The 1H-NMR spin-spin relaxation time (T2) in Triticale seeds swelling in external osmotica, polyethylene glycol 8000 or mannitol can identify both bound and free water. At the same water content, the free water spin-spin relaxation time increases for seeds imbibed with the mannitol solution, demonstrating inadequate water potential adjustment. The exchange rate of free/bound water molecules is apparently influenced by the driving force for water flow. The reciprocal lifetime of free water molecules, as a measure of water flow through the main cell barrier, was obtained. From a model of the seed as a resistance–capacitor network for water flow, a method was derived for calculating the reflection coefficient σ as a lifetime ratio of the free water molecules in seeds imbibed with two different osmotica (one penetrating across the main cell barrier and one not penetrating) at the same water potential. The 1H-NMR method and the classical method based on volume rate changes yielded reflection coefficients for mannitol for the cell wall–plasmalemma barrier of 0.78 ± 0.08 and 0.68 ± 0.06, respectively.  相似文献

Stallion semen was diluted in five different extenders (dimitro-poulus onze (Dimitro's), Kenney's modified tryode (Kenney's), modified INRA82 (INRA82), egg yolk-citrate-taurine (Citrate) and EZ-Mixin) and evaluated for motility after cooling and storage at 5 degrees C for 0, 24, 48, 72 and 96 h. EZ-Mixin extender was used as control while 70 and 100 mM of taurine were added to Dimitro's, Kenney's and INRA82 to study its effect under conditions of storage at 5 degrees C and varying processing modifications. Motility in INRA82 was 57.0, 58.4, 61.1, and 56.1% after 24, 48, 72 and 96 h, respectively and was higher (P < 0.05) than in other extenders after 48, 72, and 96 h. Motility decreased over time in Dimitro's (P < 0.05) and Kenney's (P < 0.01). Motility in INRA82 and EZ-Mixin decreased (P < 0.05) after 24 h and then remained unchanged until 96 h. In taurine (70 mM) containing extender Citrate, motility decreased throughout storage. Motility in INRA82 and Kenney's with taurine decreased (P < 0.05 and P < 0.01, respectively) after 48 h, and then remained stable until 96 h only in INRA82 with taurine. Motility in INRA82 with taurine was higher (P < 0.05) than in Citrate throughout incubation, whereas motility in INRA82 with taurine was higher than in EZ-Mixin and Kenney's with taurine after 24 and 48 h, respectively. Motility in INRA82 and Kenney's with taurine improved (P < 0.05) when osmotic pressure was increased with taurine (100 mM) but not when osmotic pressure was increased with Na (+) and K (+) salts. Motility was always higher (P < 0.01) in taurine (70 mM or 100 mM) containing extenders than in non-taurine extenders, Dimitro's, INRA82, and Kenney's, when sperm were incubated for 24 h at 5 degrees C in these extenders, then washed and incubated at 39 degrees C in Sp-TALP for 12 or 24 h. In conclusion 1) INRA82 was a better extender than the other extenders tested. 2) inclusion of taurine (100 mM) in INRA82 and Kenney's improved sperm survival until 96 and 48 h, respectively, and 3) sperm preincubation for 24 h in taurine containing extenders resulted in better sperm survival when washed and stored in Sp-TALP for further 12 or 24 h.  相似文献

The influence of varying illumination on rat hypothalamic, pituitary and prostatic steroid 5α-reductase (SR), body and prostatic weights and plasma FSH, LH and PRL levels was studied. Male rats weighing 50–60 g on arrival were divided as follows: controls (C) received 14 hours of daylight and 10 hours of darkness (14:10), constant light (CL) (24:0), and constant dark (CD) (0:24). After one month the animals were weighed, killed and the following tissues removed for analysis: prostate, hypothalamus, pituitary and blood. Prostates were weighed. Plasma concentrations of LH, FSH and PRL were determined by radioimmunoassay. Other tissues were analyzed for SR. CD animals gained less weight and had less heavy prostates/ 100 g body weight than either C or CL animals. Prostatic and hypothalamic SR activities were reduced following CD, while the pituitary enzyme was unaffected. Gonadotropin levels were unchanged in the CD group. Neither body nor prostatic weights were affected in CL treated animals. Prostatic SR activity increased following CL, while hypothalamic and pituitary enzymes remained unchanged. Plasma LH values were reduced in the CL group. FSH and PRL concentrations did not differ from controls. The prostate appeared most responsive to environmental lighting. It is suggested that observed changes in target organ (prostate) growth due to ambient lighting conditions are mediated via changes in SR.  相似文献

