Zusammenfassung Untersucht wurden zufällig ausgewählte Leberstückchen normaler Wistarratten. 25 lückenlose Übersichten von periportalen und zentralen Arealen des Leberläppchens, die aus 2564 elektronenmikroskopischen Einzelaufnahmen (Vergrößerung 4600 x) zusammengesetzt waren, wurden bei einer Gesamtvergrößerung von 53000 x mit Hilfe eines Liniengitters nach der Methode. von Loud quantitativ ausgewertet.Die mittlere Cytoplasmafläche eines Hepatocyten, der im Läppchenzentrum liegt, ist größer als diejenige einer in der Peripherie des Läppchens gelegenen Leberzelle, die Zahl der Anschnitte von Mitochondrien pro Cytoplasmafläche im Zentrum geringfügig größer als in der Peripherie. Die Zahl der Lysosomen pro Cytoplasmafläche ist in der Peripherie des Lobulus dreimal höher als in seinem Zentrum.Der Flächenanteil der Mitochondrienanschnitte an der Cytoplasmafläche im Zentrum beträgt 12,3%, im periportalen Bereich 19,3%. Die mittlere Fläche eines Mitochondrienanschnittes ist im periportalen Gebiet doppelt so groß wie im Zentrum, die Membranprofildichte in Mitochondrien periportaler Zellen ist um etwa ein Drittel größer.Schüsselförmige und schlegeloder hantelförmige Mitochondrien mit parallel zur Längsachse ausgerichteten inneren Membranen wurden nur im Zentrum des Leberläppchens gefunden. Dasselbe gilt für Plasmaprotusionen der Hepatocyten in den Disseschen Raum. Glykogenablagerungen sind gleichmäßig über den Lobulus verteilt, auffallend ist jedoch die ungleichmäßige Verteilung auf die einzelnen Zellen.Die quantitativen Daten werden mit histochemischen Befunden und biochemisch ermittelten Enzymaktivitäten verglichen. Die morphologischen Beobachtungen werden im Zusammenhang mit ähnlichen Befunden, die an pathologisch veränderten Lebern erhoben wurden, diskutiert.
Structure of the hepatic lobule of the rat
Summary Small randomly chosen pieces of liver tissue of normal Wistar rats were investigated with the electron microscope. 25 survey pictures, consisting of 2564 individual micrographs, were analyzed quantitatively at a final magnification of 53,000 x by a linear scanning method as described by Loud.The average area of cytoplasm is larger in the centrolobular than in the periportal area. The number of mitochondria per unit of cytoplasmic area was roughly the same throughout a liver lobule. However, in the periportal zone the mitochondria were about two times larger than in the center and displayed a 30% higher membrane profile concentration. It was found that the total mitochondrial area per unit of cytoplasm was 12,3% in the centrolobular region as compared to 19,3% in the periportal zone. The number of lysosomes per unit of cytoplasmic area was about three times higher in the periportal than in the centrolobular zone.Cup- and dumbbell-shaped mitochondria with densely packed and longitudinally arranged internal membranes were only found in the centrolobular area. Irregular protrusions of the liver cell into the space of Disse were also found exclusively in this area. The amount of glycogen varied considerably from cell to cell but no significant difference in this respect could be seen between the two zones of the liver lobule.The quantitative findings are discussed with reference to available bio- and histochemical data. The morphological observations are compared with similar observations made on pathologically altered liver cells.

Zusammenfassung VonFormica (Serviformica) rufibarbis, F. (S.) cunicularia undF. (Raptiformica) sanguinea werden die sonst an Ameisen angepassten Lycaenidenraupen nur in unmittelbarer Nähe des Nesteinganges eines Ameisennestes angegriffen. BeiF. (F.) rufa ist diese Agressivität nicht raum-, sondern vermutlich stimmungsabhängig. Bei verglichenen anderenFormica-Arten trat sie nicht auf.
Summary Lycaenid caterpillars which usually are adapted to the aggressive behavior of ants are, despite of their adaptations, attacked byFormica (Serviformica) rufibarbis, F. (S.) cunicularia, F. (Raptiformica) sanguinea, andF. (F.) rufa. In the three first-mentionned species this agression happens only in the immediate neighborhood of the entrance to an ant-hill. On the contrary the aggressivity ofF. rufa seems to depend upon the actual mood of the ants but not upon the distance to an entrance. In other species of the genusFormica compared in experiments with Lycaenid larvae no aggressive behavior was observed.

