Our understanding of plant responses to enhanced ultraviolet‐B (UV‐B) radiation has improved over recent decades. However, research on cryptogams is scarce and it remains controversial whether UV‐B radiation causes changes in physiology related to photosynthesis. To investigate the effects of supplementary UV‐B radiation on photosynthesis and chloroplast ultrastructure in Bryum argenteum Hedw., specimens were cultured for 10 days under four UV‐B treatments (2.75, 3.08, 3.25 and 3.41 W m–2), simulating depletion of 0% (control), 6%, 9% and 12% of stratospheric ozone at the latitude of Shapotou, a temperate desert area of northwest China. Analyses showed malondialdehyde content significantly increased, whereas chlorophyll (Chl) fluorescence parameters and Chl contents decreased with increased UV‐B intensity. These results corresponded with changes in thylakoid protein complexes and chloroplast ultrastructure. Overall, enhanced UV‐B radiation leads to significant decreases in photosynthetic function and serious destruction of the chloroplast ultrastructure of B. argenteum. The degree of negative influences increased with the intensity of UV‐B radiation. These results may not only provide a potential mechanism for supplemental UV‐B effects on photosynthesis of moss crust, but also establish a theoretical basis for further studies of adaptation and response mechanisms of desert ecosystems under future ozone depletion. 相似文献

干旱半干旱区灌木、半灌木等木本植物的分布面积持续增加(灌丛化)显著改变了原有植被景观及生态过程,但缺乏对长期封育过程中灌木物种取代草本物种进而影响草地群落组成、结构和功能的研究。为揭示长期封育措施下典型草原灌丛化特征及地上/地下植物群落的响应规律,研究通过对宁夏云雾山典型草原同一封育草地不同时间(2010年、2016年、2021年)植物群落(物种重要值、丰富度、多样性和生物量)和土壤特性(土壤容重、土壤水分及储水量、养分、pH)的定点观测,探讨了长期封育过程中灌木物种矮脚锦鸡儿(Caragana brachypoda)驱动的植被和土壤特性变化。结果表明,(1)灌木矮脚锦鸡儿已经取代草本植物成为封育草地的优势种,群落中灌木生物量、总生物量以及盖度、高度显著增加,但对植物密度的影响较小;(2)灌丛化过程中草地群落Patrick丰富度指数、Simpson优势度指数和Shannon-Wiener多样性指数显著降低,而Pielou均匀度指数显著增加;(3)灌丛化引起土壤深层(60-100 cm)容重显著增大,土层储水量随土层深度的增加呈先减少后增加的趋势;(4)灌丛化使得土壤有机碳含量在浅层(0... 相似文献

从普通栽培型番茄品种‘桃太郎’、野生醋栗番茄和潘那利番茄的幼苗中采用RT-PCR方法,扩增出转化酶抑制子基因cDNA序列的部分片段。经测序表明:不同基因型番茄的转化酶抑制子基因同源性高达97.97%以上。采用半定量RT-PCR方法对‘桃太郎’苗期根、茎、叶和花后功能叶、花期子房以及果实不同发育阶段不同部位的表达进行检测,结果表明:INH在根中表达较弱,茎中有强烈表达,从幼叶到衰老叶表达逐渐减弱;同一时期INH在果肉中表达相对较强,维管束次之,胶质胎座相对较弱;花期子房中INH的表达逐渐增强,花后3d左右达到最大,花后5~8d表达量迅速下降。 相似文献

