Lysosomes function not only as degradatory compartments but also as dynamic intracellular calcium ion stores. The transient receptor potential mucolipin 1 (TRPML1) channel mediates lysosomal Ca2+ release, thereby participating in multiple cellular functions. The pentameric Ragulator complex, which plays a critical role in the activation of mTORC1, is also involved in lysosomal trafficking and is anchored to lysosomes through its LAMTOR1 subunit. Here, we report that the Ragulator restricts lysosomal trafficking in dendrites of hippocampal neurons via LAMTOR1‐mediated tonic inhibition of TRPML1 activity, independently of mTORC1. LAMTOR1 directly interacts with TRPML1 through its N‐terminal domain. Eliminating this inhibition in hippocampal neurons by LAMTOR1 deletion or by disrupting LAMTOR1‐TRPML1 binding increases TRPML1‐mediated Ca2+ release and facilitates dendritic lysosomal trafficking powered by dynein. LAMTOR1 deletion in the hippocampal CA1 region of adult mice results in alterations in synaptic plasticity, and in impaired object‐recognition memory and contextual fear conditioning, due to TRPML1 activation. Mechanistically, changes in synaptic plasticity are associated with increased GluA1 dephosphorylation by calcineurin and lysosomal degradation. Thus, LAMTOR1‐mediated inhibition of TRPML1 is critical for regulating dendritic lysosomal motility, synaptic plasticity, and learning. 相似文献

为了减轻UV-B辐射对植物叶片的伤害,本研究以离体红芸豆叶片为实验材料,通过外源施加NaHSO3的方法探讨了UV-B辐射下NaHSO3对离体红芸豆叶片的保护作用。结果表明:与未处理对照相比较,用0.5mmol·L^-1NaHSO3处理的离体红芸豆叶片表面褐色斑减少、边缘蜷曲及萎蔫程度降低;且能延缓叶片中叶绿素和类胡萝卜素含量的降低;使类黄酮含量升高;叶片中过氧化物酶(POD)和抗坏血酸过氧化物酶(APX)活性升高,过氧化氢(H2O2)含量降低。进一步研究发现NaHSO3处理能明显延缓PSⅡ原初光能转换效率的降低;增强PSⅡ的电子传递能力,减少叶绿体内有害自由基的产生,减缓叶绿体内光合机构遭受破坏的程度。以上结果表明NaHSO3可能通过提高POD和APX的活性、降低自由基产生及保护光合色素等来实现UV-B胁迫下对红芸豆叶片的保护作用。 相似文献

The liver is the largest organ in the body, with many complex, essential functions, such as metabolism, deintoxication, and secretion, often regulated via post-translational modifications, especially phosphorylation. Thus, the detection of phosphoproteins and phosphorylation sites is important to comprehensively explore human liver biological function. The human Chang liver cell line is among the first derived from non-malignant tissue, and its phosphoproteome profile has never been globally analyzed. To develop the complete phosphoproteome and probe the roles of protein phosphorylation in normal human liver, we adopted a shotgun strategy based on strong cation exchange chromatograph, titanium dioxide and LC-MS/MS to isolate and identify phosphorylated proteins. Two types of MS approach, Q-TOF and IT, were used and compared to identify phosphosites from complex protein mixtures of these cells. A total of 1035 phosphorylation sites and 686 phosphorylated peptides were identified from 607 phosphoproteins. A search using the public database of PhosphoSite showed that approximately 344 phosphoproteins and 760 phosphorylation sites appeared to be novel. In addition, N-terminal phosphorylated peptides were a greater fraction of all identified phosphopeptides. With GOfact analysis, we found that most of the identified phosphoproteins are involved in regulating metabolism, consistent with the liver's role as a key metabolic organ. 相似文献

采用田间调查及人工模拟冰冻鉴定的方法,比较测定了4个桂花品种的抗冻性,其抗冻性强弱顺序为金桂(Osmanthusfragransvar.thunbergii)>银桂(O.fragransvar.latifolius)>丹桂(O.fragransvar.aurantiacus)>四季桂(O.fragransvar.semperflorens)。筛选出5株抗冻性最强的单株,它们的致死温度都在-26℃左右。桂花叶片膜脂脂肪酸组成随季节呈周期性变化,随着气温下降亚麻酸(18∶3)含量增加而亚油酸(18∶2)含量降低。不同品种均表现出一致的变化趋势,抗性强的品种这种变化则更为显著。在自然脱锻炼过程中,叶面喷施脱落酸(200ppm)能使亚麻酸含量保持稳定,延缓桂花的脱锻炼。 相似文献

The crystallization of 16 proteins was carried out using 60 wells on board Shenzhou 3 in 2002. Although the mission was only 7 days, careful and concerted planning at all stages made it possible to obtain crystals of improved quality compared to their ground controls for some of the proteins. Significantly improved resolutions were obtained from diffracted crystals of 4 proteins. A complete data set from a space crystal of the PEP carboxykinase yielded significantly higher resolution (1.46A vs. 1.87A), I/sigma (22.4 vs. 15.5), and a lower average temperature factor (29.2A(2) vs. 42.9A(2)) than the best ground-based control crystal. The 3-D structure of the enzyme is well improved with significant ligand density. It has been postulated that the reduced convection and absence of macromolecule sedimentation under microgravity have advantages/benefits for protein crystal growth. 目的:研究载脂蛋白E基因敲除(ApoE-/-)小鼠血脂及心肌酶学的变化,为利用其探讨动脉粥样硬化的病理生理进程提供实验依据。方法:选取28W龄雄性ApoE-/-小鼠6只和同性别、同周龄的野生型C57BL/6J(WT)小鼠10只,应用日立7600全自动生化分析仪分别测定其血清中甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、载脂蛋白B(Apo-B)、超敏C反应蛋白(hs-CRP)、游离脂肪酸(NEFA)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)和缺血修饰白蛋白(ACB)的水平。结果:与WT鼠比,ApoE-/-小鼠血清中血脂指标TG、TC、LDL-C、hs-CRP和NEFA水平显著升高(P<0.01),而HDL-C仅为0.46±0.16mmol/L,远低于WT鼠的水平(1.86±0.26mmol/L)。心肌酶学指标CK、LDH明显高于对照组(P<0.01),缺血修饰白蛋白(IMA()55.61±3.50U/mL)明显低于对照组(72.47±4.26U/mL)(P<0.01)。CK-MB与对照组相比无显著性变化。结论:ApoE-/-小鼠血脂水平... 相似文献
