This work presents a study of the effect of foliar and root application of low concentrations (0.1?C10 ??M) of potassium naphthenate on the antioxidative status of cucumber (Cucumis sativus L.), assessed for both local and systemic organs. Changes in the contents of proline and glutathione indicate that the treatment of plants with potassium naphthenate can be characterized as a mild abiotic stress. The antioxidative system of cucumber plants is sensitive to such treatment, since organs directly exposed to the chemical showed a decrease in total antioxidant activities and an increase in peroxidation. In the organs that were not directly treated, an increase in the total antioxidative activity was observed only at the lowest naphthenate concentration while at higher concentrations this activity tended to decrease. As far as the activities of antioxidant enzymes (guaiacol peroxidase, superoxide dismutase, catalase) are concerned, the responses observed differed between enzymes for a given treatment, but showed similar trends within treated local and untreated systemic organs.  相似文献

Obtaining a proper fold of affinity tagged chimera proteins can be difficult. Frequently, the protein of interest aggregates after the chimeric affinity tag is cleaved off, even when the entire chimeric construct is initially soluble. If the attached protein is incorrectly folded, chaperone proteins such as GroEL bind to the misfolded construct and complicate both folding and affinity purification. Since chaperonin/osmolyte mixtures facilitate correct folding from the chaperonin, we explored the possibility that we could use this intrinsic binding reaction to advantage to refold two difficult-to-fold chimeric constructs. In one instance, we were able to recover activity from a properly folded construct after the construct was released from the chaperonin in the presence of osmolytes. As an added advantage, we have also found that this method involving chaperonins can enable researchers to decide (1) if further stabilization of the folded product is required and (2) if the protein construct in question will ever be competent to fold with osmolytes.  相似文献

The aim of this research was to examine the applicability of some chronic toxicological tests in the determination of exposure to xenobiotics present in concentrations below No Observed Adverse Effect Level (NOAEL) and below the detection limit of analytical instruments. In the present experiment tested chemicals (Malathion, Aroclor-1254 and 3-methylcholanthrene (3-MC)) were mixed with wheat grains and given to male mice as feed over a period of 12 months. 7-ethoxyresorufin-O-deethylase (EROD) activity with the 3-MC and Aroclor-1254 treatments reached the peak at 9th month of exposure (26.7 and 42.4 pmol?1 mg(prot)-?1, respectively), while malathion did not have significant influence. Glutathione (GSH) level depletion was highest after three months of exposure. Unexpectedly, acetylcholinesterase (AChE) activity increased after treatment with malathion, an organophosphorous insecticide. In conclusion, low-level concentrations chronically administered exert certain effects on the levels of selected enzymes, e.g. biomarkers.  相似文献

Tyrosine phenol-lyase, a tetrameric pyridoxal 5'-phosphate dependent enzyme, catalyzes the reversible hydrolytic cleavage of L-tyrosine to phenol and ammonium pyruvate. Here we describe the crystal structure of the Citrobacter freundii holoenzyme at 1.9 A resolution. The structure reveals a network of protein interactions with the cofactor, pyridoxal 5'-phosphate, and details of coordination of the catalytically important K+ ion. We also present the structure of the apoenzyme at 1.85 A resolution. Both structures were determined using crystals grown at pH 8.0, which is close to the pH of the maximal enzymatic activity (8.2). Comparison of the apoenzyme structure with the one previously determined at pH 6.0 reveals significant differences. The data suggest that the decrease of the enzymatic activity at pH 6.0 may be caused by conformational changes in the active site residues Tyr71, Tyr291, and Arg381 and in the monovalent cation binding residue Glu69. Moreover, at pH 8.0 we observe two different active site conformations: open, which was characterized before, and closed, which is observed for the first time in beta-eliminating lyases. In the closed conformation a significant part of the small domain undergoes an extraordinary motion of up to 12 A toward the large domain, closing the active site cleft and bringing the catalytically important Arg381 and Phe448 into the active site. The closed conformation allows rationalization of the results of previous mutational studies and suggests that the observed active site closure is critical for the course of the enzymatic reaction and for the enzyme's specificity toward its physiological substrate. Finally, the closed conformation allows us to model keto(imino)quinonoid, the key transition intermediate.  相似文献

Depression and anxiety are co-morbid condition in diabetes as disease-related psychological reactions on this chronic metabolic illness. This study was aimed to determine the occurrence of depression and anxiety in seafarer's type 2 diabetic patients. A random sample of 52 diabetic seafarers treated with diet and oral glucose lowering agents, and 56 healthy seafarers were screened for depression with The Beck Depression Inventory (BDI) and for anxiety with State-Trait Anxiety Inventory (STAI 1, STAI 2). Depression (BDI > 18.5) and anxiety (STAI < 28.5) was significantly higher in the group of diabetic seafarers than in control group (more than 30%). Significant correlation was noted between depression and duration of diabetes mellitus, degree of obesity and poor glycaemic control (HbA1C > 8%) and longer duration of shipping routes (over 6 months). The proportion of depression and anxiety was found higher in seafarer's type 2 diabetic patients than in the healthy seafarers.  相似文献