To provide feasible primer sets for performing a polymerase chain reaction (PCR) experiment, many primer design methods have been proposed. However, the majority of these methods require a relatively long time to obtain an optimal solution since large quantities of template DNA need to be analyzed. Furthermore, the designed primer sets usually do not provide a specific PCR product size. In recent years, evolutionary computation has been applied to PCR primer design and yielded promising results. In this article, a memetic algorithm (MA) is proposed to solve primer design problems associated with providing a specific product size for PCR experiments. The MA is compared with a genetic algorithm (GA) using an accuracy formula to estimate the quality of the primer design and test the running time. Overall, 50 accession nucleotide sequences were sampled for the comparison of the accuracy of the GA and MA for primer design. Five hundred runs of the GA and MA primer design were performed with PCR product lengths of 150–300 bps and 500–800 bps, and two different methods of calculating Tm for each accession nucleotide sequence were tested. A comparison of the accuracy results for the GA and MA primer design showed that the MA primer design yielded better results than the GA primer design. The results further indicate that the proposed method finds optimal or near‐optimal primer sets and effective PCR products in a dry dock experiment. 采用四因素五水平(1/2实施)二次通用旋转组合设计,选取灌水、施氮肥、施磷肥和秸秆覆盖四因素作为试验因素,在辽西半干旱区褐土农田进行了水肥耦合效应长期定位试验。按照二次通用旋转组合设计统计分析方法建立回归模型,分析了水肥耦合对玉米籽粒全氮含量的影响。结果表明:水肥单因子对籽粒全氮的含量有较明显的影响,影响顺序为施氮秸秆覆盖施磷灌水;当灌水量为700m3.hm-2、施氮量为180kg.hm-2、施磷量为120kg.hm-2和秸秆覆盖量为7500kg.hm-2时,它们每个因素对玉米籽粒全氮含量的影响都集中11.7g·kg-1,此时籽粒蛋白质含量为73g·kg-1,产量为12000kg.hm-2,表明该处理组合下,玉米的经济效益和生态效益达到最佳,推荐生产上以此进行施肥。 相似文献

POL32 encodes a non-essential subunit of Polδ and plays a role in Polδ processivity and DNA repair. In order to understand how Pol32 is involved in these processes, we performed extensive genetic analysis and demonstrated that POL32 is required for Polζ-mediated translesion synthesis, but not for Polη-mediated activity. Unlike Polζ, inactivation of Pol32 does not result in decreased spontaneous mutagenesis, nor does it limit genome instability in the absence of the error-free postreplication repair pathway. In contrast, inactivation of Pol32 results in an increased rate of replication slippage and recombination. A genome-wide synthetic lethal screen revealed that in the absence of Pol32, homologous recombination repair and cell cycle checkpoints play crucial roles in maintaining cell survival and growth. These results are consistent with a model in which Pol32 functions as a coupling factor to facilitate a switch from replication to translesion synthesis when Polδ encounters replication-blocking lesions. When Pol32 is absent, the S-phase checkpoint complex Mrc1–Tof1 becomes crucial to stabilize the stalled replication fork and recruit Top3 and Sgs1. Lack of any of the above activities will cause double strand breaks at or near the replication fork that require recombination as well as Rad9 for cell survival. 相似文献

Elucidation of 2-hydroxybiphenyl effect on dibenzothiophene desulfurization by Microbacterium sp. strain ZD-M2 总被引:1,自引:0,他引:1
The effect of 2-hydroxybiphenyl (2-HBP), the end product of dibenzothiophene (DBT) desulfurization via 4S pathway, on cell growth and desulfurization activity was investigated by Microbacterium sp. The experimental results indicate that 2-HBP would inhibit the desulfurization activity. Providing 2-HBP was added in the reaction media, the DBT degradation rate decreased along with the increase of 2-HBP addition. By contrast, cell growth would be promoted in the addition of 2-HBP at a low concentration (<0.1mM). At high concentration of 2-HBP, the inhibition on the cell growth occurred. Meanwhile, the inhibitory effect of 2-HBP on DBT desulfurization activity was tested both in the oil/aqueous two-phase system and the aqueous system. A mathematical model was developed to explain the product formation kinetics with DBT as the sole sulfur source. The predicted results were close to the experimental data, it elucidated that along with the 2-HBP accumulation, the inhibitory effect of 2-HBP on DBT desulfurization and cell growth was enhanced. 相似文献
