In the field of biomechanics, the offer of models which are more and more realistic requires to integrate a physiological response, in particular, the controlled muscle bracing and the reflexes. The following work aims to suggest a unique methodology which couples together a sensory and motor loop with a finite element model. Our method is applied to the study of the oscillation of the elbow in the case of a biceps brachial stretch reflex. The results obtained are promising in the purpose of the development of reactive human body models.  相似文献

Hutchinson–Gilford progeria syndrome (HGPS) is caused by an LMNA mutation that results in the production of the abnormal progerin protein. Children with HGPS display phenotypes of premature aging and have an average lifespan of 13 years. We found earlier that the targeting of the transmembrane protein PLA2R1 overcomes senescence and improves phenotypes in a mouse model of progeria. PLA2R1 is regulating the JAK/STAT signaling, but we do not yet know whether targeting this pathway directly would influence cellular and in vivo progeria phenotypes. Here, we show that JAK1/2 inhibition with ruxolitinib rescues progerin‐induced cell cycle arrest, cellular senescence, and misshapen nuclei in human normal fibroblasts expressing progerin. Moreover, ruxolitinib administration reduces several premature aging phenotypes: bone fractures, bone mineral content, grip strength, and a trend to increase survival in a mouse model of progeria. Thus, we propose that ruxolitinib, an FDA‐approved drug, should be further evaluated as a drug candidate in HGPS therapy.  相似文献

John W. Salter's papers of 1856 and 1857 reported trace and body fossils from rocks of the Longmyndian Supergroup, Shropshire, that conventional wisdom had deemed literally “Azoic.” The significance of this work is reflected by its mention in On the Origin of Species, where it is cited as evidence for the existence of life prior to the Cambrian radiation. This study of Salter's historic specimens combined with recent field studies confirms that these structures likely represent microbial rather than metazoan markings. Nevertheless, this review confirms Salter as the unheralded founder of Precambrian palaeontology, many years before the existence of a Precambrian fossil record was widely known. This study also gives credit to a highly skilled palaeontologist, who appears to have struggled with psychological problems throughout his life. Salter had once been Adam Sedgwick's “youthful and cheerful companion” in the field, prior to embarking on an initially successful Geological Survey career. He was a widely renowned expert on Palaeozoic palaeontology, especially trilobites, but eventually fell into serious depression, which culminated in his suicide in 1869. Study and reinterpretation of his original materials reaffirms the importance of Salter's discoveries, and the Longmynd for our understanding of late Ediacaran palaeobiology.  相似文献

Abstract

The inactivation of Pseudomonas aeruginosa biofilms grown on glass under high shear stress and exposed to a range of dissolved ozone concentrations (2, 5 and 7?ppm) at 10 and 20?min was investigated. The regression equation, log reduction (biofilm)?=?0.64?+?0.59×(C – 2)?+?0.33×(T – 10), described the dependence of biofilm inactivation on the dissolved ozone concentration (C, ppm) and contact time (T, min). The predicted D-values were 11.1, 5.7 and 2.2?min at 2, 5 and 7?ppm, respectively. Inactivation of biofilms grown on various surfaces was tested at a single dissolved ozone concentration of 5?ppm and a single exposure time of 20?min. Biofilms grown on plastic materials showed inactivation results similar to that of biofilms on glass, while biofilms grown on ceramics were statistically significantly more difficult to inactivate, suggesting the importance of utilizing non-porous materials in industrial and clinical settings.  相似文献

