The kinetics of renaturation of heat- or formamide-denatured DNA have been studied by following the change of optical density at a constant temperature. Solvents of different ionic strength and various DNA samples have been used. At the lower ionic strengths studied, the reaction follows second-order kinetics, substantiating the hypothesis that strands of native DNA separate upon denaturation and recombine during renaturation. As the ionic strength is increased at a constant temperature, the reaction deviates from simple second-order behavior. This appears to be the result of the inhibition to rewinding caused by short helical segments in the denatured DNA which are more stable at the higher ionic strenth.  相似文献

Specialization of the right cerebral hemisphere in man for analysis of spatial relations suggests that a similar specialization may exist in macaques. This possibility is being examined in animals first trained to negotiate a maze and then subjected to transection of optic chiasm and forebrain commissures. In the one successful experiment to date, clear evidence of such specialization was obtained for the left hemisphere in a monkey consistently using its left hand for retrieval of food. Provisional evidence for the existence of unilateral engrams in such circumstances was also obtained by limiting input during training to one eye and hemisphere and testing the other before and after completion of transection of the forebrain commissures. Finally, it was found that in the split brain macaque, a lesion in the parietal-occipital area of one hemisphere initially disrupted maze performance using either hemisphere.  相似文献

The hero shrew''s (Scutisorex somereni) massive interlocking lumbar vertebrae represent the most extreme modification of the vertebral column known in mammals. No intermediate form of this remarkable morphology is known, nor is there any convincing theory to explain its functional significance. We document a new species in the heretofore monotypic genus Scutisorex; the new species possesses cranial and vertebral features representing intermediate character states between S. somereni and other shrews. Phylogenetic analyses of DNA sequences support a sister relationship between the new species and S. somereni. While the function of the unusual spine in Scutisorex is unknown, it gives these small animals incredible vertebral strength. Based on field observations, we hypothesize that the unique vertebral column is an adaptation allowing these shrews to lever heavy or compressive objects to access concentrated food resources inaccessible to other animals.  相似文献

Plants that grow and thrive under abiotic stress often do so with the help of endophytic microorganisms. Although nitrogen‐fixing (diazotrophic) endophytes colonize many wild plants, these natural relationships may be disrupted in cultivated crop species where breeding and genotype selection often occur under conditions of intensive fertilization and irrigation. Many energy crops including corn may still benefit from diazotrophic endophyte inoculations allowing for more efficient biomass production with less input of petroleum‐derived fertilizer. A selection of diazotrophic endophytes isolated from willow (Salix sitchensis, Sitka willow) and poplar (Populus trichocarpa, black cottonwood) growing in nutrient‐poor river sides were used as inoculum in three experiments testing the effect on plant growth and leaf level physiology of a sweet corn variety under various levels of applied nitrogen fertilizer. We report substantial growth promotion with improved leaf physiology of corn plants in response to diazotrophic endophyte inoculations. Significant gains of early biomass with a greater root : shoot ratio were found for plants receiving endophytic inocula over the uninoculated control groups regardless of the nitrogen level. Furthermore, inoculated plants exhibited consistently higher rates of net CO2 assimilation than did those without endophytic inoculation. These results have beneficial implications for enhanced plant growth in a low‐input system on nutrient‐poor sites. The immediate increase of root mass observed in endophyte inoculated plants has the potential to provide better establishment and early growth in resource‐limited environments. The initial results of this study also indicate that the beneficial effect from endophytes isolated from poplar and willow species is not restricted to the species from which they were initially isolated.  相似文献

When an odorant is presented to one side of the nose and air to the other, the ability to localize which side received the odorant depends upon trigeminal nerve stimulation. It has been shown that performance on this lateralization task increases as stimulus concentration increases. In this study, we determined the influences of stimulus volume and sex on the ability to localize each of 8 odorants presented at neat concentrations: anethole, geraniol, limonene, linalool, menthol, methyl salicylate, phenyl ethanol, and vanillin. At a low stimulus volume (11 mL), only menthol was localized at an above-chance level. At a high stimulus volume (21 mL), above-chance localization occurred for all odorants except vanillin. Women were significantly better than men in localizing menthol. Stimuli rated as most intense were those that were most readily localized. The detection performance measures, as well as rated intensity values, significantly correlated with earlier findings of the trigeminal detectability of odorants presented to anosmic and normosmic subjects. This study suggests that differences in stimulus volume may explain some discrepant findings within the trigeminal chemosensory literature and supports the concept that vanillin may be a "relatively pure" olfactory stimulus.  相似文献