为了探讨植物叶片对UV-B辐射增强的响应机制,采用叶绿素荧光测定技术,分别测定在人工模拟低剂量UV-B(2.4μW/cm~2)辐射条件下乌拉尔甘草叶片的叶绿素荧光诱导动力曲线、初始荧光(F_0)、最大荧光(F_m)、光合机构比活性参数(ABS/RC、TR_o/RC和ET_o/RC)和性能指数等变化规律。结果表明:(1)低剂量UV-B辐射未引起甘草叶片O-J-I-P叶绿素荧光诱导曲线中的相数发生改变,UV-B辐射对PSⅡ的影响主要发生在其受体侧,而非供体侧;(2)低剂量UV-B辐射引起了甘草叶片光合系统F_v/F_m以及F_m、F_0的明显变化,同时也影响了光合机构的开放程度和电子从Q_A向Q_B传递效率,从而影响了光转化效率;相应性能指数(PI_(abs)和PI_(total))的改变亦验证了此结果。研究认为,低剂量UV-B辐射抑制乌拉尔甘草叶片光合系统Ⅱ受体侧Q_A至PQ之间的电子传递效率,从而影响了Q_A之后的光化学反应及非光化学反应。  相似文献

The aim of this study was to use whole body calorimetry to directly measure the change in body heat content (DeltaH(b)) during steady-state exercise and compare these values with those estimated using thermometry. The thermometry models tested were the traditional two-compartment model of "core" and "shell" temperatures, and a three-compartment model of "core," "muscle," and "shell" temperatures; with individual compartments within each model weighted for their relative influence upon DeltaH(b) by coefficients subject to a nonnegative and a sum-to-one constraint. Fifty-two participants performed 90 min of moderate-intensity exercise (40% of Vo(2 peak)) on a cycle ergometer in the Snellen air calorimeter, at regulated air temperatures of 24 degrees C or 30 degrees C and a relative humidity of either 30% or 60%. The "core" compartment was represented by temperatures measured in the esophagus (T(es)), rectum (T(re)), and aural canal (T(au)), while the "muscle" compartment was represented by regional muscle temperature measured in the vastus lateralis (T(vl)), triceps brachii (T(tb)), and upper trapezius (T(ut)). The "shell" compartment was represented by the weighted mean of 12 skin temperatures (T(sk)). The whole body calorimetry data were used to derive optimally fitting two- and three-compartment thermometry models. The traditional two-compartment model was found to be statistically biased, systematically underestimating DeltaH(b) by 15.5% (SD 31.3) at 24 degrees C and by 35.5% (SD 21.9) at 30 degrees C. The three-compartment model showed no such bias, yielding a more precise estimate of DeltaH(b) as evidenced by a mean estimation error of 1.1% (SD 29.5) at 24 degrees C and 5.4% (SD 30.0) at 30 degrees C with an adjusted R(2) of 0.48 and 0.51, respectively. It is concluded that a major source of error in the estimation of DeltaH(b) using the traditional two-compartment thermometry model is the lack of an expression independently representing the heat storage in muscle during exercise.  相似文献

Phenanthrene imidazole 3 (MF63) has been identified as a novel potent, selective, and orally active mPGES-1 inhibitor. This new series was developed by lead optimization of a hit from an internal HTS campaign. Compound 3 is significantly more potent than the previously reported indole carboxylic acid 1 with an A549 whole cell IC50 of 0.42 μM (50% FBS) and a human whole blood IC50 of 1.3 μM. It exhibited a significant analgesic effect in a guinea pig hyperalgesia model when orally dosed at 30 and 100 mg/kg.  相似文献

蒙古扁桃的植株可分为长花丝植株、短花丝植株和中花丝植株。群落花期约50d,单花花期约8d,分为露粉、微开、盛开、凋谢4个时期。过氧化物法测定4个时期花粉均具活力,可保持30d左右,联苯胺-过氧化氢测定柱头可授性,花粉活力与柱头可授性重叠,长花丝植株为8d左右,而中花丝约为5d。蒙古扁桃花一般在9:00开始泌蜜,11:00分泌量达到高峰,之后产蜜量减少直至停止,日泌蜜和散粉集中在10:00~14:00。蒙古扁桃开花受环境的影响。  相似文献
