A C3 symmetric (R)‐phenylglycinol N‐1,3,5‐benzenetricarboxylic acid‐derived chiral stationary phase (CSP) and three C2 symmetric (R)‐phenylglycinol CSPs were newly synthesized using o‐, m‐, and p‐phthaloyl dichlorides. © 2016 Wiley Periodicals, Inc. These CSPs were used to compare the resolution of 25 chiral samples using a previously reported 3,5‐dinitrobenzoyl (R)‐phenylglycinol‐derived CSP. An efficient DNA extraction protocol and polymerase chain reaction (PCR) assay for detecting Leptosphaeria maculans from infected seed lots of oilseed rape were developed. L. maculans, the causal agent of blackleg, a damaging disease in oilseeds rape/canola worldwide, was listed as a quarantine disease by China in 2009. China imports several millions of tons of oilseeds every year. So there is a high risk that this pathogen will be introduced to China via contaminated seeds. Seed contamination is one of the most significant factors in the global spread of phytopathogens. Detection of L. maculans in infected seed lots by PCR assay is difficult due to the low level of pathogen mycelium/spores on seeds and PCR inhibitors associated with the seeds of oilseed rape. In our study, these two major obstacles were overcome by the development of a two‐step extraction protocol combined with a nested PCR. This extraction protocol (kit extraction after CTAB method) can efficiently extract high‐quality DNA for PCR. Amplification results showed that the detection threshold for conventional PCR and nested PCR was, respectively, 1 ng and 10 fg of DNA per μl in mycelia samples. On contaminated seed lots of oilseed rape, the detection threshold of conventional and nested PCR was 709 fg/μl and 709 ag/μl of DNA, respectively. The DNA extraction protocol and PCR assay developed in this study can be used for rapid and reliable detection of L. maculans from infected seeds of oilseed rape . 相似文献

Black spot leads to great marigold losses worldwide. The disease is characterized by black spots on leaves and stems in its early stages, and the whole plant has black rot at the advanced stage. In this report, 6 of 217 Alternaria strains isolated from lesions of marigold plants in Beijing were randomly selected. The morphological characteristics and a pathogenic tree based on two protein‐coding genes (gpd and alt a 1) indicated that Alternaria tagetica is the causal agent of marigold black spot in Beijing. All six Alternaria strains could successfully re‐infect marigold, but they could not infect carrot or zinnia by either spore spray in a greenhouse or planting experiments in the epidemic area. This is the first report of the A. tagetica pathogen being isolated from marigold in Beijing. 相似文献

Excessive and long-term alcohol consumption leads to liver disease and low immunity. Extensive evidence suggests that C-phycocyanin (C-PC), a chromophore phycocyanobilin derived from Arthrospira (Spirulina) platensis, exerts protective effects against chemical-induced organ damage and improves immunity. In this study, we investigated whether C-PC could protect against ethanol-induced subacute liver injury and improve immunity. KM mice with ethanol-induced liver injury were established, and animals were divided into three groups that were treated with high, medium, and low doses of C-PC. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), total cholesterol (CHOL), low-density lipoprotein (LDL), total bilirubin (TBIL), liver homogenate malondialdehyde (MDA), and superoxide dismutase (SOD) levels were measured. In addition, the number of thymus T cell subsets was assessed, and liver sections were examined pathologically. C-PC exhibited obvious inhibitory effects on serum ALT, AST, TG, CHOL, LDL, and MDA levels and increased SOD content significantly in the liver. C-PC also increased serum CD3+ and CD4+ cell activation and T cell proliferation significantly compared with the model group. The structure of the hepatic lobules was clear, the liver sinus returned to normal, and the liver cell cords were arranged in neat rows. Therefore, C-PC could protect against ethanol-induced subacute liver injury significantly. 相似文献

为研究外阴阴道假丝酵母菌病(Vulvovaginal Candidiasis,VVC)患者合并支原体和衣原体感染情况,对86例复发性VVC(recurrent Vulvovaginal Candidiasis,RVVC)患者、219例单纯性VVC患者以及健康妇女200例,分别进行解脲脲原体(Ureaplasma urealyticum,Uu)、人型支原体(mycoplasma hominis,Mh)和沙眼衣原体(Chlamydia trachomatis,CT)检测;所有VVC病例均进行真菌培养。RVVC组和单纯VVC组与对照组之间Uu、Mh及Uu混合Mh阳性率比较,差异均有统计学意义(P0.05)。而三组之间CT和CT混合支原体阳性率比较,均无显著性差异(P0.05)。真菌培养阳性组与阴性组之间,Uu阳性率比较有显著性差异(P0.05),而CT阳性率比较无显著性差异(P0.05)。结果表明,VVC患者合并Uu感染较正常人群明显增高,假丝酵母菌与Uu混合感染可能导致VVC的发生和复发。 相似文献

