The treatment of the simulated lead-contaminated solid waste by composting with white-rot fungus was studied at laboratory scale. The composting system without the inocula of white-rot fungus was prepared as control, and the composting of the uncontaminated solid waste with the inocula of white-rot fungus was carried out as the other control. The results indicated that the solid waste inoculated with white-rot fungus could be successfully processed. The final compost was mature with 70.5% of lead (Pb) in residual fraction and none in exchangeable fraction. Germination index reached 120%. All the results indicated that the bioavailability of Pb in compost was reduced and the potential harm of Pb in compost was alleviated by composting with the inocula of white-rot fungus.  相似文献

This study attempted to investigate the time course of meiotic progression after transferring primary spermatocyte (PS) into ooplasm at different maturing stages. In present experiments, PSs were introduced into maturing ooplasts or oocytes by electrofusion. Higher fusion rate was obtained by phytohemagglutinin (PHA) agglutination than by perivitelline space (PVS) insertion. When the ooplasms prepared at 0, 2, 5, and 8.5 hr of in vitro maturation (IVM) were used as recipients and PSs were used as donors, the reconstructed cells extruded the first polar body (PB1) approximately 8.5, 7, 5.5, and 3 hr after electrofusion, respectively. Especially, when ooplasm cultured for 8.5 hr in vitro after GV removal was fused with PS, the PB1 was emitted 7-11 hr after electrofusion. Additionally, the PB1 extrusions of GV and pro-MI oocytes fertilized with PSs were 2.5 hr earlier than control oocytes. The results suggest that (1) PSs undergo the first meiosis in different time courses when introduced into ooplasm at different maturing stages; (2) GV material plays an important role in determining the timing of PB1 extrusion; and (3) first meiotic division of GV and pro-MI oocytes can be accelerated by introducing PS.  相似文献

This study investigates the therapeutic effect and the underlying mechanisms of ergothioneine (EGT) on the testicular damage caused by varicocele (VC) in vivo, in vitro, and in silico. This preclinical study combines a series of biological experiments and network pharmacology analyses. A total of 18 Sprague Dawley (SD) male rats were randomly and averagely divided into three groups: the sham-operated, VC model, and VC model with EGT treatment (VC + EGT) groups. The left renal vein of the VC model and the VC + EGT groups were half-ligated for 4 weeks. Meanwhile, the VC + EGT group was intragastrically administrated with EGT (10 mg/kg). GC1 and GC2 cells were exposed to H2O2 with or without EGT treatment to re-verify the conclusion. The structure disorder of seminiferous tubules ameliorated the apoptosis decrease in the VC rats receiving EGT. EGT can also increase the sperm quality of the VC model rats (p < 0.05). The exposure to H2O2 decreased proliferation and increased apoptosis of GC1 and GC2 cells, which was revisable by adding EGT to the plates (p < 0.05). The network pharmacology and molecular docking were conducted to explore the potential targets of EGT in VC, and HSP90AA1 was identified as the pivotal gene, which was validated by western blot, immunohistochemistry, and RT-qPCR both in vivo and in vitro (p < 0.05). Overall, EGT attenuates the testicular injury in the VC model both in vivo and in vitro by potentially potentiating the expression of HSP90AA1.  相似文献

Members of the Nanorana genus (family Dicroglossidae) are often referred to as excellent model species with which to study amphibian adaptations to extreme environments and also as excellent keystone taxa for providing insights into the evolution of the Dicroglossidae. However, a complete mitochondrial genome is currently only available for Nanorana pleskei. Thus, we analyzed the complete mitochondrial genomes of Nanorana parkeri and Nanorana ventripunctata to investigate their evolutionary relationships within Nanorana and their phylogenetic position in the family Dicroglossidae. Our results showed that the genomes of N. parkeri (17,837 bp) and N. ventripunctata (18,373 bp) encode 13 protein‐coding genes (PCGs), two ribosomal RNA genes, 23 transfer RNA (tRNA) genes, and a noncoding control region. Overall sequences and genome structure of the two species showed high degree of similarity with N. pleskei, although the motif structures and repeat sequences of the putative control region showed clear differences among these three Nanorana species. In addition, a tandem repeat of the tRNA‐Met gene was found located between the tRNA‐Gln and ND2 genes. On both the 5′ and 3′‐sides, the control region possessed distinct repeat regions; however, the CSB‐2 motif was not found in N. pleskei. Based on the nucleotide sequences of 13 PCGs, our phylogenetic analyses, using Bayesian inference and maximum‐likelihood methods, illustrate the taxonomic status of Nanorana with robust support showing that N. ventripunctata and N. pleskei are more closely related than they are to N. parkeri. In conclusion, our analyses provide a more robust and reliable perspective on the evolutionary history of Dicroglossidae than earlier analyses, which used only a single species (N. pleskei).  相似文献

