (G), a sympathetic neuron toxic agent, significantly decreased E and NE responses to stress and partly antagonized the inhibitory effects exerted by immobilization on T biosynthesis. Adrenalectomy totally suppressed circulating E and reduced the stress-induced NE increase while partly antagonizing the inhibitory effects exerted on T biosynthesis. Combined G and adrenalectomy treatments totally suppressed plasma E and NE, and completely blocked the effects of immobilization on T levels. Treatment of the animals with the alpha 1-adrenergic blocker, prazosin, and the beta 1-adrenergic blocker, metoprolol, did not modify the effects of stress on T biosynthesis. Treatment with propranolol or with butoxamine, a nonspecific beta- and a specific beta 2-adrenergic receptor blocker, respectively, antagonized the testicular hyposensitivity to hCG induced by stress. Stress- or treatment-induced changes of plasma luteinizing hormone (LH) and hCG levels were not consistently correlated with plasma T modifications. These findings suggest that at least part of the inhibitory effects of immobilization stress on T biosynthesis is exerted by catecholamines through a beta 2-adrenergic receptor. 相似文献

Gonadotropins, prolactin (PRL), testosterone (T), delta 4-androstenedione, dehydroepiandrosterone sulfate and cortisol (F) levels were determined from 14 days before birth to term in 3 female and 3 male ovine fetuses with a chronically implanted venous catheter, and in the same animals from birth to 72 h of age. In both sexes, plasma gonadotropins and androgens were low throughout the period of study while plasma F increased with gestational age. After birth, plasma gonadotropins and PRL tended to increase progressively with time while PRL concentrations were significantly higher in female than in male lambs. F and T concentrations decreased significantly within the first 12 and 6 h of postnatal life. Higher T values were again observed at 36 h in male lambs. These data indicate that the fetal hypothalamic-pituitary-gonadal axis is relatively quiescent in the last 14 days of gestation but is activated within the first 72 h after birth. 相似文献

A number of highly potent PDE4 inhibitors are being developed for the treatment of asthma, chronic obstructive pulmonary disease, rheumatoid arthritis, multiple sclerosis and Crohn's disease. Cilomilast (Ariflo, SB 207499, SmithKline Beecham), the most advanced member of the class in Phase III clinical trials, was reported to have a limited therapeutic window. Other inhibitors with improved profiles in preclinical models are entering into (or are in) clinical trials. The recent developments in understanding PDE4 catalysis, inhibitor binding and their emetic response should facilitate the design of the next generation of PDE4 inhibitors. 相似文献

Background
Comparative sequence analysis of complex loci such as resistance gene analog clusters allows estimating the degree of sequence conservation and mechanisms of divergence at the intraspecies level. In banana (Musa sp.), two diploid wild species Musa acuminata (A genome) and Musa balbisiana (B genome) contribute to the polyploid genome of many cultivars. The M. balbisiana species is associated with vigour and tolerance to pests and disease and little is known on the genome structure and haplotype diversity within this species. Recurrence and progression to higher grade lesions are characteristic behaviorsof gliomas. Though IDH1 mutation frequently occurs and is considered as an early event in gliomagenesis, little is known about its role in the recurrence and progression of gliomas. We therefore analysed IDH1 and IDH2 statusat codon 132 of IDH1 and codon 172 of IDH2 by direct sequencing and anti-IDH1-R132H immunohistochemistry in 53 paired samples and their recurrences, including 29 low- grade gliomas, 16 anaplastic gliomas and 8 Glioblastomas. IDH1/IDH2 mutation was detected in 32 primarytumors, with 25 low- grade gliomas and 6 anaplastic gliomas harboring IDH1 mutation and 1 low- grade glioma harboring IDH2 mutation. All of the paired tumors showed consistent IDH1 and IDH2 status. Patients were analyzed according to IDH1 status and tumor-related factors. Malignant progression at recurrence was noted in 22 gliomas and was not associated with IDH1 mutation. Survival analysis revealed patients with IDH1 mutated gliomas had a significantly longer progression-free survival (PFS) and overall survival (OS). In conclusion, this study demonstrated a strong tendency of IDH1/IDH2 status being consistent during progression of glioma. IDH1 mutation was not a predictive marker for malignant progression and it was a potential prognostic marker for gliomas of Chinese patients. 相似文献

目的:探讨Orem自理模式在经尿道膀胱肿瘤电切患者护理中的应用,以Orem自护理论作为指导,对膀胱肿瘤患者电切术后实行护理干预,观察Orem自理模式对膀胱肿瘤电切术后患者生活质量及术后复发率的影响。方法:选择2008年10月至2012年6月我科经尿道膀胱肿瘤电切术患者160例随机分为两组,即对照组和实验组,各80例。其中对照组采用术后常规护理,实验组将Orem自理模式贯穿于常规护理的全过程。比较两组患者术后下床活动时间、住院时间、患者对相关知识的掌握情况及患者满意度。结果:实验组患者术后下床活动时间、住院时间、患者对疾病的认知度、患者满意度均明显高于对照组(P〈0.05)。结论iOrem自理模式应用于经尿道膀胱肿瘤电切术后患者的护理,训练患者及家属自我护理技术和能力,不仅能调动和激发经尿道膀胱肿瘤电切患者主观能动性,变“替代护理”为“自我护理”,而且能减少并发症的发生,提高患者的自理能力及生存质量,初步实现了对经尿道膀胱肿瘤电切患者教育的个体化、系统性、连贯性和重复性。 相似文献

An SAR campaign aimed at decreasing the overall lipophilicity of renin inhibitors such as 1 is described herein. It was found that replacement of the northern appendage in 1 with an N-methyl pyridone and subsequent re-optimization of the benzyl amide handle afforded compounds with in vitro and in vivo profiles suitable for further profiling. An unexpected CV toxicity in dogs observed with compound 20 led to the employment of a time and resource sparing rodent model for in vivo screening of key compounds. 目的:观察原儿茶酸对鱼藤酮诱导的帕金森模型小鼠脑组织中相关抗氧化酶活性的影响,为原儿茶酸治疗帕金森病提供理论依据。方法:选用昆明小鼠为实验动物,连续5周腹腔注射鱼藤酮(1mg/kg.d)建立帕金森模型,之后2周分别注射原儿茶酸(5mg/kg.d)和阳性药物美多芭(125mg/kg.d)。应用生物化学方法,观察原儿茶酸对帕金森模型小鼠中脑和纹状体内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活力以及脂质过氧化物丙二醛(MDA)含量的影响。结果:实验数据显示原儿茶酸可提高帕金森模型小鼠中脑和纹状体SOD、CAT、GSH-PX活力,降低MDA含量。同时病理切片染色显示原儿茶酸可减轻鱼藤酮诱导的脑组织损伤。结论:原儿茶酸具有神经保护作用,保护机制可能是提高脑组织内源性抗氧化酶活力,减少体内自由基的产生,进而减轻了脑组织的病理损伤。 相似文献