识别扩散限制和环境筛选在群落物种共存中的相对作用,是土壤动物群落物种共存机制研究的重要内容,然而少有针对地表和地下土壤动物群落的探讨。在三江平原农田生态系统,设置一个50 m×50 m的空间尺度,探讨环境筛选和扩散限制对地表和地下土壤螨群落物种共存的调控作用。基于Moran特征向量图(MEMs)和变差分解的方法来区分环境筛选和扩散限制的调控作用;采用偏Mantel检验进一步分析环境距离和空间距离的相对贡献;使用RDA分析环境因子对螨群落物种组成的解释能力。变差分解结果表明,空间变量对地表、地下和地表-地下土壤螨群落具有较大的显著方差解释量,而环境变量和空间环境结构的解释量相对较小且不显著;偏Mantel检验没有发现环境距离或空间距离的显著贡献;RDA分析表明土壤p H值、大豆株高和土壤含水量对土壤螨群落具有显著的解释能力,说明环境变量对螨群落物种组成的重要作用。研究表明,在三江平原农田生态系统,地表和地下土壤螨群落物种共存主要受到扩散限制的调控作用,同时环境筛选的调控作用也不容忽视。 相似文献

We propose a novel parallel computing framework for a nonlinear finite element method (FEM)-based cell model and apply it to simulate avascular tumor growth. We derive computation formulas to simplify the simulation and design the basic algorithms. With the increment of the proliferation generations of tumor cells, the FEM elements may become larger and more distorted. Then, we describe a remesh and refinement processing of the distorted or over large finite elements and the parallel implementation based on Message Passing Interface to improve the accuracy and efficiency of the simulation. We demonstrate the feasibility and effectiveness of the FEM model and the parallelization methods in simulations of early tumor growth. 相似文献

利用屋顶绿化隔热,对降低夏季空调耗能尤为重要。目前中国还缺少针对其日照环境差异下夏季昼夜热效应的全面考察研究。借助建筑日照分析将试验屋顶划分为4个日照区间,利用HOBO小气象站测取环境温度数据、热电偶昼夜连续测取不同日照区间多点温度数据,综合分析上海佛甲草模块式屋顶绿化的昼夜隔热效果。研究结果表明:试验屋顶绿化在夏季白天的隔热效果明显,最大屋面降温值可达28℃以上,日照越强隔热效果越明显;夜间的散热效果不及裸屋面;且存在最高温度时滞效应。其对建筑节能的昼夜综合效应,需要结合建筑屋顶构造自身的保温隔热性能、屋面日照条件以及建筑的昼夜使用节律做进一步的深入研究,以期达到屋顶绿化经济价值与节能效益的最大化。 相似文献

As essential B vitamin for humans, folates accumulation in edible parts of crops, such as maize kernels, is of great importance for human health. But its breeding is always limited by the prohibitive cost of folate profiling. The molecular breeding is a more executable and efficient way for folate fortification, but is limited by the molecular knowledge of folate regulation. Here we report the genetic mapping of folate quantitative trait loci (QTLs) using a segregated population crossed by two maize lines, one high in folate (GEMS31) and the other low in folate (DAN3130). Two folate QTLs on chromosome 5 were obtained by the combination of F2 whole-exome sequencing and F3 kernel-folate profiling. These two QTLs had been confirmed by bulk segregant analysis using F6 pooled DNA and F7 kernel-folate profiling, and were overlapped with QTLs identified by another segregated population. These two QTLs contributed 41.6% of phenotypic variation of 5-formyltetrahydrofolate, the most abundant storage form among folate derivatives in dry maize grains, in the GEMS31×DAN3130 population. Their fine mapping and functional analysis will reveal details of folate metabolism, and provide a basis for marker-assisted breeding aimed at the enrichment of folates in maize kernels. 相似文献