The prevalence of SARS-CoV-2 variants of concern (VOCs) is still escalating throughout the world. However, the level of neutralization of the inactivated viral vaccine recipients’ sera and convalescent sera against all VOCs, including B.1.1.7 (Alpha), B.1.351 (Beta), P.1 (Gamma), B.1.617.2 (Delta), and B.1.1.529 (Omicron) remains to be lack of comparative analysis. Therefore, we constructed pseudoviruses of five VOCs using a lentiviral-based system and analyzed their viral infectivity and neutralization resistance to convalescent and BBIBP-CorV vaccinee serum at different times. Our results show that, compared with the wild-type strain (WT), five VOC pseudoviruses showed higher infection, of which B.1.617.2 and B.1.1.529 variant pseudoviruses exhibited higher infection rates than wild-type or other VOC strains, respectively. Sera from 10 vaccinated individuals at the 1, 3 and 5-month post second dose or from 10 convalescent at 14 and 200 days after discharge retained neutralizing activity against all strains but exhibited decreased neutralization activity significantly against the five VOC variant pseudoviruses over time compared to WT. Notably, 100% (30/30) of the vaccinee serum samples showed more than a 2.5-fold reduction in neutralizing activity against B.1.1.529, and 90% (18/20) of the convalescent serum samples showed more than 2.5-fold reduction in neutralization against B.1.1.529. These findings demonstrate the reduced protection against the VOCs in vaccinated and convalescent individuals over time, indicating that it is necessary to have a booster shot and develop new vaccines capable of eliciting broad neutralization antibodies.  相似文献

Moderate allele variation was found in these loci with two to eight alleles per locus. The advancement of leaf senescence is accompanied by a reduction in cellular protein content together with the induction of specific proteins which are probably involved in the process. In the present study, with parsley, we followed the changes in the levels of proteins functionally and immunogenically related to pathogenesis‐related proteins during both senescence of detached leaves and natural senescence of attached leaves. Both chitinase activity and protein level were found to be induced during senescence, as was the level of two other proteins immunologically related to β‐1,3‐glucanase and P4 pathogenesis‐related proteins of citrus and tomato, respectively. A high correlation between the advancement of senescence and the induction of these proteins was demonstrated. Treatments with CO2 or gibberellic acid, which retard senescence, reduced both chitinase activity and the level of the pathogenesis‐related proteins, whereas enhancement of senescence with ethylene induced them further. The induction of pathogenesis‐related proteins during senescence suggests that these proteins may have a primary role in this process.  相似文献

目的探讨NAS对肝缺血再灌注所诱导的脂质过氧化损伤产生的保护作用。方法采用夹闭肝蒂法30min、再灌注6h制作肝缺血再灌注模型,冰冻切片,HE染色,光学显微镜下观察肝细胞形态结构的变化;比色法检测损伤后血清中谷丙转氨酶(ALT)水平及肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH—Px)的含量。结果夹闭肝蒂30min、再灌注6h后,肝小叶结构紊乱、肝血窦淤血,其间有白细胞浸润、肝细胞出现变性、坏死;血清中ALT水平升高,肝组织中s0D和GSH—Px的含量降低,MDA升高;NAS可减少缺血再灌注后血清ALT的释放,使肝组织中SOD和GSHPx的含量升高,MDA的含量降低;NAS+Luz可逆转NAS的这一作用。结论NAS对肝缺血再灌注小鼠的氧化应激损伤具有保护作用。  相似文献
