A mild and rapid method is described for isolating various milk proteins from bovine rennet whey. β-Lactoglobulin from bovine rennet whey was easily adsorbed on and desorbed from a weak anion exchanger, diethylaminoethyl-Toyopearl. However, α-lactalbumin could not be adsorbed onto the resin. α-Lactalbumin and β-lactoglobulin from rennet whey could also be adsorbed and separated using a strong anion exchanger, quaternary aminoethyl-Toyopearl. The rennet whey was passed through a strong cation exchanger, sulphopropyl-Toyopearl, to separate lactoperoxidase and lactoferrin. α-Lactalbumin and β-lactoglobulin were adsorbed onto quaternary aminoethyl-Toyopearl. α-Lactalbumin was eluted using a linear (0–0.15 M) concentration gradient of NaCl in 0.05 M Tris–HCl buffer (pH 8.5). Subsequently, β-lactoglobulin B and β-lactoglobulin A were eluted from the column with 0.05 M Tris–HCl (pH 6.8), using a linear (0.1–0.25 M) concentration gradient of NaCl. The yields were 1260 mg α-lactalbumin, 1290 mg β-lactoglobulin B and 2280 mg β-lactoglobulin A from 1 l rennet whey. 相似文献

The formation of hyphae that grow solely by apical extension is a defining feature of filamentous fungi. Hyphal morphogenesis involves several key steps, including the establishment and maintenance of a stable polarity axis, as well as cell division via the deposition of septa. Several filamentous fungi have been employed in attempts to decipher the mechanisms underlying these steps. Amongst these fungi, Aspergillus nidulans has proven to be a particularly valuable model. The genetic tractability of this fungus coupled with the availability of sophisticated post-genomics resources has enabled the identification and characterization of numerous genes involved in hyphal morphogenesis. Here, we summarize current progress towards understanding the function of these genes and the mechanisms involved in polarized hyphal growth and septation in A. nidulans. A series of metal carboxylates containing pyridine N-oxide are prepared via one pot synthesis and solid phase synthesis. The structural variations from metal to metal are observed. In the case of reactions of manganese(II) acetate with pyridine N-oxide in the presence of aromatic carboxylic acids, polymeric complexes with bridging aromatic carboxylate as well as bridging pyridine N-oxide are observed. Whereas, the reaction of copper(II) acetate with pyridine N-oxide in the presence of an aromatic carboxylic acid led to mononuclear or binuclear paddle wheel carboxylate complexes with monodentate pyridine N-oxide. Co-crystal of two neutral complexes having composition [Cu2(OBz)4(MeOH)2][Cu2(OBz)4(pyO)2] (where OBz = benzoate, pyO = pyridine N-oxide) each neutral parts have paddle wheel structure. Solid phase reaction of zinc chloride with sodium benzoate prepared in situ and pyridine N-oxide leads to a tetra-nuclear zinc complex. 相似文献

Irreversible chemical programming of monoclonal aldolase antibody (mAb) 38C2 has been accomplished with β-lactam equipped mono- and bifunctional targeting modules, including a cyclic-RGD peptide linked to either the peptide (d-Lys6)-LHRH or another cyclic RGD unit and a small-molecule integrin inhibitor SCS-873 conjugated to (d-Lys6)LHRH. We also prepared monofunctional targeting modules containing either cyclic RGD or (d-Lys6)-LHRH peptides. Binding of the chemically programmed antibodies to integrin receptors α(v)β(3) and α(v)β(5) and to the luteinizing hormone releasing hormone receptor were evaluated. The bifunctional and bivalent c-RGD/LHRH and SCS-783/LHRH, the monofunctional and tetravalent c-RGD/c-RGD, and the monofunctional bivalent c-RGD chemically programmed antibodies bound specifically to the isolated integrin receptor proteins as well as to integrins expressed on human melanoma M-21 cells. c-RGD/LHRH, SCS-783/LHRH, and LHRH chemically programmed antibodies bound specifically to the LHRH receptors expressed on human ovarian cancer cells. This approach provides an efficient, versatile, and economically viable route to high-valency therapeutic antibodies that target defined combinations of specific receptors. A high-performance liquid chromatographic method is described for the determination of opipramol in human plasma. Opipramol was extracted into tert.-butylmethyl ether, separated on a cyanopropyl silica column and detected at 254 nm. Imipramine was used as internal standard. The limit of quantitation was 250 pg/ml using 1.5 ml plasma. Precision was better than 9%, inaccuracy less than 8%. The assay is more sensitive than previously published methods, and it has been applied to the analysis of plasma samples from a pharmacokinetic study. 相似文献

