The concentration of lignin in plant tissue is a major factor controlling organic matter degradation rates in forest ecosystems. Microbial biomass and lignin and cellulose decomposition were measured for six weeks in forest soil microcosms in order to determine the influence of pH, moisture, and temperature on organic matter decomposition. Microbial biomass was determined by chloroform fumigation; lignin and cellulose decomposition were measured radiometrically. The experiment was designed as a Latin square with soils of pH of 4.5, 5.5, and 6.5 adjusted to 20, 40, or 60% moisture content, and incubated at temperatures of 4, 12, or 24°C. Microbial biomass and lignin and cellulose decomposition were not significantly affected by soil acidity. Microbial biomass was greater at higher soil moisture contents. Lignin and cellulose decomposition significantly increased at higher soil temperatures and moisture contents. Soil moisture was more important in affecting microbial biomass than either soil temperature or soil pH.  相似文献

The fragile site at Xq27.3 is an unstable microsatellite repeat, p(CCG)n. In fragile-X syndrome pedigrees, this sequence exhibits variable amplification, the length of which correlates with fragile-site expression. There is a direct relationship between increased p(CCG)n copy number and propensity for instability: individuals having large amplifications exhibit somatic variation due to increased instability. The instability of the p(CCG)n repeat, when transmitted through affected pedigrees, explains the unusual segregation patterns of fragile-X phenotype, referred to as the Sherman paradox. All individuals of fragile-X genotype were found (where testing was possible) to have a parent with amplified p(CCG)n repeat, indicating that few, if any, cases of fragile-X syndrome are not familial.  相似文献

Two commonly used measures of genetic diversity for intraspecies DNA sequence data are based, respectively, on the number of segregating sites, and on the average number of pairwise nucleotide differences. Expressions are derived for their variance in the presence of intragenic recombination for a panmictic population of fixed size that is at neutral equilibrium at the region sequenced. We show that, in contrast to the slow decrease in variance with increasing sample size, if the recombination rate is nonzero, the asymptotic rate of decrease of variance with increasing sequence length, for fixed sample size, is quite rapid. In particular, it is close to that which would be obtained by sequencing independent chromosome regions. The correlation between measures of diversity from linked regions is also examined. For a given total number of bases sequenced in a particular region, optimal sequencing strategies are derived. These typically involve sequencing relatively few (three to 10) long copies of the region. Under optimal strategies, the variances of the two measures are very similar for most parameter values considered. Results concerning optimal sequencing strategies will be sensitive to gross departures from the underlying assumptions, such as population bottlenecks, selective sweeps, and substantial population substructure.  相似文献

During the last decade increasing attention has been given to the role of free radicals in biological oxidations. The subject has been of increasing interest to both the food scientist and the physiologist. Free radical scavengers in the form of both indigenous and added antioxidants are necessary for the successful preservation of food; free radicals are increasingly being implicated in the onset of, among others, ischaemic heart disease and for protection against these diseases it is suggested that the dietary intake of the antioxidant vitamins should be increased especially for diets high in polyunsaturated fats. 1,2 Convenience and snack foods which absorb substantial amounts of frying oils are being increasingly consumed. Since poly-unsaturated fatty acids are particularly susceptible to oxidation by free radicals during the storage, cooking and frying of foods, the potential risk of exposure to lipid degradation products' is likely to have increased. In foods the non-enzymic and lipoxygenase catalysed oxidation of polyunsaturated fatty acids, β-carotene and vitamin A can result in the loss of essential nutrients and the development of off-flavours.  相似文献