Indo-Pacific Lionfish (Pterois volitans and P. miles) are venomous marine fishes in the family Scorpaenidae that invaded the Caribbean Sea, Gulf of Mexico, and western North Atlantic Ocean beginning in the mid-1980s. Lionfish are generalist, opportunistic predators that consume a variety of invertebrates and small reef fishes, such that the presence of Lionfish can significantly reduce reef fish abundance, diversity, and recruitment on invaded reefs. This study focused on the feeding ecology of Lionfish in Biscayne National Park (BNP), located in southeast Florida, USA. BNP consists of multiple marine habitats, including mangroves, seagrass beds, coral reefs, and limestone keys that support a diverse array of species resulting in multi-million dollar fishing and tourism industries. These habitats within BNP are at risk from the predatory impacts of invasive Lionfish. Through morphological prey identification of stomach contents, supplemented with DNA barcoding for identification of highly-digested prey items, Lionfish diet was analyzed and compared among fish sizes (immature, transitional and mature), BNP region (bay, shelf, and edge), and seasons (wet and dry). A total of 513 stomachs, containing more than 2600 prey items, were examined. We report that Lionfish in BNP fed predominantly on small reef fishes and small crustaceans, with a dietary shift from crustaceans to fishes occurring with increasing Lionfish size. Diets differed among BNP regions for medium-sized (100–179 mm) transitional Lionfish but not for large-sized (≥?180 mm) mature individuals. Furthermore, dietary differences between seasons were observed in mature Lionfish, but no seasonal differences were detected for smaller Lionfish (i.e., immature and transitional Lionfish). Based on the diet habits observed, Lionfish in BNP could have significant ecological and economic consequences for BNP and south Florida coastal habitats.  相似文献

Bartonella infections are common in rodents. From 1994 to 2006, longitudinal studies of a rodent community, consisting mainly of deer mice (Peromyscus maniculatus), were conducted in southwestern Colorado to study hantaviruses. Blood samples from deer mice captured one or more times during the period 2003 to 2006 (n = 737) were selected to study bartonellae in deer mice. Bartonellae were found to be widely distributed in that population, with an overall prevalence of 82.4% (607/737 mice). No correlation was found between bartonella prevalence and deer mouse weight or sex. Persistent or successive infections with bartonellae were observed in deer mice captured repeatedly, with a prevalence of 83.9% (297/354), and the infection appeared to last for more than 1 year in some of them. Persistent infection with bartonellae may explain the high prevalence of these bacteria in deer mice at this site and, perhaps, elsewhere. Genetic analysis demonstrated that deer mouse-borne bartonella isolates at this site belong to the same species, B. vinsonii subsp. arupensis, demonstrating a specific relationship between B. vinsonii subsp. arupensis and deer mice.  相似文献

The relative influences of intertnal interval and sniff-bottlevolume on phenyl ethyl alcohol odor detection thresholds weredetermined in two experiments. In the first, thresholds weremeasured for 20 men and 20 women using 8, 16, 32 and 64 s intertrialintervals within a repeated measures counterbalanced design.No significant influence of intertnal interval was apparent,although females evidenced lower average thresholds than males.In the second, thresholds were established for 24 men and 24women in a similar design using sniff-bottles of 65, 120 and285 ml volumes. Sniff-bottle volume was inversely related tothe magnitude of the detection threshold measures. These findingssuggest that (i) rigid control of intertrial interval is notnecessary for the meaningful determination of phenyl ethyl alcoholdetection thresholds and (ii) sniff-bottle volume is significantlyrelated to such measures.  相似文献