Bioturbation by benthic infauna has important implications for the fate of contaminants as well as for changes to the sediment
structure, chemistry and transport characteristics. There is an extensive literature dealing with the influence of sedimentary
variables on the structure and function of infaunal marine and estuarine organisms but less is known of the converse, the
influence of biota on sedimentary structure. Although some work has been carried out regarding spatial and temporal patterns
of bioturbation, little attention has been given to the effects of pollution. The paper gives a framework of animal sediment
relationships in an intertidal environment and discusses the general role of macrofauna in structuring and modifying sedimentary
features. A brief outline of the various techniques used for quantifying the degree of bioturbation is given and some of these
techniques have then been used to demonstrate the effect of a petrochemical discharge on the bioturbation potential of intertidal
communities in the Humber estuary, eastern England. These studies indicate an increase in bioturbation with increasing distance
from the source of pollution, not only because of differences in abundance, animal size and depth of activity but also because
of the difference in species composition between the communities. As a means of interpreting the responses, the species present
have been broadly classified in terms of their feeding strategy and sediment modification potential. The paper concludes by
discussing the potential impact, in terms of effect on sediment transport, of selectively removing the different guilds (by
pollution).

The movement ability of individuals has become increasingly important to a variety of ecological questions. In this study,
I investigate how plant structure and changes in body size through development affect the movement ability of a predaceous
stinkbug, Podisus maculiventris, on three species of goldenrod (Solidago spp.) representing a wide range of surface complexities. I adapt existing techniques for quantifying movement in two dimensions
to the study of movement on natural plant structures in three dimensions. These experiments indicate that plant structure
and insect size are significant factors affecting the movement ability of P. maculiventris. Changes in movement ability due to factors of ontogeny and different habitat structures suggest that the scale of an individual’s
ambit or ecological sphere of influence may vary within its lifespan. Considering the influence of ontogeny and habitat structure
on movement ability may be useful to investigations of population dynamics, foraging behavior, and pest management.

The presence of litter has the potential to alter the population dynamics of plants. In this paper, we explore the effects
of litter on population dynamics using a simple experimental laboratory system with populations of the annual crucifer, Cardamine pensylvanica. Using a factorial experiment with four densities and three litter levels, we determined the effect of litter on biomass
and plant fecundity, and the life stages responsible for these changes in yield. Although litter had significant effects on
seed germination and on seedling survivorship, we show, using a population dynamics model, that these effects were not demographically
significant. Rather, the potential effect of litter on population dynamics resulted almost entirely from its effect on biomass.
Persistent litter suppressed plant biomass and apparently removed the direct density effect present in the absence of litter.
Thus, litter changed the shape of the recruitment curve from slightly humped to asymptotic. In addition to changing the shape
of the recruitment curve, litter reduced the carrying capacity of the populations. Thus, the population dynamics model indicated
that not all statistically significant responses were dynamically significant. Given the potential complexity of litter effects,
simple population models provide a powerful tool for understanding the potential consequences of short-term responses.

Identification of Culturable Oligotrophic Bacteria within Naturally Occurring Bacterioplankton Communities of the Ligurian Sea by 16S rRNA Sequencing and Probing 总被引:10,自引:0,他引:10

Abstract
Typical marine bacteria (i.e., obligately oligotrophic) that were numerically dominant members of naturally occurring marine
communities were identified by cloning and sequencing the amplified 16S rRNA genes obtained from dilution cultures of the
original samples. The data reported here refer to two different habitats of a marine pelagic environment (28 miles offshore,
in the northwestern Mediterranean Sea). The samples were taken from the water column at two representative layers, i.e., the
30-m depth, corresponding to the chlorophyll maximum layer, and the 1800-m depth, representative of a deep, oligotrophic environment.
Three major lineages were found in the 16S rDNA clone libraries prepared from the two samples, two of which could be assigned
to the Vibrio and the Rhodobacter groups. The third lineage was a distant relative of the genus Flavobacterium, but it was not closely related to any marine isolate. Six oligonucleotide probes, either complementary to the conserved sequence
domains or selectively hybridizing to the clone sequences, were designed for use as hybridization group-specific and strain-specific
probes. A single-mismatch discrimination between certain probes and nontarget sequences was demonstrated by detecting the
probes' specificity at different hybridization and washing conditions. The screening of the clone libraries with the obtained
probes revealed that neither the 30-m sample higher dilution nor the 1800-m one were pure cultures. While some representatives
of the Vibrio group were found in both the surface and the deep sample, the members of the Flavobacterium and Rhodobacter lineages were detected only in the deep and the euphotic layers, respectively. We suggest an approach for analyzing autochthonous
marine bacteria able to grow in unamended seawater.
