The bottleneck in elucidating gene function through high-throughput gain-of-function genome screening is the limited availability of comprehensive libraries for gene overexpression. Lentiviral vectors are the most versatile and widely used vehicles for gene expression in mammalian cells. Lentiviral supernatant libraries for genome screening are commonly generated in the HEK293T cell line, yet very little is known about the effect of introduced sequences on the produced viral titer, which we have shown to be gene dependent. We have generated an arrayed lentiviral vector library for the expression of 17,030 human proteins by using the GATEWAY® cloning system to transfer ORFs from the Mammalian Gene Collection into an EF1alpha promoter-dependent lentiviral expression vector. This promoter was chosen instead of the more potent and widely used CMV promoter, because it is less prone to silencing and provides more stable long term expression. The arrayed lentiviral clones were used to generate viral supernatant by packaging in the HEK293T cell line. The efficiency of transfection and virus production was estimated by measuring the fluorescence of IRES driven GFP, co-expressed with the ORFs. More than 90% of cloned ORFs produced sufficient virus for downstream screening applications. We identified genes which consistently produced very high or very low viral titer. Supernatants from select clones that were either high or low virus producers were tested on a range of cell lines. Some of the low virus producers, including two previously uncharacterized proteins were cytotoxic to HEK293T cells. The library we have constructed presents a powerful resource for high-throughput gain-of-function screening of the human genome and drug-target discovery. Identification of human genes that affect lentivirus production may lead to improved technology for gene expression using lentiviral vectors.  相似文献

The first structurally characterised oxomolybdenum(V) complexes with thienyl carboxylate ligands were prepared by the reaction of [Mo2O3(C5H7O2)4] or (NH4)2[MoOCl5] with the corresponding acid (2-thiophenecarboxylic, 5-methyl-2-thiophenecarboxylic or 3-(3-thienyl)acrylic acid). Complexes [Mo2O3(μ-OC2H5)(μ-O2CR)(C5H7O2)2](R = -C4H3S (1), -C4H2S(CH3) (2) or -CHCHC4H3S (3)) were obtained upon substitution of two acetylacetonate ligands from [Mo2O3(C5H7O2)4] with RCOO in dry ethanol. Reactions of (NH4)2[MoOCl5] with the corresponding thienyl carboxylic acid in the presence of γ-picoline (C6H7N) yielded complexes (C6H7NH)[Mo2O4(μ-O2CR)Cl2(C6H7N)2] (R = -C4H3S (4), -C4H2S(CH3) (5) or -CHCHC4H3S (6)). All of the six new complexes were characterised as dinuclear. The molecular structures of 1, 3, 4·0.5CH3CN and 5 were determined by the single crystal X-ray diffraction method. In the complexes the two molybdenum atoms are doubly bridged either by one oxygen and one ethoxy-oxygen, or alternatively by two oxo-oxygens, and are additionally bridged by the thienyl carboxylate ion in a didentate bridging manner. All complexes were further characterised by means of chemical analysis, IR spectroscopy, TG and in some cases by the one and two-dimensional NMR method.  相似文献

Numerous white surface proliferations appeared in cultures of Ceratonia siliqua L. grown three to four weeks on medium containing 0.5 mg l-1 BA and 0.1 mg l-1 IBA. It was histologically confirmed that these proliferations were hypertrophied lenticels. Proliferations appeared first at the basal shoot internode and gradually spread acropetally, covering eventually the whole shoot except the uppermost internodes. Variations in 5HT-related genes contribute to the alterations of serotonergic neurotransmission, which is implicated in the etiopathology of alcoholism. In this preliminary study we have tested polymorphisms of genes involved in 5HT transport and turnover for their association with alcohol dependence. A case group of males with type 2 alcoholism (N=59) and a control group of healthy males (N=282), both of Croatian origin, were analyzed for the frequency distribution of polymorphisms in 5HT transporter (5HTT-VNTR2, 5HTT-LPR), monoamine oxidase A (MAOA-uVNTR) and B (MAOB-A/G) and tryptophan hydroxylase 1 (TPH1 A218C) and 2 (TPH2 G-703T) genes. An increase in the frequencies of 10-repeat allele (p = 0.010; OR = 1.73; 95% CI = 1.14-2.60) and 10/10 genotype (p = 0.006; OR = 2.57; 95% CI = 1.32-5.00) of the 5HTT-VNTR2 polymorphism was found in alcoholic patients. No differences between case and control groups were observed for the other tested polymorphisms. Present results support earlier studies implicating the role of 5HTT gene in alcoholism. The increase of sample size (in progress) is expected to enable search of more subtle differences, as well as re-evaluation of these preliminary findings.  相似文献

