In the Moroccan Atlas, sedimentary deposits provide important data on reef events that characterize the Jurassic period. Recent work allows us to enhance knowledge of the Jurassic reefs in the Atlas, in particular their age, character and palaeogeographic distribution. Numerous localities with sponge-microbial mud mounds, coral reefs, and lithiotid bioherms are recorded from the Middle and High Atlas regions. These different biogenic constructions occupy different palaeogeographic settings: on the top of tilted blocks within the basin center; in slightly deeper positions, at the basin platform junction; and on adjacent platforms in the middle of the coastal area. The main episodes of reef building span nearly 30 million years, as follows: (1) Sinemurian, (2) early Pliensbachian, (3) late Toarcian, (4) Aalenian–early Bajocian (pars), and (5) late Bajocian. These five distinct reef events can be linked with general fluctuations of sea level and tectonism, and have palaeoclimatic implications.  相似文献

Serpulid-dominated shell beds from the Lower Bajocian (Middle Jurassic) of the Central High Atlas (Morocco) are primarily formed by quadrangular tubes of the serpulid polychaete Nogrobs moroccensis sp. nov. The transverse tube ornament consisting predominately of wrinkles, the lack of narrow spirals and the shortness or absence of attached posterior tube portion represent the diagnostic features of this new species. Tubes of Nogrobs moroccensis co-occur with gastropods and oyster–crinoidal debris. We suggest that these serpulids were free-lying in their adult stage and formed benthic meadows on soft-bottom habitats. They frequently show parallel alignment in plane views in both graded and non-graded, few-cm-thick shell beds, corresponding to distal tempestites deposited close to and below storm wave base in an outer-ramp environment. The excellent preservation of serpulids implies short residence time on the seafloor, and the lack of compositional mixing with other communities and their restriction to outer-ramp habitats indicate that they represent parautochthonous assemblages. Analyses of serpulid orientation on the bottom bedding planes of shell beds revealed weak unidirectional arrangement of tubes, whereas orientation on the top bedding planes revealed strong unidirectional arrangement of tubes, with significantly different orientation relative to the bottom planes. This change could suggest a shift from predominantly a current component of the combined storm flow (with uneven seafloor surface and adhesion of skeletal grains to muddy substrate) to significant oscillatory component of the waning storm, as also suggested by bidirectional orientation of gastropods.  相似文献

The development of covalent drugs, specifically in cancer therapeutics, has recently sparked interest among the pharmaceutical research community. While representing a significant fraction of the drugs in the market, very few have been deliberately designed to interact covalently with their biological target. One of the enzymes that have been both covalently and noncovalently targeted is the Neural Precursor Cell Expressed Developmentally Downregulated gene 4-1 (Nedd4-1). This enzyme has been found to have multiple physiological implications, including its involvement in cancer invasion. A critical gap still remains in the molecular understanding of the structural mechanism upon the covalent and noncovalent binding to Nedd4-1. In this study, we explore the most optimal binding mechanism in the inhibition of the catalytic site of the Nedd4-1. Our results exhibited a greater stability in the covalent complex compared with the noncovalent complex. This was supported by the secondary structure elements that were more dominant in the covalently inhibited complex. This complex disclosed an optimal free binding energy landscape, induced by the catalytic site energy contributions that showed to be more favorable. The insights demonstrating the above binding mechanism of Nedd4-1 establishes covalent inhibition as the preferred method of inhibition of the enzyme. This investigation aids in the understanding of the structural mechanism of Nedd4-1 inhibition and would assist in the design of more potent covalent inhibitors at the catalytic site of Nedd4-1.  相似文献

Background/Purpose

A slight increase in the proportion of circulating regulatory T (Treg) cells has been reported in systemic lupus erythematosus (SLE) patients taking oral prednisone. The effects of intravenous (IV) high dose methylprednisolone (MP) on Tregs have not yet been described, especially in active SLE.