Elephant seals maintain rates of endogenous glucose production (EGP) typical of post-absorptive mammals despite enduring prolonged periods of food deprivation concurrent with low rates of glucose oxidation. These high rates of EGP suggest extensive glucose recycling during fasting. We investigated lactate metabolism in fasting elephant seals to assess its role in glucose recycling. Whole-animal glucose and lactate fluxes were measured as the rates of appearance of glucose and lactate (Ra gluc and Ra lac, respectively) using a primed constant infusion of [U-14C] lactate and [6-3H] glucose, and we calculated the minimum contribution of lactate to gluconeogenesis (GNG lac). Ra lac was high compared to resting values in other species (3.21 ± 0.71 mmol min?1* kg?1), did not change between 14 ± 1 and 31 ± 8 days of fasting and varied directly with Ra glu. The minimum GNG lac was 44.6 ± 6.0 % of EGP, varied directly with plasma lactate levels, and did not change over the fast. Ra lac and Ra glu both varied directly with plasma insulin concentrations. These data suggest that lactate is the predominant gluconeogenic precursor in fasting elephant seals and that high rates of glucose recycling through Cori cycle activity contribute to the maintenance of EGP during fasting. High levels of Cori cycle activity and EGP may be important components of metabolic adaptations that maintain glucose production while avoiding ketosis during extended fasting or are related to sustained metabolic alterations associated with extended breath-holds in elephant seals.  相似文献

We present evidence from ethnography and experimental processing of foxtail millet (Setaria italica (L.) P. Beauv.) in China that spikelets containing incompletely filled (or immature) grains constitute a significant portion of typical millet harvests and are removed along with other by-products after threshing and winnowing. This study provides a baseline for the identification of immature foxtail grains in archaeobotanical assemblages. Immature millet grains are a frequent component of archaeobotanical assemblages in Neolithic and Bronze Age China, and criteria for their recognition are presented based on our modern experimental result and illustrated with archaeobotanical examples from Shandong and Henan. It is seed morphology rather than size that plays a determinative role in the identification of foxtail millet. It is suggested that those grains with a narrow egg-shaped embryo, which is about 5/6 of the whole grain, and having a round shape can be classed as foxtail millet even though they are small, flat and squashed. While different grades of immaturity in millet grains might be defined, the interpretative potential of these appears to be negligible as all immature grains are concentrated in winnowing waste. This study confirms the suggestion that the ratio of immature to mature millet grains can be employed in archaeobotany in considering whether or not early stage crop processing (threshing and winnowing) contributed to the formation of particular archaeological millet assemblages.  相似文献

The complexity of behavioural interactions in predator-prey systems has recently begun to capture trait-effects, or non-lethal effects, of predators on prey via induced behavioural changes. Non-lethal predation effects play crucial roles in shaping population and community dynamics, particularly by inducing changes to foraging, movement and reproductive behaviours of prey. Prey exhibit trade-offs in behaviours while minimizing predation risk. We use a novel evolutionary ecosystem simulation EcoSim to study such behavioural interactions and their effects on prey populations, thereby addressing the need for integrating multiple layers of complexity in behavioural ecology. EcoSim allows complex intra- and inter-specific interactions between behaviourally and genetically unique individuals called predators and prey, as well as complex predator-prey dynamics and coevolution in a tri-trophic and spatially heterogeneous world. We investigated the effects of predation risk on prey energy budgets and fitness. Results revealed that energy budgets, life history traits, allocation of energy to movements and fitness-related actions differed greatly between prey subjected to low-predation risk and high-predation risk. High-predation risk suppressed prey foraging activity, increased total movement and decreased reproduction relative to low-risk. We show that predation risk alone induces behavioural changes in prey which drastically affect population and community dynamics, and when interpreted within the evolutionary context of our simulation indicate that genetic changes accompanying coevolution have long-term effects on prey adaptability to the absence of predators.  相似文献

