Summary A complex network of atrial natriuretic factor-producing cells has been delineated by biochemical and morphological techniques in the rat ventricular myocardium. The chordae tendineae spuriae (CTS; false tendons) contain ANF mRNA and the ANF propeptide (Asn 1-Tyr 126) as assessed by Northern blot analysis, high-pressure liquid chromatography and immunohisto- and -cytochemistry, using three different affinity-purified antibodies: monoclonal and polyclonal antibodies against C-terminal ANF (Arg 101-Tyr 126) and polyclonal antibodies against N-terminal ANF (Asp 11-Ala 37). Two types of cells harboring ANF-containing secretory granules constitute the CTS: the majority (Purkinje type I) have ultrastructural similarities with both atrial and classical Purkinje fibers. Purkinje type-II fibers resemble working ventricular cardiocytes. Both cell types harbor a large paranuclear Golgi complex. The subendocardial Purkinje network is also made up of these two cell types. In this location, Purkinje type-I fibers form cable-like structures while Purkinje type-II fibers are either located beneath the former or abut directly on the endocardium. The latter are not separated from adjacent working ventricular cardiocytes by connective tissue septa. Coronary arteries and arterioles, as in birds, are surrounded by a cushion of Purkinje type-II fibers which blend with the surrounding myocardium. Specificity of the effect of prostaglandins (PGs) on hormone release by the anterior pituitary gland was studied using cells in primary culture. Growth hormone (GH) release is stimulated by all eight PGs studied, PGE1 and E2 being 1000-fold more potent than the corresponding PGFs. The release of luteinizing hormone (LH), follicle-stimulating hormone (FSH), and prolactin (PRL) remains unchanged upon addition of PGEs. While the basal release of thyrotropin (TSH) is only slightly stimulated by concentrations of PGEs above 10−6M, an important potentiation of the stimulatory effect of thyrotropin-releasing hormone on TSH release is observed. The release of GH, TSH and LH is stimulated equally well by PGAs and PGBs at concentrations higher than 10−6M, 3 × 10−6M, and 10−5M, respectively. PGFs do not affect the release of any of the measured pituitary hormones at concentrations below 10−4M. The stimulation of GH release by PGE2 can be inhibited by the PG antagonist 7-oxa-13-prostynoic acid, a half-maximal inhibition being found at a concentration of 4 × 10−5M of the antagonist in the presence of 10−6M PGE2. In the presence of somatostatin (10−8M), the inhibition of GH release cannot be reversed by PGE2 at concentrations up to 10−4M. 8-bromo-cyclic AMP-induced GH release is additive with that produced by PGE2.The present data show that 1) of the five pituitary hormones measured, only GH release is stimulated by prostaglandins at relatively low concentrations, 2) the PGE-induced GH release can be competitively inhibited by 7-oxa-13-prostynoic acid, 3) the inhibition of GH release by somatostatin cannot be reversed by PGE2 and 4) the PGEs increase the responsiveness of the thyrotrophs to TRH. 相似文献

The authors have investigated the electrical activity of corpus cavernous, first according to Wagner and Gerstenberg’ method, then, since one year, according to Stief method, in the basal state and following intracavernosal injection of vasoactive agents, or following various stimulation tests. They have found again the electrical activity described by these authors, but have been confronted with the difficulty in quantifying the data. Specially single potential analysis seems to them little reliable and reproducible for an objective interpretation. At this stage of their experience, they test two simpler criteria interpretation: the richness of the electrical activity, and the reactivity of the recording to various stimulations. Their preminary results suggest a correlation between those criteria and the state of the autonomic nervous system of the penis. 相似文献

Electron microscopy (EM) provides much higher resolution than that obtained with light microscopy (LM). Until now, however, no chromosome banding procedure specifically adapted for EM was available. To produce an easy and reproducible banding method that would allow accurate chromosome identification, we investigated the applicability of an immunochemical method. BrdU-substituted chromosomal regions can be accurately visualized by applying a monoclonal antibody against BrdU, followed by a gold-tagged secondary antibody. Since BrdU is incorporated during the last part of the S-phase, regions of darkly stained G- and C-bands are substituted. A characteristic C-banding pattern is revealed, and the G-banding obtained is sharp and allows discrimination between subbands. Its similarity with the classical G-banding observed by LM makes it easy to interpret and facilitates karyotyping. 相似文献

Complete sequences of the rRNA genes of Drosophila melanogaster 总被引:19,自引:0,他引:19
In this, the first of three papers, we present the sequence of the
ribosomal RNA (rRNA) genes of Drosophila melanogaster. The gene regions of
D. melanogaster rDNA encode four individual rRNAs: 18S (1,995 nt), 5.8S
(123 nt), 2S (30 nt), and 28S (3,945 nt). The ribosomal DNA (rDNA) repeat
of D. melanogaster is AT rich (65.9% overall), with the spacers being
particularly AT rich. Analysis of DNA simplicity reveals that, in contrast
to the intergenic spacer (IGS) and the external transcribed spacer (ETS),
most of the rRNA gene regions have been refractory to the action of
slippage-like events, with the exception of the 28S rRNA gene expansion
segments. It would seem that the 28S rRNA can accommodate the products of
slippage-like events without loss of activity. In the following two papers
we analyze the effects of sequence divergence on the evolution of (1) the
28S gene "expansion segments" and (2) the 28S and 18S rRNA secondary
structures among eukaryotic species, respectively. Our detailed analyses
reveal, in addition to unequal crossing-over, (1) the involvement of
slippage and biased mutation in the evolution of the rDNA multigene family
and (2) the molecular coevolution of both expansion segments and the
nucleotides involved with compensatory changes required to maintain
secondary structures of RNA.

A 3-D nonlinear mathematical model is used to analyze the mechanical response of a lumbar L2-3 motion segment including the posterior elements when subjected to combined sagittal plane loads. The loadings consist of axial compression force, anterior and posterior shear forces, and flexion and extension moments. The facet articulation is modelled as a general moving contact problem and the ligaments as a collection of uniaxial elements. The disk nucleus is considered as an inviscid fluid and the annulus as a composite of collagenous fibers embedded in a matrix of ground substance. The presence of axial compression force reduces the segmental stiffness in flexion whereas a reverse trend is predicted in extension. In the presence of axial compression with and without sagittal shear force, flexion considerably increases the intradiscal pressure while extension reduces it. In other words, under an identical compression force, disk pressure is predicted to be noticeably larger in flexion than in extension. The segmental mechanical response in extension loadings is markedly influenced by the changes in the relative geometry of the articular surfaces at the lower regions. Finally, the deformation of the bony structures plays a significant role in the segmental mechanics under relatively large loads. 相似文献

Pro-natriodilatin (PND) is the precursor for atrial natriuretic peptide (ANP), a hormone which plays an important role in cardiovascular homeostasis. Since the effects of thyroid hormone (T3) on the cardiovascular and renal systems appear to mimic those elicited by ANP, we studied the effect of T3 on PND gene expression using rat neonatal cardiocytes in primary cultures. Treatment of cardiocytes for 48 h with T3 (5 X 10(-9) M) results in a maximal increase in PND mRNA levels; this increase is two fold in atrial and four fold in ventricular cell cultures. These results taken together with a previous report showing decreased plasma ANP in hypothyroid and increased plasma ANP in hyperthyroid rats suggest that at least some of the cardiovascular and renal effects of T3 may be mediated by a T3-dependent increase in PND gene expression. 相似文献