Methods

We prospectively analyzed the proportion of circulating CD4+ Treg cell subsets defined as follows: (1) naïve Treg (nTreg) FoxP3lowCD45RA+ cells; (2) effector Treg (eTreg) FoxP3highCD45RA cells; and (3) non-suppressive FoxP3lowCD45RA cells (non-regulatory Foxp3low T cells). Peripheral blood mononuclear cells of patients with active SLE were analyzed before the first infusion of IV high dose MP (day 0) and the following days (day 1, day 2, ±day 3 and ±day 8). The activity of SLE was assessed by the SLEDAI score.

Results

Seventeen patients were included. Following MP infusions, the median (range) percentage of eTregs significantly increased from 1.62% (0.53–8.43) at day 0 to 2.80% (0.83–14.60) at day 1 (p = 0.003 versus day 0), 4.64% (0.50–12.40) at day 2 (p = 0.06 versus day 1) and 7.50% (1.02–20.70) at day 3 (p = 0.008 versus day 2), and declined to baseline values at day 8. Expanding eTreg cells were actively proliferating, as they expressed Ki-67. The frequency of non-regulatory FoxP3low T cells decreased from 6.39% (3.20–17.70) at day 0 to 4.74% (1.03–9.72) at day 2 (p = 0.005); nTreg frequency did not change. All patients clinically improved immediately after MP pulses. The absence of flare after one year of follow up was associated with a higher frequency of eTregs at day 2.

Conclusion

IV high dose MP induces a rapid, dramatic and transient increase in circulating regulatory T cells. This increase may participate in the preventive effect of MP on subsequent flares in SLE.  相似文献

Plasmodium falciparum infection can cause microvascular dysfunction, cerebral encephalopathy and death if untreated. We have previously shown that high concentrations of free heme, and C-X-C motif chemokine 10 (CXCL10) in sera of malaria patients induce apoptosis in microvascular endothelial and neuronal cells contributing to vascular dysfunction, blood-brain barrier (BBB) damage and mortality. Endothelial progenitor cells (EPC) are microvascular endothelial cell precursors partly responsible for repair and regeneration of damaged BBB endothelium. Studies have shown that EPC’s are depleted in severe malaria patients, but the mechanisms mediating this phenomenon are unknown. Toll-like receptors recognize a wide variety of pathogen-associated molecular patterns generated by pathogens such as bacteria and parasites. We tested the hypothesis that EPC depletion during malaria pathogenesis is a function of heme-induced apoptosis mediated by CXCL10 induction and toll-like receptor (TLR) activation. Heme and CXCL10 concentrations in plasma obtained from malaria patients were elevated compared with non-malaria subjects. EPC numbers were significantly decreased in malaria patients (P < 0.02) and TLR4 expression was significantly elevated in vivo. These findings were confirmed in EPC precursors in vitro; where it was determined that heme-induced apoptosis and CXCL10 expression was TLR4-mediated. We conclude that increased serum heme mediates depletion of EPC during malaria pathogenesis.  相似文献

Antioxidant supplementation has become a common practice among athletes to boost sport achievement. Likewise, melatonin (MEL) has been ingested as an ergogenic aid to improve physical performance. To date, no study has checked whether the multiple beneficial effects of MEL have an outcome during a maximum running exercise until exhaustion. Therefore, the present study aimed to evaluate the effect of MEL ingestion on physical performance and biochemical responses (i.e., oxidative stress) during exhaustive exercise. In a double blind randomized study, thirteen professional soccer players [age: 17.5 ± 0.8 years, body mass: 70.3 ± 3.9 kg, body height: 1.80 ± 0.08 m; maximal aerobic speed (MAS): 16.85 ± 0.63 km/h; mean ± standard deviation], members of a first league squad, performed a running exercise until exhaustion at 100% of MAS, after either MEL or placebo ingestion. Physical performance was assessed, and blood samples were obtained at rest and following the exercise. Compared to placebo, MEL intake prevented the increase in oxidative stress markers (i.e., malondialdehyde), alleviated the alteration of antioxidant status (i.e., glutathione peroxidase, uric acid and total bilirubin) and decreased post-exercise biomarkers of muscle damage (i.e., creatine kinase and lactate dehydrogenase) (p < 0.05). However, physical performance was not affected by MEL ingestion (p > 0.05). In conclusion, acute MEL intake before a maximal running exercise protected athletes from oxidative stress and cellular damage but without an effect on physical performance.  相似文献