Water level, ash content, proximate (protein, lipid, carbohydrate and chitin) and elemental (carbon and nitrogen) composition were analyzed in twentythree species of Antarctic Zooplankton collected during the austral fall (1986) and winter (1988) from the Scotia/Weddell Sea region. Extremes in water level, ash content and organic components were typified by copepods and gelatinous forms. Ostracods and polychaetes were generally similar in composition to copepods, being only slightly higher in water level and ash content. Chaetognaths exhibited a composition intermediate in character with some components similar in value to that shown by crustaceans (i.e. protein) while other components were more in the range of values seen in gelatinous forms (i.e. water level and ash content). Protein was the major proximate component and measured values (as % Afdw) were fairly uniform among non-gelatinous species (x=33.9±6.9). Lipid levels were variable, with high values (>30% AFDW) only found for the copepods Calanoides acutus, Calanus propinquus and Euchaeta antarctica. Carbohydrate values were low in all species examined. Chitin was measured in crustacean species only. With the exception of C. acutus (x=2.5% AFDW chitin), values were similar among species with mean values being slightly higher in fall (x=11.8±2.5) than in winter (x=6.7±1.8). Among non-gelatinous species, the ratio of carbon to nitrogen was positively correlated with the lipid to protein ratio, underscoring the compositional association between elemental and proximate components in these groups. In gelatinous species, the relationship between carbon:nitrogen and lipid:protein was inconsistent and less pronounced. Caloric content was estimated from recovered organic matter for nongelatinous species. As a function of wet weight and dry weight, values reflected differences in water level and ash content among individual species. As a function of ashfree dry weight, values were similar among all species (x=3.6±0.9 kcal/g).Seasonal comparisons were possible for 12 of the 23 species. Among crustaceans, changes in water level and organic components were variable reflecting dissimilar trophic, reproductive or ecological habits among different species. Essentially no change in composition between fall and winter was observed for diapause species (e.g. Calanoides acutus and Rhincalanus gigas) as well as for omnivorous/ carnivorous species (e.g. Gaetanus tenuispinus). Conversely, large compositional changes were evident for Calanus propinquus, a small-particle grazer that relies heavily on lipid reserves. Chaetognaths and some gelatinous species exhibited a considerable decrease in ash content from fall to winter which, for most cases, was mirrored by some degree of increase in lipid level. At present, however, scant data are available to help explain the observed patterns of compositional change within non-crustacean species.  相似文献

Obesity and low levels of physical and metabolic fitness are risk factors for cardiovascular disease and diabetes. The purpose of this investigation was to attenuate obesity and improve physical and metabolic fitness in elementary school children. Schools have the opportunity, mechanisms, and personnel in place to deliver nutrition education, fitness activities, and a school food service that is nutritious and healthy. Cohorts from grades 3 to 5 in two school districts in rural Nebraska (Intervention/Control) participated in a 2-year study of physical activity and modified school lunch program. Data collection for aerobic capacity, body composition, blood chemistry, nutrition knowledge, energy intake, and physical activity was at the beginning and end of each year. Int received enhanced physical activity, grade specific nutrition education, and a lower fat and sodium school lunch program. Con continued with a regular school lunch and team sports activity program. At year 2, Int lunches had significantly less energy (9%), fat (25%), sodium (21%), and more fiber (17%). However, measures of 24-hour energy intake for Int and Con showed significant differences for sodium only. Physical activity in the classroom was 6% greater for Int compared to Con (p < 0.05) but physical activity outside of school was ?16 % less for Int compared to Con (p < 0.05). Body weight and body fat were not different between schools for normal weight or obese children. No differences were found for cholesterol, insulin, and glucose; however, HDL cholesterol was significantly greater and cholesterol/HDL was significantly less for Int compared to Con (p < 0.05). It appears that compensation in both energy intake and physical activity outside of school may be responsible for the lack of differences between Int and Con.  相似文献

The paper is concerned with methods for the estimation of the coalescence time (time since the most recent common ancestor) of a sample of intraspecies DNA sequences. The methods take advantage of prior knowledge of population demography, in addition to the molecular data. While some theoretical results are presented, a central focus is on computational methods. These methods are easy to implement, and, since explicit formulae tend to be either unavailable or unilluminating, they are also more useful and more informative in most applications. Extensions are presented that allow for the effects of uncertainty in our knowledge of population size and mutation rates, for variability in population sizes, for regions of different mutation rate, and for inference concerning the coalescence time of the entire population. The methods are illustrated using recent data from the human Y chromosome.  相似文献

Mapping of a Gene for Long QT Syndrome to Chromosome 4q25-27   总被引:21,自引:0,他引:21  
Long QT syndrome (LQTS) is a heterogeneous inherited disorder causing syncope and sudden death from ventricular arrhythmias. A first locus for this disorder was mapped to chromosome 11p15.5. However, locus heterogeneity has been demonstrated in several families, and two other loci have recently been located on chromosomes 7q35-36 and 3p21-24. We used linkage analysis to map the locus in a 65-member family in which LQTS was associated with more marked sinus bradycardia than usual, leading to sinus node dysfunction. Linkage to chromosome 11p15.5, 7q35-36, or 3p21-24 was excluded. Positive linkage was obtained for markers located on chromosome 4q25-27. A maximal LOD score of 7.05 was found for marker D4S402. The identification of a fourth locus for LQTS confirms its genetic heterogeneity. Locus 4q25-27 is associated with a peculiar phenotype within the LQTS entity.  相似文献
