Summary Extracts of 9-day-old mycelia of Aspergillus parasiticus NRRL 2999 were assayed for peroxidase activity and for their ability to degrade aflatoxin. A positive relationship existed between rates of aflatoxin degradation and amount of peroxidase activity in these extracts. The supernatant fluid of homogenates from mycelia grown under similar conditions varied in amount of peroxidase present (170 to 2215 U/g). The fraction obtained, by precipitation with (NH4)2SO4 at 45% of saturation, from six different homogenates prepared from three mycelial mats contained peroxidase and degraded aflatoxin. Rates of aflatoxin degradation by and amounts of peroxidase activity in each sample obtained from mycelial homogenates with (NH4)2SO4 at 60% of saturation varied; however, when increased amounts of peroxidase activity were present, more aflatoxin was degraded and vice versa. Relatively little peroxidase activity was present in the fraction obtained with (NH4)2SO4 at 30% of saturation and little or no aflatoxin was degraded by this precipitate. Trends for degradation of aflatoxin when more or less peroxidase activity was present in mycelial preparations suggest that the enzyme may be involved in degradation of aflatoxin by the Aspergillus.  相似文献

Development of secretagogue response in rat pancreatic acinar cells   总被引:4,自引:0,他引:4  
Two to 3 days prior to birth, acinar cells of the rat pancreas acquire morphologic and biochemical characteristics of the adult gland. To determine if differentiation of the secretory apparatus coincides temporally with the capacity of the cell to respond to secretory stimuli, lobules of embryonic, neonatal, and adult rat pancreas were compared for their ability to respond to secretagogues presumed to act directly via hormone receptors [caerulein and carbamylcholine (carbachol)] or indirectly (cyclic nucleotide analogs and the Ca2+ ionophore A23187). Of all agents tested, only dibutyryl cAMP elicited discharge of secretory proteins at day 20 in utero and preceded hormone stimulation by 1 day. A23187 elicited discharge by Day 21 in utero; its action was near adult levels in contrast to hormonal stimuli whose effect was maximal only at birth. All secretagogues required Ca2+ and energy to induce discharge. Pulse-chase autoradiography of lobules from Day 20 embryonic glands indicated that the acinar cells were capable of transporting [3H]leucine-labeled proteins to zymogen granules at rates roughly equivalent to those in adult glands. SDS gel electrophoretograms confirmed that the bulk of 14C-amino acid incorporation into proteins at a given age was primarily into exportable proteins. The results indicate that acinar cells synthesize and package secretory proteins into zymogen granules about 2 days before they are capable of responding to hormonal stimuli and to intracellular effectors.  相似文献

Infrared spectroscopy of collagen and collagen-like polypeptides.   总被引:11,自引:0,他引:11  
The set of synthetic polytripeptides and polyhexapeptides which can adopt a triple-helical form constitute a good model system for investigating collagen structure. Here we consider previous and new infrared spectroscopic studies on collagen and present the infrared spectra of a number of polymers with collagen-like features. The amide A band position for all triple-helical polypeptides is higher than that observed for most proteins and polypeptides, and this high frequency appears to be related to the degree of supercoiling of the triple helix. It is possible that with increased supercoiling of the three chains the angles between the groups involved in the intramolecular hydrogen bonds become less favorable, or these bonds may become unusually long. The frequency of the amide I band varies considerably for triple-helical polypeptides with different amino acid sequences, and often minor bands are observed. This finding contrasts with the observations for polypeptides in a pleated sheet or α-helical form, where the same amide I frequency is observed regardless of the amino acid composition. An explanation for this variation is proposed in terms of the hydrogen bonding properties of imino acids. Significant spectral changes in the amide I region are observed on hydration in the spectra of some triple-helical polypeptides, but corresponding changes have not been found in the collagens examined.  相似文献

Ultrastructural examinations of the flagellum of Compylobacter (Vibrio) fetus were performed throughout the growth cycle. Filament diameters, exceeding 17.6 nm during the exponential phase, were substantially greater than those reported for unsheathed flagella of other genera with the exception of Pseudomonas fluorescens. Filament diameters increased during growth, reaching a mean width of 21.2 nm in middle to late stationary phase. Internal flagellar structure, principally of the parallel lined variety, was observed during the later periods of growth but not during exponential or early stationary phase. Despite the unusually large filament sizes, no evidence of a flagellar sheath was observed after selected treatments (0.01 N HCl, 6 M urea, tris(hydroxymethyl) amino-methane-hydrochloride buffer, warm water) or examination of thin sections. To determine whether alterations in filament size and variable ability to demonstrate filament fine structure were correlated with progressive changes in serological activity, agglutination and immobilization tests were conducted with antisera directed against intact flagella, the principal flagellar antigen, the O antigen, and a superficial glycoprotein which has been found in association with the flagellum and the cell envelope. Significant differences in the serological activity of cells at different growth intervals were not noted with any of the sera employed.  相似文献

The phytohemagglutinin, concanavalin A (Con A), interacts specifically and reversibly with the polyglucosyl glycerol phosphate teichoic acid of Bacillus subtilis 168 cell walls. Advantage has been taken of this interaction to examine the organization of the surface teichoic acid at the ultrastructural level. Con A-treated whole cells and cell walls contain an irregular, fluffy layer 25 to 60 nm thick which is absent in untreated or alpha-methyl glucoside-treated preparations. This discontinuous layer is present only on the outer profile of Con-A-treated cell walls. The surface teichoic acid is proposed to be oriented perpendicular to the long axis of the cell. Fixation and embedment for electron microscopy result in condensation of this layer which then contributes to the stainable portion of the wall. Con A treatment binds adjacent teichoic acid molecules in their native configuration producing the irregular, fluffy layer visualized.  相似文献

The population structure of 4 California Iridaea cordata (Turner) Bory populations was studied. Random sampling procedures were used to measure seasonal variations in standing crops, density and size-class distribution of life history stages. The results describe aspects of the in situ life history; a prerequisite to realistically considering the distribution, ecology or life history expressions of an alga. Seasonal fluctuations in density occur only in the juvenile stage, which is initiated during the winter (November to January) predominantly from the basal perennial crusts. Both the gametangial and tetrasporangial stages are present throughout the year. The tetrasporangial stage is dominant in relation to the sexual stages in both density and biomass during most of the year, except spring when the new crop in just maturing and all stages are abundant. Density in nearly constant and observable changes in the populations are due to biomass fluctuations. Seasonal lows in biomass occur during the winter with the majority of thalli in the smaller size-classes. Growth and maturation culminate in peak summer crops and dominance of the tetrasporangial stage, followed by autumnal senescence and die-back in winter. Carrageenans analyzed from immature thalli showed a predominance of lambda-type, previously determined as specific for tetrasporangial plants. This indirect evidence for the tetrasporangial nature of immature plants suggests that dominance of the diploid stage occurred prior to blade development and most likely at the spore level. Alternatively, field results indicate a major contribution and possible replacement of alternation of gametangial and tetrasporangial stage by thallus perennation and vegetative reproduction.  相似文献

During the course of a one year field study of the Lesser Bushbaby (Galago senegalensis moholi), using a technique of direct observation with red light, it was discovered that family groups which slept together during the daytime split up at night, with each member spending most of the time alone. During the night individuals would come into contact with one another or with members of adjacent groups and the resulting social behaviour varied considerably. An interpretation has been made of more than 120 interactions observed in the field based on the behaviour of bushbabies introduced to one another under controlled semi-natural conditions.  相似文献