Chez les autres fourmis étudiées à titre comparatif, on n'a pas constaté d'agressivité envers les chenilles de Lycénides.

Summary The Herring bodies in the posterior lobe of the bovine hypophysis are very large (2–600 ) and can be classified into three types. The type I Herring body contains an accumulation of neurosecretory granules. These Herring bodies are very scarce and should not be confused with the numerous, but small, axonal swellings which also contain neurosecretory granules.The type II Herring body is characterized by the presence of a varying number of normal, moderately electron dense and empty vesicles, autophagic vacuoles, multilamellate bodies and occasional mitochondria. These Herring bodies are frequently observed.The type III Herring body is typified by the presence of dense vesicles connected to tubular formations which contain material of variable electron density, of filaments, and of long slender and very numerous mitochondria.The presence of multilamellate bodies and autophagic vacuoles suggests that the type II Herring body is in a degenerating phase. This concept is further substantiated by the similarity between this type of Herring body and transected neurosecretory axons in which degeneration is occurring.A similar comparison suggests that the type III Herring body is undergoing a regenerative process. Summary The structural changes of the zona juxtamedullaris of the rat adrenal cortex at birth, have been examined by the light and the electron microscope. In this zone clusters of medullary cells lying among the strands of cortical tissue were observed. In the inner portion of the zona juxtamedullaris two types of adrenocortical cells were found: light and very-dark cells. The latter are smaller than the light cells and are always in close connection with the medullary tissue. The ultrastructural features of the very-dark cells suggest that these elements are in degeneration. This finding supports the hypothesis that at birth there is a partial degeneration of the rat zona juxtamedullaris, i.e. the zone corresponding to the fetal zone of some mammalian species.It is proposed that in all mammalian species at birth there is a partial regression of the zona juxtamedullaris and that the regression of the fetal zone is only the quantitative increase of this phenomenon. This hypothesis is discussed in relation to numerous data demonstrating that there are enzymatic conditions in the rat during fetal life, which permit a discrete hypertrophy of the adrenal cortex.The author wishes to express his appreciation to Dr. A. Two cytologically distinguishable species have been found in material ofChironomus australis. These species have been calledCh. australis andCh. duplex. Both belong to thepseudothummi-cytological grouping because they have the chromosome arm combinations AE, BF, CD, G.Ch. duplex shows a modified arm pattern due to a tandem fusion of arm G to arm E.—The banding patterns of the polytene chromosomes of the two species are compared to each other and to the Australian standard species,Ch. oppositus. Ch. australis is very close cytologically toCh. oppositus, whileCh. duplex, which is considered a derived species because of the tandem fusion, shows a number of inversion differences from the morphologically similarCh. australis. Ch. duplex is polymorphic for six inversions, four of which are simple inversions, the other two are complex involving also the transposition of some bands.Ch. australis appears to be monomorphic.  相似文献

Human serum and urine contain polypeptides which bind mannose 6-phosphate (M6P) and insulin-like growth factor II (IGF II) and crossreact with antibodies against the M6P/IGF II receptor. These polypeptides are considered to be fragments of the M6P/IGF II receptor. The major Mr approx. 205,000 fragment in serum and urine is about 10 kDa smaller in size than the membrane-associated receptor and is accompanied by minor forms with Mr values ranging from 104,000 to 180,000. The presence of receptor fragments in biological fluids indicates that shedding is one of the mechanisms contributing to the turnover of the M6P/IGF II receptor and that receptor fragments are part of the heterogenous group of serum proteins whic bind IGF II.  相似文献

