The keto (linear) form of exogenous fructose 1,6-bisphosphate, a highly charged glycolytic intermediate, may utilize a dicarboxylate transporter to cross the cell membrane, support glycolysis, and produce ATP anaerobically. We tested the hypothesis that fumarate, a dicarboxylate, and 3-phosphoglycerate (3-PG), an intermediate structurally similar to a dicarboxylate, could support contraction in vascular smooth muscle during hypoxia. To assess ATP production during hypoxia we measured isometric force maintenance in hog carotid arteries during hypoxia in the presence or absence of 20 mM fumarate or 3-PG. 3-PG improved maintenance of force (p < 0.05) during the 30-80 min period of hypoxia. Fumarate decreased peak isometric force development by 9.5% (p = 0.008) but modestly improved maintenance of force (p < 0.05) throughout the first 80 min of hypoxia. 13C-NMR on tissue extracts and superfusates revealed 1,2,3,4-13C-fumarate (5 mM) metabolism to 1,2,3,4-13C-malate under oxygenated and hypoxic conditions suggesting uptake and metabolism of fumarate. In conclusion, exogenous fumarate and 3-PG readily enter vascular smooth muscle cells, presumably by a dicarboxylate transporter, and support energetically important pathways. 相似文献

Failure of a continuously aerated sequencing batch reactor (SBR) pilot plant-enhanced biological phosphorus removal (EBPR) process, designed to remove phosphorus from the clarified effluent from a conventional non-EBPR wastewater treatment plant, was associated with the dominance ( c . 50% of the biovolume) of gammaproteobacterial coccobacilli. Flow cytometry and subsequent clone library generation from an enriched population of these Gammaproteobacteria showed that their 16S rRNA genes were most similar to partial clone sequences obtained from an actively denitrifying SBR community, and from anaerobic : aerobic EBPR communities. Under the SBR operating conditions used here, these cells stained for poly-β-hydroxyalkanoates, but never polyphosphate. Applying FISH probes designed against them in combination with microautoradiography showed that they could also assimilate acetate 'aerobically'. FISH analyses of biomass samples from the full-scale treatment plant providing the pilot plant feed showed that they were present there in high numbers. However, they were not detected by FISH in laboratory-scale communities of the same aerated laboratory-scale EBPR process even when EBPR had failed, or from several full-scale EBPR plants or other activated sludge processes. 相似文献

Women's waist-to-hip ratio (WHR) varies with age, and a lower WHR is associated with a higher estrogen-to-androgen ratio and possibly higher fecundity, at least in some populations. Consequently, it has been argued that selection has favored a universal male preference for a low female WHR. In previous studies using frontal pictures, men in the United States preferred a low WHR of 0.7, but men among Hadza hunter–gatherers and a few other small-scale societies preferred higher ratios. Unlike the actual WHR of women, measured with a tape around the waist and the hips and buttocks, the WHR in frontal pictures excludes the buttocks. Because frontal WHR gives only a partial picture, we used profile views of women to measure men's preferences for the profile WHR. Hadza men preferred a lower profile WHR (more protruding buttocks) than American men. Since Hadza men preferred higher frontal WHR but lower profile WHR, and since both contribute to the actual WHR, these results imply there is less disparity between American and Hadza preferences for the actual WHR of real women. We suggest men's preferences vary with the geographic variation in the shape of women who have wider hips in some populations and more protruding buttocks in others. 相似文献

Plant community distribution along water table and grazing gradients in montane meadows of the Sierra Nevada Range (California, USA) 总被引:1,自引:0,他引:1
This study aimed to identify dominant plant communities across five wet and mesic meadows in the Sierra Nevada Range (California, USA) and examine the impacts of environmental and grazing gradients on plant community distribution and diversity. Species composition and environmental conditions were recorded in 100 plots over two years. Classification and ordination analyses were used to classify plant communities and identify relationships between community types and both environmental and grazing gradients. We identified the following six plant community types: Carex jonesii, Carex leporinella, Carex nebrascensis, Carex utriculata, Eleocharis pauciflora, and Veratrum californicum. We found strong connections between plant communities and water table variables, with low water table (r 2?=?0.56) and mean water table (r 2?=?0.30) significantly correlated with Axis 1 while high water table (r 2?=?0.29) and elevation were correlated with Axis 3 (r 2?=?0.49). We found significant differences among community types for all three water table variables and for elevation. We found no correlation between grazing and community type classification, but there was a significant difference in grazing levels among community types. The plant communities and relationships to water table found in this work may aid managers in understanding present conditions and identifying future changes in meadow ecosystems. 相似文献

Cu homeostasis depends on a tightly regulated network of proteins that transport or sequester Cu, preventing the accumulation of this toxic metal while sustaining Cu supply for cuproproteins. In Rhodobacter capsulatus, Cu‐detoxification and Cu delivery for cytochrome c oxidase (cbb3‐Cox) assembly depend on two distinct Cu‐exporting P1B‐type ATPases. The low‐affinity CopA is suggested to export excess Cu and the high‐affinity CcoI feeds Cu into a periplasmic Cu relay system required for cbb3‐Cox biogenesis. In most organisms, CopA‐like ATPases receive Cu for export from small Cu chaperones like CopZ. However, whether these chaperones are also involved in Cu export via CcoI‐like ATPases is unknown. Here we identified a CopZ‐like chaperone in R. capsulatus, determined its cellular concentration and its Cu binding activity. Our data demonstrate that CopZ has a strong propensity to form redox‐sensitive dimers via two conserved cysteine residues. A ΔcopZ strain, like a ΔcopA strain, is Cu‐sensitive and accumulates intracellular Cu. In the absence of CopZ, cbb3‐Cox activity is reduced, suggesting that CopZ not only supplies Cu to P1B‐type ATPases for detoxification but also for cuproprotein assembly via CcoI. This finding was further supported by the identification of a ~150 kDa CcoI‐CopZ protein complex in native R. capsulatus membranes. 相似文献
