Synthesis of indole-3-acetylaspartic acid

A simple synthesis of indole-3-acetyl-d,l-aspartic acid and its l-isomer is described and their physical properties are listed.     

Fixation of 14CO2 in tissue cultured red raspberry prior to and after transfer to soil

The ¹⁴CO₂ uptake of an aseptically cultured red raspberry clone (Rubus ideaus L.) was examined prior to and after transfer to soil. Individual leaves of transplants, both persistent from culture and new ones, were tested 5 weeks after transplant for ¹⁴CO₂ uptake capability. Transplant leaves of successive weekly age classes took up ¹⁴CO₂ at increasing rates per unit area, displaying a spectrum of photosynthetic competence from low levels close to that of leaves from culture, to that of control plants. This is illustrative of acclimatization to the soil environment and was related to transplant light intensity.     

A simple model for calculating residential 60-Hz magnetic fields

A model is presented that permits the calculation of densities of 60-Hz magnetic fields throughout a residence from only a few measurements. We assume that residential magnetic fields are produced by sources external to the house and by the residential grounding circuit. The field from external sources is measured with a single probe. The field produced by the grounding circuit is calculated from the current flowing in the circuit and its geometry. The two fields are combined to give a prediction of the total field at any point in the house. A data-acquisition system was built to record the magnitude and phase of the grounding current and the field from external sources. The model's predictions were compared with measurements of the total magnetic field at a single location in 23 houses; a correlation coefficient of .87 was obtained, indicating that the model has good predictive capability. A more detailed study that was carried out in one house permitted comparisons of measurements with the model's predictions at locations throughout the house. Again, quite reasonable agreement was found. We also investigated the temporal variability of field readings in this house. Daily magnetic field averages were found to be considerably more stable than hourly averages. Finally, we demonstrate the use of the model in creating a profile of the magnetic fields in a home.     

Formaldehyde treated gelatin as a gel exclusion agent.

A high molecular weight fraction of gelatin precipitated from a solution of Difco gelatin with polyethylene glycol has several favourable properties in gel exclusion chromatography. To manufacture the gel exclusion agent, the gelatin was dissolved in hot water, allowed to set in the cold and rendered insoluble by reaction with formaldehyde. When finely disrupted and used as bed material in columns this gel is capable of separating protein components from mixtures of proteins of widely different molecular weights. Favourable properties include elasticity at relatively low concentration, its availability and its inexpensive nature.     

Down-regulation of Ia expression on macrophages by sea star factor

Sea star factor (SSF), a protein of 39,000 Da isolated from the coelomocytes of Asterias forbesi, was found to inhibit the induction of Ia expression on murine macrophages by concanavalin A supernatants. Addition of SSF to cultured macrophages at the same time as the lymphokine preparation significantly reduced the percentage Ia+ cells after 5 days culture, compared to cultures given lymphokine only. Intraperitoneal injection of SSF also reduced the percentage Ia+ peritoneal exudate macrophages by three-fourths in Listeria-infected mice. Addition of the cyclo-oxygenase inhibitor, indomethacin, to macrophage cultures reversed this Ia-suppressive effect of SSF. Since macrophages from endotoxin-unresponsive and endotoxin-responsive mice were both sensitive to the Ia-inhibitory effect of SSF, the induction of arachidonic acid metabolism and the inhibition of Ia appear to be independent of the action of endotoxin. The SSF-induced down regulation of Ia expression may be a major factor in the suppression of primary immune responses to T-dependent antigens previously noted in studies with SSF.     

Gentamicin-based medium for the isolation of group D streptococci and application of the medium to water analysis.

Gentamicin-thallous-carbonate (GTC) medium contained (per liter): 40.0 g of Trypticase soy agar, 5.0 g of KH(2)PO(4), 2.0 g of NaHCO(2), 1.0 g of glucose, 1.0 g of esculin, 0.5 g of thallous acetate (TA), 0.5 g of ferric citrate, 0.75 ml of Tween 80, and 2.5 mg of gentamicin sulfate. The NaHCO(3) (20 ml of a 10% solution that had been heated to boiling) was added after sterilization of the basal medium. The spread plate technique was used to compare GTC agar with Pfizer selective enterococcus, TA, and KF agars by using sewage as well as bovine and swine fecal samples. Significantly greater numbers of group D streptococci were recovered on GTC agar than on Pfizer selective enterococcus or KF agars, within and over all samples. Higher counts also were obtained on GTC than on TA agar, but the differences were not statistically significant. The percentage of false positives was about the same for all four media. Samples of riverwater also were plated on GTC, TA, and KF agars, and significantly higher recoveries were obtained with GTC agar. GTC agar was superior to the other media examined primarily because of increased recoveries of Streptococcus bovis and S. equinus; other advantages of GTC agar were large colony size and short (24-h) incubation period. The percentage of false positives from riverwater was 13% for GTC agar and 0% for TA and KF agars; therefore, confirmation would be necessary when GTC agar is used with some types of environmental samples.     

Challenges and progress in setting a standard for PCR for invasive aspergillosis

Despite extensive experience in the past decade using polymerase chain reaction for diagnosing aspergillosis, the technique was not included as mycologic evidence in either the first consensus definitions of invasive fungal disease or in recent revisions because there was no standardized or validated method. Consequently, several research groups decided to join forces and form a working party of the International Society of Human and Animal Mycology to establish a standard for Aspergillus polymerase chain reaction testing. This group opted for screening tests on blood because this is most likely to find a place in integrated care pathways for managing invasive aspergillosis. Progress is being made such that a standard will be proposed in the near future. This standard will then be formally validated in a prospective trial.
