Substances known to alter cyclic nucleotide levels in cells were applied to the isolated toad retina and effects on rod electrical and adaptive behavior were studied. The retina was continually superfused in control ringer’s or ringer’s containing one or a combination of drugs, and rod activity was recorded intracellularly. Superfusion with cGMP, Bu(2)GMP, isobutylmethylxanthine (IBMX; a phosphodiesterase inhibitor), or PGF(2α) (a prostaglandin) caused effects in rods that closely match those observed when extracellular Ca(2+) levels were lowered. For example, short exposures (up to 6 min) of the retina to these substances caused depolarization of the membrane potential, increase in response amplitudes, and some changes in waveform; but under dark-adapted or partially light-adapted conditions receptor sensitivity was virtually unaffected. That is, the position of the V-log I curve on the intensity axis was determined by the prevailing light level, not by drug level. These drugs, like lowered extracellular Ca(2+), also decreased the period of receptor saturation after a bright-adapting flash, resulting in an acceleration of the onset of membrane and sensitivity recovery during dark adaptation.
Long-term (6-15 min) exposure of a dark-adapted retina to 5 mM IBMX or a combination of IBMX and cGMP caused a loss of response amplitude and a desensitization of the rods that was similar to that observed in rods after a long-term low Ca(2+) (10(-9)M) treatment. Application of high (3.2 mM) Ca(2+) to the retina blocked the effects of applied Bu(2)cGMP. PGE(1) superfusion mimicked the effects of increasing extracellular Ca(2+). The results show that increased cGMP and lowered Ca(2+) produce similar alterations in the electrical activity of rods. These findings suggest that Ca(2+) and cGMP are interrelated messengers. We speculate that low Ca(2+) may lead to increased intracellular cGMP, and/or that applied cGMP, and/or that applied cGMP may lower cytosol Ca(2+), perhaps by stimulating Ca(2+)- ATPase pumps in the outer segment.

While larval sea lampreys exist as eyeless filter feeders for several years, they transform into free-swimming juveniles (transformers) that attach parasitically to prey fish as they develop sexual maturity. This study examines lamprey lens development and optics and, since the lens is often the only refractive component of an aquatic eye, the data also provide an indication of visual ability during transformer and adult periods of life. Seven adult sea lampreys (0.40–0.55 m) and eight transformers (0.15–0.18 m) were sacrificed, the eyes removed and lenses dissected, measured, and placed in an automated laser scanning instrument. Back vertex focal length (spherical aberration) was measured for 14 beam positions across each lens by using a digital camera to record the position of the refracted beam. Transformer lenses exhibit positive spherical aberration, with average focal lengths varying from about 2.40 mm near the lens center and 1.06 mm at the lens periphery. On the other hand, the lenses from adults are largely corrected for spherical aberration, with average focal lengths varying from 2.19 mm to 2.44 mm. This result indicates that the younger lenses do not have a gradient refractive index necessary to mitigate the aberration and that further study of this model may reveal the relation between lens embryology and the development of such a gradient. 相似文献
The adaptive function of male masturbation is still poorly understood, despite its high prevalence in humans and other animals. In non‐human primates, male masturbation is most frequent among anthropoid monkeys and apes living in multimale–multifemale groups with a promiscuous mating system. In these species, male masturbation may be a non‐functional by‐product of high sexual arousal or be adaptive by providing advantages in terms of sperm competition or by decreasing the risk of sexually transmitted infections. We investigated the possible functional significance of male masturbation using behavioral data collected on 21 free‐ranging male rhesus macaques (Macaca mulatta) at the peak of the mating season. We found some evidence that masturbation is linked to low mating opportunities: regardless of rank, males were most likely to be observed masturbating on days in which they were not observed mating, and lower‐ranking males mated less and tended to masturbate more frequently than higher‐ranking males. These results echo the findings obtained for two other species of macaques, but contrast those obtained in red colobus monkeys (Procolobus badius) and Cape ground squirrels (Xerus inauris). Interestingly, however, male masturbation events ended with ejaculation in only 15% of the observed masturbation time, suggesting that new hypotheses are needed to explain masturbation in this species. Examination of the thyroid hormone levels of C57BL/6J ob/ob mice and their lean littermates between the ages of 6 days and 22 weeks revealed that obese mice have significantly reduced hormone concentrations between 10 and 21 days of age. Thereafter, the values remained equal to, or above those or their lean littermates. The present data indicate that the hypometabolism and abnormal thermal regulation of weanling mice could be a result of diminished thyroid hormone levels, but that other mechanisms must be responsible for the persistence of these abnormalities in adult ob/ob mice. 相似文献
