The importance of top predators in controlling ecological processes in large, intact ecosystems is unclear. In grasslands that support abundant ungulates, top–down control by predators may be particularly important, because of the tight biogeochemical linkages of ungulate prey with plants and soil microbes. Here, I examined the effects of the recent reintroduction of the gray wolf Canis lupus on ecosystem processes in Yellowstone National Park, where herds of grazing ungulates previously have been shown to stimulate several processes, including soil net nitrogen (N) mineralization. Rates of ungulate grazing intensity and soil net N mineralization were compared before and after wolf reintroduction in grasslands ranging five‐fold in aboveground production. Grazing intensity and grassland net N mineralization declined after wolf reintroduction, a likely partial function of fewer ungulates; wolf predation has been one of several factors implicated in causing the decline in Yellowstone ungulates. In addition, the spatial pattern of grazing and net N mineralization changed after reintroduction. A shift in the spatial patterns of grazer‐associated processes is consistent with a growing body of work indicating that wolves have changed habitat use patterns of ungulates in Yellowstone National Park. These findings suggest widespread wolf effects on ungulate prey, plants, and microbial activity that have spatially reorganized grassland energy and nutrient dynamics in Yellowstone Park.  相似文献

The present studies show that hydrolysis of a phosphodiester bond, most likely ATP, is a distinct, second step required to complete import of the F1-ATPase beta-subunit into the mitochondria. This step follows a membrane potential-dependent first step. We show, using an inhibitor of adenine nucleotide transport and the analogue beta,gamma-AMP-PCP, that the activity required for this phosphodiester hydrolysis-dependent completion of protein import resides outside the mitochondrial inner membrane. This activity is proposed to act on the precursor at the site of translocation either to render it competent or to catalyze its vectorial movement directly through the import apparatus. This activity shares properties ascribed to proteins of the heat-shock family, which are proposed to participate in the ATP-dependent refolding of partially denatured proteins and nascent peptides.  相似文献

Phase, darkfield, and computer-enhanced microscopy were used to observe the surface microenvironment of flow cells during bacterial colonization. Microbial behavior was consistent with the assumptions used previously to derive surface colonization kinetics and to calculate surface growth and attachment rates from cell number and distribution. Surface microcolonies consisted of closely packed cells. Each colony contained 2n cells, where n is the number of cell divisions following attachment. Initially, cells were freely motile while attached, performing circular looping movements within the plane of the solid-liquid interface. Subsequently, cells attached apically, maintained a fixed position on the surface, and rotated. This type of attachment was reversible and did not necessarily lead to the formation of microcolonies. Cells became irreversibly attached by progressing from apical to longitudinal attachment. Longitudinally attached cells increased in length, then divided, separated, moved apart laterally, and slid next to one another. This resulted in tight cell packing and permitted simultaneous growth and adherence. After approximately 4 generations, individual cells emigrated from developing microcolonies to recolonize the surface at new locations. Surface colonization byPseudomonas fluorescens can thus be subdivided into the following sequential colonization phases: motile attachment phase, reversible attachment phase, irreversible attachment phase, growth phase, and recolonization phase.  相似文献

The rapid increase in protein synthesis that occurs on addition of insulin (1 mU/ml) to stepped-down 3T3 cells was blocked by pre-incubation of the cells with pertussis toxin. Cholera toxin on the other hand stimulated protein synthesis and this effect was insensitive to actinomycin D and inhibited by pro-treatment of the cells with phorbol dibutyrate to deplete cell protein kinase C. Insulin was found to cause a rapid and transient increase in diacylglycerol (DAG) synthesis. The insulin-induced increase in diacylglycerol was blocked by pertussis toxin. Exogenous DAG (10 M) stimulated protein synthesis within 1 hour. The results suggest that insuIin stimulates ribosomal activity through a signal mechanism that involves a G-protein mediated activation of phospholipase C to increase DAG levels.  相似文献

Using an immunofluorescence assay developed to identify serogroups(i.e. clusters of strains labelled by one antiserum), the compositionof natural populations of phycoerythrin-containing Synechococcusspp. was examined. The 7803 (open ocean clone)-serogroup wasfound in most oceanic regions, but was most prevalent (up to85%) in tropical and subtropical waters during spring and summer.At coastal Long Island stations it was most abundant (up to65%) when water temperatures were >22°C. The seasonaland geographic distribution of the 7803-serogroup appeared tobe limited by water temperature. No consistent pattern was observedin the per cent composition with depth in the Sargasso Sea orat coastal to offshore stations in the North-west Atlantic Oceanor eastern tropical North Pacific Ocean. The 8016 (coastal clone)-serogroupwas abundant at coastal and estuarine stations off Long Island(up to 95 %) and its appearance was also correlated with warmwater temperature (> 15°C). However, this serogroup remaineda constant proportion of the population at the Long Island Soundstation during early winter months (through January) when abundanceof the 7803-serogroup was negligible. Owing to limited data,the oceanic distribution of the 8016-serogroup is not yet discernible.Lastly, antisera to the phycocyanin-dominant Synechococcus spp.clones failed to label any cells in samples collected from severaloceanic stations. Thus, these strains appear to be limited tocoastal and estuarine regions, which is consistent with predictionsfrom experiments comparing the photosynthetic performance ofthe phycoerythrin-dominant and phycocyanin-dominant clones. 1Present address: Department of Oceanography, University ofHawaii, Honolulu, HI 96822, USA  相似文献