Increased platelet serotonin level (PSL) has been consistently found in a portion of autistic patients. Suggested mechanisms for hyperserotonemia in autism have been increased synthesis of serotonin (5HT) by tryptophan hydroxylase (TPH), increased uptake into platelets through 5HT transporter (5HTt), diminished release from platelets through 5HT2A receptor (5HT2Ar) and decreased metabolism by monoamine oxydase (MAOA). The allelic influence of genes, encoding the mentioned 5HT elements, on PSL was investigated in 63 autistic subjects. Our study shows that 5HTt-LPR and -1438AG 5HT(2Ar) genotypes did not significantly affect PSL. However, significantly higher PSLs were observed in subjects with "cc" genotype of a218c TPH and subjects with "4" genotype of uVNTR MAOA. In addition, when TPH-cc and MAOA-4 were combined as "high 5HT" genotypes, a correlative increase in PSL was observed with the increase in the number of "high 5HT" genotypes. These results suggest a possible synergistic effect of genes regulating 5HT synthesis/degradation in dysregulation of the peripheral 5HT homeostasis of autistic patients.  相似文献

The relationships and interactions within a methanotrophic-heterotrophic groundwater community were studied in a closed system (shake culture) in the presence of methane as the primary carbon and energy source and with the addition of the pure linear alkylbenzenesulfonate (LAS) congener 2-[4-(sulfophenyl)]decan as a cometabolic substrate. When cultured under different conditions, this community was shown to be a stable association, consisting of one obligate type II methanotroph and four or five heterotrophs possessing different nutritional and physiological characteristics. The results of experiments examining growth kinetics and nutritional relationships suggested that a number of complex interactions existed in the community in which the methanotroph was the only member able to grow on methane and to cometabolically initiate LAS transformation. These growth and metabolic activities of the methanotroph ensured the supply of a carbon source and specific nutrients which sustained the growth of four or five heterotrophs. In addition to the obligatory nutritional relationships between the methanotroph and heterotrophs, other possible interactions resulted in the modification of basic growth parameters of individual populations and a concerted metabolic attack on the complex LAS molecule. Most of these relationships conferred beneficial effects on the interacting populations, making the community adaptable to various environmental conditions and more efficient in LAS transformation than any of the individual populations alone.  相似文献

There is relatively little information on the role of high density lipoprotein (HDL) oxidation in atherogenesis although there are indications that oxidation might affect atheroprotective activities of HDL. Recently we reported the study on LDL oxidation initiated and sustained by traces of the transition metal ions under conditions, which favor slow oxidation. Here we report the results of the analogous study on the oxidation of the two HDL subclasses. The oxidation process was monitored by measuring the time dependence of oxygen consumption and concentration of the spin-trapped free radicals using EPR spectroscopy. In both HDL2 and HDL3 subclasses, the dependence of the oxidation process on the copper/lipoprotein molar ratio is different from that in LDL dispersions. Comparison of the kinetic profiles of HDL2 and HDL3 oxidation revealed that under all studied experimental conditions HDL2 was more susceptible to copper-induced oxidation than HDL3.  相似文献

piRNAs and Piwi proteins have been implicated in transposon control and are linked to transposon methylation in mammals. Here we examined the construction of the piRNA system in the restricted developmental window in which methylation patterns are set during mammalian embryogenesis. We find robust expression of two Piwi family proteins, MIWI2 and MILI. Their associated piRNA profiles reveal differences from Drosophila wherein large piRNA clusters act as master regulators of silencing. Instead, in mammals, dispersed transposon copies initiate the pathway, producing primary piRNAs, which predominantly join MILI in the cytoplasm. MIWI2, whose nuclear localization and association with piRNAs depend upon MILI, is enriched for secondary piRNAs antisense to the elements that it controls. The Piwi pathway lies upstream of known mediators of DNA methylation, since piRNAs are still produced in dnmt3L mutants, which fail to methylate transposons. This implicates piRNAs as specificity determinants of DNA methylation in germ cells.  相似文献

Serum sickness is immune response to a foreign antigen, usually a heterologous protein. Incidence rate is less than 0.5%. Antigens and responding antibodies form circulating immunocomplex that is characteristic for serum sickness. The condition occurs 7-15 days after exposure to the antigen, usually with clinical picture of glomerulonephritis. The immunocomplex circulates to other tissues where it sediments and causes inflammation, such as arteritis, neuritis, synovitis. The aim of this research is to present the break out of serum sickness in form of anterior uveitis due to azithromycin therapy administered by mouth. Identifying of anterior uveitis may help in early diagnostics and treatment of serum sickness.  相似文献

An investigation has been made within the expertise regarding teeth of 10 left halves of mandibles of recent sows as well as teeth of 18 pig mandibles originating from Kostolac culture (3,250-3,000 B.C.) from Vucedol archaeological location. TWS (Tooth Wear Stage) has been determined at all molars, according to Grant method and a corresponding numerical equivalent has been added, so MWS (Mandibular Wear Stage) has been obtained by summing up the above. On the basis of these values and ranges of MWS, an approximate age at the time of death has been determined for each unit. All the recent units have been older than 17 months, while the age of Kostolac units has come out to be between 14 and 17 months.  相似文献