As evidence mounts that the feral Cat (Felis catus) is a significant threat to endemic Australian biodiversity and impedes reintroduction attempts, uncertainty remains about the impact a residual population of cats following control will have on a mammal reintroduction programme. Also, behavioural interactions between cats and their prey continue to be an area of interest. Within the framework of an ecosystem restoration project, we tested the hypotheses that successful reintroductions of some medium‐sized mammals are possible in locations where feral cats are controlled (but not eradicated) in the absence of European Red Fox (Vulpes vulpes), and that hare‐wallabies that dispersed from their release area are more vulnerable to cat predation compared with those that remain at the release site. We used radiotelemetry to monitor the survivorship and dispersal of 16 Rufous Hare‐wallabies (Lagorchestes hirsutus spp.) and 18 Banded Hare‐wallabies (Lagostrophus fasciatus fasciatus) reintroduced to four sites within Shark Bay, Western Australia. Nearly all foxes were removed and feral cats were subject to ongoing control that kept their indices low relative to prerelease levels. All monitored hare‐wallabies were killed by cats within eight and 10 months following release. Significant predation by feral cats was not immediate: most kills occurred in clusters, with periods of several months where no mortalities occurred. Once a hare‐wallaby was killed, however, predation continued until each population was eliminated. Animals remaining near their release site survived longer than those that dispersed. The aetiology of predation events observed offers new insights into patterns of feral cat behaviour and mammal releases. We propose a hypothesis that these intense per capita predation events may reflect a targeted hunting behaviour in individual feral cats. Even where feral cats are controlled, the outcome from consistent predation events will result in reintroduction failures. Managers considering the reintroduction of medium‐sized mammals in the presence of feral cats should, irrespective of concurrent cat control, consider the low probability of success. We advocate alternative approaches to cat‐baiting alone for the recovery of cat‐vulnerable mammals such as hare‐wallabies.  相似文献

Neomegalonema perideroedes (formerly Meganema perideroedes) str. G1 is the type strain and only described isolate of the genus Neomegalonema (formerly Meganema) which belongs to the Alphaproteobacteria. N. perideroedes is distinguished by the ability to accumulate high amounts of polyhydroxyalkanoates and has been associated with bulking problems in wastewater treatment plants due to its filamentous morphology. In 2013, its genome was sequenced as part of the Genomic Encyclopedia of Bacteria and Archaea (GEBA), which aims to improve the sequencing coverage of the poorly represented regions of the bacterial and archaeal branches of the tree of life. As N. perideroedes str. G1 is relatively distantly related to well described species—being the only sequenced member of its proposed family—the in silico prediction of genes by nucleotide homology to reference genes might be less reliable. Here, a proteomic dataset for the refinement of the N. perideroedes genome annotations is generated which clearly indicates the shortcomings of high‐throughput in silico genome annotation.  相似文献

Prion diseases are fatal neurodegenerative disorders caused by proteinaceous infectious pathogens termed prions (PrP(Sc)). To date, there is no prophylaxis or therapy available for these transmissible encephalopathies. Passive immunization with monclonal antibodies recognizing the normal host-encoded prion protein (PrP(C)) has been reported to abolish PrP(Sc) infectivity and to delay onset of disease. Because of established immunologic tolerance against the widely expressed PrP(C), active immunization appears to be difficult to achieve. To overcome this limitation, papillomavirus-like particles were generated that display a nine amino acid B-cell epitope, DWEDRYYRE, of the murine/rat prion protein in an immunogenic capsid surface loop, by insertion into the L1 major capsid protein of bovine papillomavirus type 1. The PrP peptide was selected on the basis of its previously suggested central role in prion pathogenesis. Immunization with PrP-virus-like particles induced high-titer antibodies to PrP in rabbit and in rat, without inducing overt adverse effects. As determined by peptide-specific ELISA, rabbit immune sera recognized the inserted murine/rat epitope and also cross-reacted with the homologous rabbit/human epitope differing in one amino acid residue. In contrast, rat immune sera recognized the murine/rat peptide only. Sera of both species reacted with PrP(C) in its native conformation in mouse brain and on rat pheochromocytoma cells, as determined by immunoprecipitation and fluorescence-activated cell sorting analysis. Importantly, rabbit anti-PrP serum contained high-affinity antibody that inhibited de novo synthesis of PrP(Sc) in prion-infected cells. If also effective in vivo, PrP-virus-like particle vaccination opens a unique possibility for immunologic prevention of currently fatal and incurable prion-mediated diseases.  相似文献