Electron probe microanalysis of transverse sections was used to provide semiquantitative estimates of Al and other cations in the leaf tissues of some Al-accumulating plants of the cerrado vegetation of central Brazil. Concentrations of Al were generally higher than those of other cations in the phloem elements of these plants growing on dystrophic as well as fertile acid soils. When one of the Al-accumulating species,Vochysia thyrsoidea, was grown in a calcareous soil, the concentration of Al in the phloem elements of leaves was lower than that of Ca and K though the cotyledons showed higher concentrations of Al than those of other cations.  相似文献

The geometries of interacting arginine-carboxyls in proteins   总被引:3,自引:0,他引:3  
The geometries are reported for interacting arginine-carboxyl pairs obtained from 37 high resolution protein structures solved to a resolution of 2.0 A or better. The closest interatomic distance between the guanidinium and carboxyl is less than 4.2 A for 74 arginine and carboxyl groups, with the majority of these lying within hydrogen-bonding distance (2.6-3.0 A). Interacting pairs have been transformed into a common orientation, and arginine-carboxyl, and carboxyl-arginine geometries have been calculated. This has been defined in terms of the spherical polar angles T theta, T phi, and the angle P, between the guanidinium and carboxyl planes. Results show a clear preference for the guanidinium and carboxyl groups to be approximately coplanar, and for the carboxyl oxygens to hydrogen bond with the guanidinium nitrogens. Single nitrogen-single oxygen is the most common type of interaction, however twin nitrogen-twin oxygen interactions also occur frequently. The majority of these occur between the carboxyl oxygens and the NH1 and NE atoms of the arginine, and are only rarely observed for NH1 and NH2. The information presented may be of use in the modelling of arginine-carboxyl interactions within proteins.  相似文献

The silicified plates of certain testate amoebae have the necessary characteristics to be useful paleoindicators. These plates are usually abundant and well preserved in sedimentary deposits, where they can be studied using the same preparation techniques developed for other siliceous microfossils. Individual plates can usually be identified to the generic and, in some cases, to the species level. A preliminary survey of plates recorded in the surficial (top centimetre) sediments of 38 Adirondack Mountain (NY) lakes suggests that these testaceans may reflect catchment features (e.g. bog development) more closely than lakewater characteristics. The siliceous, species-specific scales of heliozoans are also shown to preserve in sedimentary profiles.  相似文献

In order to examine the influence of protein structure on the post-translational import of a protein into mitochondria, the carboxyl-terminal 129 residues of F1-ATPase beta-subunit precursor (511aa) have been replaced with 61 residues of yeast copper metallothionein. Import of the F1 beta-copper metallothionein (beta CuMT) hybrid into mitochondria was as efficient as that of the F1 beta precursor in the absence of copper. Addition of copper to mitochondrial import reactions, which had no significant effect on import of the F1 beta-subunit precursor, blocked import of the beta CuMT protein. This copper-dependent transport block for the beta CuMT precursor occurred after the precursor was bound to mitochondria. Expression of the beta CuMT protein in vivo revealed that beta CuMT would bind copper and allow growth of a copper-sensitive yeast host on an otherwise inhibitory level of the cation as long as it was localized in the cytoplasm. These data indicate that the binding of copper by beta CuMT renders it refractile for partial unfolding which is necessary for its translocation into mitochondria. These observations provide an alternative scheme for the selection of mutants defective in mitochondrial import.  相似文献

Glutamine synthetase isozymes in elasmobranch brain and liver tissues   总被引:1,自引:0,他引:1  
Glutamine synthetase is present as isozymic forms in the elasmobranchs Squalus acanthias (dogfish shark) and Dasyatis sabina (stingray). Subcellular fractionation of elasmobranch brain and liver tissue shows the enzyme to be predominantly cytosolic in the former tissue and mitochondrial in the latter. For the cytosolic brain enzyme, the subunit Mr equals 42,000 in the stingray and 45,000 in the shark, as determined by sodium dodecyl sulfate-gel electrophoresis/Western blotting. The subunit Mr = 45,000 and 47,000, respectively, for stingray and dogfish mitochondrial liver enzymes. Translation of total brain RNA from both species gives immunoprecipitable nascent peptides of the same size as their respective mature enzymes. However, in liver tissue, translation of glutamine synthetase mRNA yields peptides of higher Mr than that of the mature enzymes. In dogfish liver, Mr = 50,000 for the translation product and, in stingray liver, Mr = 48,000. This suggests that the translocation of the enzyme into liver mitochondria may be via a signal or leader sequence mechanism. The larger liver isozyme of elasmobranch glutamine synthetase is found in kidney where it is also known to be mitochondrial. The smaller cytosolic isozyme occurs in retina, heart, gill, and rectal gland tissue as well as in brain.  相似文献
