Results in this study are consistent with those of Murdock and his colleagues who clearly demonstrated that clonidine, an agonist of octopaminergic receptors in some insects, significantly increases sucrose feeding. Their studies, however, did not examine the effect of clonidine on protein feeding. Injection of a 20 microg/microl/fly dose of clonidine significantly reduces protein feeding in both sexes of Phormia regina, instead of stimulating feeding as is observed with carbohydrate feeding. The manner in which the flies are fed prior to starvation and the method of testing influences the amounts of diet consumed. It is proposed that the biogenic amines influence the state of hunger (i.e., protein versus carbohydrates) while other chemicals and neural mechanisms (i.e., such as sulfakinins and stretch receptors, respectively) affect satiety.  相似文献

The invertebrate immune system produces melanotic nodules in response to bacterial infections and this has previously been shown to be mediated by eicosanoids. Nodulation occurs in two phases: the first involves hemocyte degranulation and activation of the prophenoloxidase cascade; the second involves formation of a cellular capsule by attachment and spreading of hemocytes. We demonstrate that inhibitors of eicosanoid biosynthesis affect both of these phases of nodulation in Galleria mellonella. The phospholipase A(2) inhibitor, dexamethasone, as well as the cyclooxygenase inhibitor, indomethacin, significantly inhibit phagocytosis in vitro and prophenoloxidase activation in vivo. The inhibitory effects of dexamethasone were abolished by the addition of exogenous arachidonic acid. Furthermore, 5,8,11,14- eicosatetraynoic acid, dexamethasone and indomethacin inhibit hemocyte spreading in vitro. The findings support the idea that eicosanoid derivatives mediate both phases of the nodulation response and are consistent with previous studies which attribute roles for eicosanoids in other species as modulators of cell activity.  相似文献

The harbor seal (Phoca vitulina) has the most extensive distribution of any phocid seal species. An analysis of population structure in this species across its European range was made using 7 phocid derived microsatellites in a sample of 1,029 individuals from 12 separate geographic areas. Despite the species potential for long-distance movement, significant genetic differentiation between areas was observed using an unbiased estimator of RST. Six distinct population units were identified: Ireland-Scotland, English east coast, Waddensea, western Scandinavia (Norway-Kattegat-Skagerrak-west Baltic), east Baltic, and Iceland. Little local substructuring is present along coastlines with a continuous distribution of breeding animals, but differentiation does increase with geographic distance. The degree of differentiation is greater over equivalent distances where the distribution is discontinuous, such as along coasts where breeding colonies are separated by large distances or by stretches of open sea. Patterns of population differentiation derived from microsatellites are very similar to those obtained from previous mitochondrial DNA analysis and suggest that philopatry in harbor seals operates over 300-500 km. In Europe, harbor seals have experienced a complex demographic history and patterns of population structure are likely to have been affected by natural environmental influences such as Pleistocene glaciations and epizootics. Comparison of Nm values from an unbiased estimator of RST, GST, and theta are consistent and, in some cases, may indicate populations where conditions deviate from the expectations of the RST model.   相似文献

The mandibular closer muscles of the cricket, Gryllus domestica, contain a 5-hydroxytryptamine (5-HT)-sensitive receptor that is coupled to adenylate cyclase. A structure-activity study of the 5-HT molecule indicates that the integrity of the ethylamine sidegroup and the presence of a negatively charged moiety at the 5 position (-OH, -OCH3) are essential for activity. A pharmacological profile is presented for this receptor. The receptor differs from any reported mammalian 5-HT receptor in that none of the mammalian agonists tested were effective. However, the mammalian antagonists for 5-HT receptors, spiperone, mianserin, and ketanserin as well as the anti-histaminic cyproheptadine were all effective antagonists in this preparation. Preliminary analysis of antagonism, particularly by spiperone, shows that these antagonists are probably acting non-competitively. On the basis of the pharmacological data, and comparisons with other insect systems, the 5-HT receptor present in the cricket mandibular muscles has been tentatively classified as 5-HT2-like.  相似文献

The response of rainbow trout (Salmo gairdneri Richardson) to sublethal levels of sodium pentachlorophenate (NaPCP) was determined by measuring brain concentrations of the amines dopamine (DA), norepinephrine (NE), 5-hydroxytryptamine (5-HT) and the free amino acid tryptophan (TP) using high performance liquid chromatography with electrochemical detection. Sodium pentachlorophenate had no major effect on the concentrations of the biogenic amines. Mean control concentrations (ng g−1 brain, S.E.) were: DA, 146.2, 9.9; NE, 205.8, 16.6; and 5-HT, 110.5, 8.8. There was a significant dose-dependent increase in TP concentration. Fish exposed to 200ng I−1 NaPCP showed a 128% increase in TP level relative to the mean control concentration of 1980 (50) ng g−1 brain. The effects of diets varying in lipid and carbohydrate content, anaesthetization with tricaine methanesulphonate or 2-phenoxyethanol, and electroshock were also investigated. No differences between treatment groups were detected.  相似文献

Biopesticides and agrichemicals are applied using basically the same equipment. The limitations imposed on biological or conventional chemical pesticides by current application systems are discussed. In general, any pesticide must be applied into a crop using the commonly used application system in that crop. This will normally be the hydraulic nozzle. Researchers attempting to increase the efficacy of a biopesticide by changing application system should bear in mind the constraints set by farmers on altering their usual spraying practices. These constraints are considered along with the criteria required for the successful market introduction of a novel application system. A novel application system, the “double nozzle” is introduced, which fulfils the criteria discussed, in particular the reduction in terms of amount of active ingredient required for pest control. The scientific rationale for this new system is explained, and its performance discussed.  相似文献

The lipid composition of flight muscle mitochondria was determined in adult male acclimated for 30 days at 31°C and 45°C respectively. Locusts held at 31°C showed lower levels of phosphatidylcholine and higher levels of phosphatidylethanolamine than the 45°C-acclimated insects. A trend towards an increased cholesterol:phospholipid ratio was also observed at the higher temperature. Wide angle X-ray diffraction procedures indicated a difference of 5°C in the lipid phase transition temperatures of mitochondrial preparations derived from the two groups of insects with the 45°C-acclimated samples demonstrating the higher transition temperature.  相似文献

Antibody against cockroach trehalase inhibitor was prepared and tested against the plasma of adult locusts and larval silkworms to determine whether these species possess a similar protein. An immunopositive response was elicited in both species. Studies using immunogold labeling show that adult cockroaches have trehalase inhibitor protein in granules of plasmatocytes and in oenocytoid-like structures. Localization of the immunoreactive protein with trehalase inhibitor antibody in locust hemocytes indicated that the protein is also contained in the granules of plasmatocytes. However, in the hemolymph of silkworm larvae, the immunoreactive protein was found only in the spherules of spherulocytes. The results suggest that insect hemolymph commonly contains trehalase inhibitor both in plasma and in certain hemocytes.  相似文献
