In the crystal structure of the bovine heart mitochondrial F(1)-ATPase (Abrahams, J. P., Leslie, A. G. W., Lutter, R., and Walker, J. E. (1994) Nature 370, 621-628), the two liganded beta subunits, one with MgAMP-PNP bound to the catalytic site (beta(T)) and the other with MgADP bound (beta(D)) have closed conformations. The empty beta subunit (beta(E)) has an open conformation. In beta(T) and beta(D), the distance between the carboxylate of beta-Asp(315) and the guanidinium of beta-Arg(337) is 3.0-4.0 A. These side chains are at least 10 A apart in beta(E). The alpha(3)(betaD311C/R333C)(3)gamma subcomplex of TF(1) with the corresponding residues substituted with cysteine has very low ATPase activity unless it is reduced prior to assay or assayed in the presence of dithiothreitol. The reduced subcomplex hydrolyzes ATP at 50% the rate of wild-type and is rapidly inactivated by oxidation by CuCl(2) with or without magnesium nucleotides bound to catalytic sites. Titration of the subcomplex with iodo[(14)C]acetamide after prolonged treatment with CuCl(2) in the presence or absence of 1 mM MgADP revealed nearly two free sulfhydryl groups/mol of enzyme. Therefore, one pair of introduced cysteines is located on a beta subunit that exists in the open or partially open conformation even when catalytic sites are saturated with MgADP. Since V(max) of ATP hydrolysis is attained when three catalytic sites of F(1) are saturated, the catalytic site that binds ATP must be closing as the catalytic site that releases products is opening.  相似文献

Intestinal microbiota is considered to play an integral role in maintaining health of host by modulating several physiological functions including nutrition, metabolism and immunity. Accumulated data from human and animal studies indicate that intestinal microbes can affect lipid metabolism in host through various direct and indirect biological mechanisms. These mechanisms include the production of various signalling molecules by the intestinal microbiome, which exert a strong effect on lipid metabolism, bile secretion in the liver, reverse transport of cholesterol and energy expenditure and insulin sensitivity in peripheral tissues. This review discusses the findings of recent studies suggesting an emerging role of intestinal microbiota and its metabolites in regulating lipid metabolism and the association of intestinal microbiota with obesity. Additionally, we discuss the controversies and challenges in this research area. However, intestinal micro-organisms are also affected by some external factors, which in turn influence the regulation of microbial lipid metabolism. Therefore, we also discuss the effects of probiotics, prebiotics, diet structure, exercise and other factors on intestinal microbiological changes and lipid metabolism regulation.  相似文献

The values and variation characteristics of energy components, their relationship with net radiation and the characteristics of water balance in the forest were analyzed, based on the observation data of energy fluxes, meteorological parameters and biomass in a tropical seasonal rain forest in Xishuangbanna from January 2003 to December 2004. The results show that annual net radiation was 3516.4 MJ/(m2 · a) and 3516.6 MJ/(m2 · a) in 2003 and 2004, respectively, of which 46% and 44% were used in latent heat flux, and 12% and 11% were lost as sensible heat flux. Annual mean canopy surface conductance was 10.3 mm/s and 10.0 mm/s in 2003 and 2004, respectively. Moreover, canopy surface conductance was lower in dry-hot seasons than in fog-cool and rainy seasons. Canopy surface conductance correlated significantly and positively with leaf area index, but negatively with water vapor pressure deficit. In general, canopy surface conductance was not affected directly by soil water content, but highly depended on soil moisture status when soil water content was below 0.15 m3/m3. Annual total evapotranspiration of this forest ecosystem in dry seasons was lower than that in rainy seasons, which was considered as one of the most important reasons that tropical seasonal rain forest could survive and flourish in Xishuangbanna at limit of water and heat.  相似文献

To push the energy density limit of supercapacitors, a new class of electrode materials with favorable architectures is strongly needed. Binary metal sulfides hold great promise as an electrode material for high‐performance energy storage devices because they offer higher electrochemical activity and higher capacity than mono‐metal sulfides. Here, the rational design and fabrication of NiCo2S4 nanosheets supported on nitrogen‐doped carbon foams (NCF) is presented as a novel flexible electrode for supercapacitors. A facile two‐step method is developed for growth of NiCo2S4 nanosheets on NCF with robust adhesion, involving the growth of Ni‐Co precursor and subsequent conversion into NiCo2S4 nanosheets through sulfidation process. Benefiting from the compositional features and 3D electrode architectures, the NiCo2S4/NCF electrode exhibits greatly improved electrochemical performance with ultrahigh capacitance (877 F g?1 at 20 A g?1) and excellent cycling stability. Moreover, a binder‐free asymmetric supercapacitor device is also fabricated by using NiCo2S4/NCF as the positive electrode and ordered mesoporous carbon (OMC)/NCF as the negative electrode; this demonstrates high energy density (≈45.5 Wh kg?1 at 512 W kg?1).  相似文献

Despite the effectiveness of sodium as a p‐type dopant for lead chalcogenides, its solubility is shown to be very limited in these hosts. Here, a high thermoelectric efficiency of ≈2 over a wide temperature range is reported in multiphase quaternary (PbTe)0.65(PbS)0.25(PbSe)0.1 compounds that are doped with sodium at concentrations greater than the solubility limits of the matrix. Although these compounds present room temperature thermoelectric efficiencies similar to sodium doped PbTe, a dramatically enhanced Hall carrier mobility at temperatures above 600 K for heavily doped compounds results in significantly enhanced thermoelectric efficiencies at elevated temperatures. This is achieved through the composition modulation doping mechanism resulting from heterogeneous distribution of the sodium dopant between precipitates and the matrix at elevated temperatures. These results can lead to further advances in designing high performance multiphase thermoelectric materials with intrinsically heterogeneous dopant distributions.  相似文献

Pseudomonas aeruginosa XMG, isolated from soil, utilizes lactate. Here we present a 6.45-Mb assembly of its genome sequence.