8 antioxidants were tested in the SOS chromotest for induction of SOS function and for modulation of benzo[a]pyrene-induced SOS function. None of the antioxidants leads to increased beta-galactosidase activity by itself. Butylated hydroxytoluene at concentrations between 10(-5) M and 3 X 10(-4) M enhances benzo[a]pyrene-induced SOS function at benzo[a]pyrene concentrations between 10(-6) M and 3 X 10(-5) M. Butylated hydroxyanisole, ethoxyquin, propyl gallate and octyl gallate also slightly enhance benzo[a]pyrene-induced SOS function at concentrations up to 3 X 10(-4) M though to a lesser degree than butylated hydroxytoluene. Dodecyl gallate, vitamin C and alpha-tocopherol do not increase benzo[a]pyrene action. In concentrations exceeding 3 X 10(-4) M all synthetic antioxidants tested but not vitamin C and alpha-tocopherol decrease beta-galactosidase activity both in the absence and, more extensively, in the presence of benzo[a]pyrene. Preliminary data suggest that the apparent suppression of benzo[a]pyrene-induced SOS function is not due to an effect on the formation of benzo[a]pyrene metabolites by the metabolizing system used.  相似文献

A patient with Philadelphia-chromosome positive chronic myelogenous leukemia developed interferon antibodies on treatment with recombinant interferon alpha-2b. Clinically this event corresponded with progressive disease. No cross-reactivity of antibodies with human leukocyte interferon was found by Western blot. Treatment was switched to human leukocyte interferon with an obvious clinical effect: WBC was reduced and platelet count stabilized, but the effect was transient and no hematologic remission was achieved. Human leukocyte interferon may be an alternative in CML-patients with neutralizing antibodies to recombinant interferon alpha.  相似文献

We describe a transgenic mouse line, Pax8-rtTA, which, under control of the mouse Pax8 promoter, directs high levels of expression of the reverse tetracycline-dependent transactivator (rtTA) to all proximal and distal tubules and the entire collecting duct system of both embryonic and adult kidneys. Using crosses of Pax8-rtTA mice with tetracycline-responsive c-MYC mice, we established a new, inducible model of polycystic kidney disease that can mimic adult onset and that shows progression to renal malignant disease. When targeting the expression of transforming growth factor beta-1 to the kidney, we avoided early lethality by discontinuous treatment and successfully established an inducible model of renal fibrosis. Finally, a conditional knockout of the gene encoding tuberous sclerosis complex-1 was achieved, which resulted in the early outgrowth of giant polycystic kidneys reminiscent of autosomal recessive polycystic kidney disease. These experiments establish Pax8-rtTA mice as a powerful tool for modeling renal diseases in transgenic mice.  相似文献

CD1d-restricted invariant NKT (iNKT) cells can enhance immunity to cancer or prevent autoimmunity, depending on the cytokine profile secreted. Antitumor effects of the iNKT cell ligand alpha-galactosylceramide (alphaGC) and iNKT cell adoptive transfer have been demonstrated in various tumor models. Together with reduced numbers of iNKT cells in cancer patients, which have been linked to poor clinical outcome, these data suggest that cancer patients may benefit from therapy aiming at iNKT cell proliferation and activation. Herein we present results of investigations on the effects of human iNKT cells on Ag-specific CTL responses. iNKT cells were expanded using alphaGC-pulsed allogeneic DC derived from the acute myeloid leukemia cell line MUTZ-3, transduced with CD1d to enhance iNKT cell stimulation, and with IL-12 to stimulate type 1 cytokine production. Enhanced activation and increased IFN-gamma production was observed in iNKT cells, irrespective of CD4 expression, upon stimulation with IL-12-overexpressing dendritic cells. IL-12-stimulated iNKT cells strongly enhanced the MART-1 (melanoma Ag recognized by T cell 1)-specific CD8(+) CTL response, which was dependent on iNKT cell-derived IFN-gamma. Furthermore, autologous IL-12-overexpressing dendritic cells, loaded with Ag as well as alphaGC, was superior in stimulating both iNKT cells and Ag-specific CTL. This study shows that IL-12-overexpressing allogeneic dendritic cells expand IFN-gamma-producing iNKT cells, which may be more effective against tumors in vivo. Furthermore, the efficacy of autologous Ag-loaded DC vaccines may well be enhanced by IL-12 overexpression and loading with alphaGC.  相似文献