Peaks of ecdysteroids were observed during the different phases of embryonic development of intact Carausius eggs or eggs precociously deprived of their exochorion and cultivated under paraffin oil. Several groups of ecdysteroids were separated and analyzed by thin-layer chromatography (TLC) and high performance liquid chromatography (HPLC) combined with radioimmunoassay. Ecdysteroids were similar in the two categories of eggs, including high-polarity products (essentially conjugates hydrolyzable by Helix pomatia digestive juice, or alkaline phosphatase), possible ecdysonoic acids (unhydrolyzable polar substances), free hormones, and nonpolar ecdysteroids. Four ecdysteroids were identified by co-elution during HPLC with reference compounds of 20,26-dihydroxyecdysone, 20-hydroxyecdysone, ecdysone, and 2-deoxy-20-hydroxyecdysone. Concentrations of these substances (free and conjugated forms) were studied during the different stages of embryonic development: 20-hydroxyecdysone and 2-deoxy-20-hydroxyecdysone were the major free ecdysteroids. They showed parallel variations with large peaks at stages VI8 and VII6 whereas ecdysone titers were consistently low. Injected labelled ecdysone was converted efficiently into 20-hydroxyecdysone, and both compounds underwent 26-hydroxylation and/or conjugation to polar or apolar metabolites.  相似文献

Given the fact that, according to our knowledge, no study has compared the agro-environmental use of decontaminated with non-decontaminated sludge, a greenhouse experiment was carried out to test the growth of maize (Zea mays L., G-4011 Hybrid) and bioaccumulation of metals in the presence of four different sludges (MUC, QUC, BEC and DAI), before and after their decontamination by a novel process (METIX-AC). Data showed that decontaminated sludge ameliorated plant growth and biomass production, and decreased bioaccumulation of metals, more than control soil, inorganic chemical fertilization, or conventional non-decontaminated sludge. Since chemicals used by the METIX-AC process contained S and Fe, decontaminated sludge introduced large amounts of these elements, while the overall presence of metals was reduced. Often, sludge dose also affected maize growth and bioaccumulation of metals. Overall, no toxicity to plants was noticed and bioaccumulation and transfer of many metals remained below the limits reported in the literature.  相似文献

The involvement of apoplastic invertase (Ap Inv) and sucrose synthase (SuSy) in the somatic embryo development of black spruce (Picea mariana) was investigated under different maturation conditions. Replacing 6% sucrose with 3% or 1% sucrose in the maturation medium drastically decreased Ap Inv activity and amount in embryogenic tissues. This was accompanied by a decrease in the hexose pool that resulted in a lower starch deposition and protein amount in embryogenic tissues together with a lower embryo production. Conversely, SuSy activity was stable during maturation regardless of the sucrose concentration used in the medium. The presence of an extracellular enzyme responsible for sucrose hydrolysis in the maturation medium was also verified. An immunodetection experiment with anti-acid invertase antibodies revealed the presence of an active 53 kDa polypeptide in the medium, which had a similar molecular mass to that of the Ap Inv polypeptide found in embryogenic tissues. Utilization of sucrose from the medium by the tissues was also studied using labelled 14C-sucrose. Distribution of the radioactivity between tissular sucrose, glucose, and fructose showed that sucrose was diffused into the cell wall of embryogenic tissues and partly hydrolyzed by Ap Inv. These results show that the utilization of sucrose from the medium, the Ap Inv activity in embryogenic tissues, and the release of an active invertase into the medium operate together for the utilization of the carbohydrates during somatic embryo development in black spruce.  相似文献
