Antisera raised against vimentin, the protein subunit of nonspecific intermediate-sized filaments (IFs), were used in conjunction with neurofilament (NF) antisera to study the early development of neurons and glia in the rat embryo. Vimentin-positive fibers spanning the entire thickness of the neural tube including the cerebral vesicles were first observed on Day 12, concomitant with the appearance of NF protein in more confined areas (anterolateral regions of spinal cord and brain stem; motor roots emerging from the NF-positive areas). From Day 15 onwards vimentin and NF antisera selectively decorated glia and neurons, respectively, both in vivo and in vitro. Before Day 15 it appeared that NF-positive structures also stained with antivimentin in cryostat sections. In vitro experiments confirmed the presence of vimentin in early differentiating neurons. NF-positive cells were observed which also reacted with antivimentin in cultures obtained from 13- and 14-day embryos, but not later in development. Most neurons in these cultures became vimentin negative after 2–3 days in vitro.  相似文献

The organization of 14 exons covering 97% of the cDNA sequence of human cerebroside sulfate activator protein precursor has been determined from two overlapping EMBL-4 human genomic clones extending over 17kb. All exons and exon/intron splice junctions and five introns were sequenced. Exon 8 consists of only 9 bp and is involved in alternative splicing which generates three different mRNAs of cerebroside sulfate activator precursor.  相似文献

The structure of hen erythrocyte chromatin fibers was studied with the electron microscope. Chromatin fiber fragments with a length of about 5,000 Å and an average diameter of 320 Å are composed of 13 globular subunits (superbeads) which contain different numbers of nucleosomes. Their number average corresponds to 17 nucleosomes. — The interaction of lysine-rich histones with nucleosome chains was investigated by reconstitution experiments and was found to be semi-cooperative.  相似文献

Summary Liver glycogen synthase b phosphatase, chromatographically separable from phosphorylase a phosphatase, is decreased in 48-hour alloxan diabetic rats. The phosphatase activities are measured in an in vitro system using exogenous isolated phospho-enzyme as substrates with added phosphatases. A number of the temperature-sensitive mutants of Sindbis virus originally isolated and characterized by Burge and Pfefferkorn (1966, 1968) were reexamined for their abilities to grow and complement one another in cultured BHK-21 and Aedes albopictus (mosquito) cells. The response of the mutants to conditions of high and low temperature was similar in cultured cells of both the vertebrate and invertebrate hosts. Complementation experiments in BHK-21 cells produced growth patterns similar to those described by Burge and Pfefferkorn for chicken embryo fibroblast cells (1966) and placed the mutants into six nonoverlapping complementation groups. When examined in the cultured mosquito cells, only three of the nine mutants used in this study demonstrated complementation under a variety of experimental conditions. Homologous interference experiments demonstrated that the unusual patterns of complementation obtained in the A. albopictus cells did not result from an inefficient infection of the invertebrate cells by the mutants.  相似文献