Incidence rate of skin tumours, both, non-melanoma and melanoma, is increasing nowadays. Various etiological factors are of relevance for the occurrence of the diseases. The solar radiation, as well, long-term exposure to ultraviolet (UV) radiation, have the greatest impact on development of these skin tumours. Non-melanoma skin tumours, Basal Cell Carcinoma (BCC) and Squamous Cell Carcinoma (SCC), are the most common skin tumours in humans, and usually develop on the chronically photo-exposed areas. As for the Malignant Melanoma (MM), one of the most aggressive skin tumours, the exposure to solar radiation also plays an important role. This study investigates the correlation between the skin tumours and UV radiation in the area of West Herzegovina, on the sample of 1676 patients. It presents the occurrence of skin tumours in the period from 1997 to 2003. The study investigates the incidence and the risk factors separately for every skin tumour which can be etiologically related to the occurrence of skin tumours and UV radiation: occupation, exposure to UV radiation, skin type, and family history on malignan tumours within the patient's family. The exact incidence rate of non-melanoma and melanoma skin tumours in Bosnia and Herzegovina is still unknown, for the reason that the united National Cancer Register does not exist yet.  相似文献

We investigated the concentration of Aluminium (Al), Cobalt (Co), Chromium (Cr), Copper (Cu), Iron (Fe), Manganese (Mn), Nickel (Ni) and Zinc (Zn) in the root and aboveground organs of four halophyte species (Salicornia europaea, Suaeda maritima, Salsola soda and Halimione portulacoides), as well as in the soil from maritime and inland saline areas. The aim of our research was to evaluate the capability of some halophyte species to absorb different heavy metals and to detect differentiation of heavy metal accumulation within populations from inland and maritime saline areas. Generally, the plant roots had significantly higher concentrations of metals when compared to stems and leaves. Zinc was the only metal with concentrations significantly higher in the leaves than in the root and stem. Populations from maritime saline areas had higher trace root and stem metal concentrations than populations from inland saline areas. Excepting zinc, populations from inland saline areas had higher heavy metal concentrations in the leaves. The factors that affected metal accumulation by halophytes included the percentage of salt in the soil. We also discuss the potential use of these halophytes in phytoremediation.  相似文献

doi: 10.1111/j.1741‐2358.2011.00589.x Residual ridge atrophy in complete denture wearers and relationship with densitometric values of a cervical spine: a hierarchical regression analysis Background: The rate of residual ridge atrophy (RRR) and its association with mineral density of other bones have not yet been fully explained. Objective: To measure RRR over a 5‐year period in complete denture wearers and relate it to the density of a cervical spine (CSBD). Materials and methods: Sixty‐two patients (different gender, age, body mass index, duration of edentulousness (DE) and different denture‐wearing habits) participated. A copper stepwedge was attached to the cassette, and 50 lateral radiograms met the criteria to be included. Results: A significant decrease in vertical height was observed in all measured sites. The amount of RRR was highest in frontal areas of both jaws and decreased gradually towards lateral regions. Hierarchical regression analysis revealed that the amount of RRR in the maxillary frontal area could be explained up to 48.4% by the variable DE and only up to 6.1% by the CSBD, while gender had almost no influence (1%). Similar results were obtained for the lateral maxillary RRR (33.9%; 7%; 2%), frontal mandibular RRR (40; 8.4; 0.4%) and lateral mandibular RRR (31.5%; 3.4%; 7.7%). Conclusion: Skeletal bone density, reflecting systemic and hereditary factors, is weakly related to RRR (3.4–8.4%).  相似文献

We employed the self-organizing map (SOM) method to investigate the spatio-temporal pattern of the Chironomidae community in the Southern Morava River basin (Serbia) and to examine to what extent the Chironomidae community is affected by environmental factors. Additionally, this study explores the problems of utilizing chironomids in bioassessment programs. The SOM analysis of the chironomid community data produced 3 groups of sites. The indicator species analysis presented indicator taxa for two groups. Twenty taxa (at species, species group and genus level), according to the Kruskal–Wallis test, showed the most pronounced differences among the temporal units. Out of 15 measured environmental parameters, one-way ANOVA pointed out that 10 significantly differ between the groups. Elevation had the most important influence on the chironomid community, also affecting other environmental parameters. According to our findings, the winter season and the periods with high water level are the main sources of natural variability. To avoid such variability and to successfully incorporate Chironomidae in bioassessment programs, we suggest exclusion of the arguable months from monitoring programs.  相似文献
