DNA损伤生物学反应中ATM对p21~(WAF1/CIP1)蛋白的直接磷酸化   总被引:3,自引:0,他引:3  
毛细血管扩张性共济失调症突变蛋白 (mutatedinataxiatelangiectasia ,ATM)是直接感受DNA双链断裂损伤并起始诸多DNA损伤信号反应通路的主开关分子 .已有研究发现 ,DNA损伤生物学反应中 ,ATM可通过磷酸化活化p5 3,继而转录活化细胞周期检查点蛋白p2 1WAF1 CIP1的表达 ,而对于ATM是否直接参与p2 1WAF1 CIP1的早期活化迄今尚无实验证明 .通过免疫共沉淀反应 ,检测到细胞电离辐射 (ionizingradiation ,IR)反应早期ATM与p2 1WAF1 CIP1蛋白存在相互作用 .将p2 1WAF1 CIP1蛋白编码基因全长克隆入原核表达载体pGEX4T 2 ,经诱导表达及亲和层析纯化获取GST p2 1融合蛋白作为磷酸化底物 .体外磷酸化实验检测证明 ,IR活化的ATM具磷酸化p2 1WAF1 CIP1蛋白的功能 ,并且此磷酸化功能可被PI3K家族特异性抑制剂Wortmannin所抑制 .结果揭示了IR后ATM可通过直接磷酸化p2 1WAF1 CIP1蛋白 ,在IR致DNA损伤生物学反应早期调控p2 1WAF1 CIP1蛋白的快速活化过程  相似文献

目的:建立从全血中提取基因组DNA的方法,并评价提取DNA的产量和质量.方法:分别采用传统酚-氯仿抽提法和试剂盒法制备基因组DNA,比较这两种方法提取DNA的产量、纯度、稳定性以及方法本身的优缺点、费用等.结果:试剂盒法简单快速,但其制备的DNA产率、纯度、稳定性低于酚-氯仿抽提法.结论:采用酚-氯仿抽提法可以提取较高质量的基因组DNA,适用于下游的分子生物学实验.  相似文献

Mangroves are essential for maintaining local biodiversity and human well-being, and mangrove structure and functioning depend on the macrobenthos. Although exotic cordgrass, Spartina alterniflora, is an increasing threat to the mangrove wetlands (including the associated unvegetated shoals) of China, its effects on the macrobenthic fauna in such wetlands is poorly understood. The macrobenthic faunal communities were compared in (1) an Avicennia marina monoculture vs. an S. alterniflora-invaded A. marina stand (a mixture of A. marina and S. alterniflora) and in (2) an unvegetated shoal vs. an S. alterniflora-invaded shoal that had rapidly become an S. alterniflora monoculture in Zhanjiang, China. S. alterniflora invasion significantly increased plant density regardless of invaded habitat but significantly increased the contents of total carbon, organic matter, and total sulfur in the sediment only in the unvegetated shoal. The presence of S. alterniflora had little influence on indices of the macrobenthic faunal community in the A. marina monoculture, but significantly decreased the density and biomass of macrobenthic faunal community in the unvegetated shoal. These results indicate that the effects of S. alterniflora on the macrobenthic faunal community depend on which type of mangrove habitat is invaded. The composition of the macrobenthic faunal community was more similar between the invaded and non-invaded A. marina stand than between the invaded and non-invaded unvegetated shoal. Overall, the differences in the macrobenthic faunal community between invaded and non-invaded habitats were associated with increases in the sediment organic matter content and plant density.  相似文献

Ninety-one lakes distributed along the Tatra Mountains (most of lakes > 1 ha and 65% of lakes > 0.01 ha) were sampled and analysed for ionic and nutrient composition in September 2004 (15 years after reduction in acid deposition). Eighty-one lakes were in alpine zone and ten lakes in Norway spruce forest. The results were compared to similar lake surveys from 1994 (the beginning of water recovery from acidification) and 1984 (maximum acidification). Atmospheric deposition of SO 4 2? and inorganic N decreased 57% and 35%, respectively, in this region from the late 1980s to 2000. Lake water concentrations of SO 4 2? and NO 3 ? have decreased both by ~50% on average (to 23 and 19 μmol L?1, respectively, in 2004) since 1984. While the decrease in SO 4 2? concentrations was stable throughout 1984–2004, most of the NO 3 ? decrease occurred from 1994 to 2004. The declines in SO 4 2? and NO 3 ? concentrations depended on catchment coverage with vegetation, being most rapid for SO 4 2? in forest lakes and for NO 3 ? in rocky lakes. Concentrations of the sum of base cations (dominated by Ca2+) significantly decreased between 1984 and 2004, with the highest change in rocky lakes. Most of this decline occurred between 1994 and 2004. Acid neutralising capacity (ANC) did not change in the 1984–1994 period, but increased on average by 29 μmol L?1 between 1994 and 2004, with the highest change in rocky lakes. Over the last decade, the proportion of lakes with ANC > 150 μmol L?1 increased from 15% to 21% and that of ANC < 20 μmol L?1 decreased from 37% to 20%. The highest decline in H+ and Al concentrations occurred in the most acid lakes. On a regional basis, no significant change was observed for total phosphorus, total organic nitrogen, and dissolved organic carbon (DOC) in the 1994–2004 period. However, these parameters increased in forest lakes, which exhibited an increasing trend in DOC concentrations, inversely related (P < 0.001) to their decreasing ionic strength (30% on average in 1994–2004).  相似文献

目的:探讨红花注射液对大鼠在慢性低O2高CO2下肺动脉高压的抑制作用。方法:将SD大鼠分为对照组,慢性低O2高CO2组,慢性低O2高CO2+红花注射液组。用电镜、放免等方法,观察各组大鼠肺动脉平均压、颈动脉平均压、肺细小动脉显微结构、血浆和肺匀浆TXB2及6-keto-PGF1a含量的变化。结果:①慢性低O2高CO2组mPAP比对照组显著增高,红花注射液组的mPAP比慢性低O2高CO2组显著降低,3组间mCAP比较差异无显著性。②慢性低O2高CO2组与对照组相比血浆和肺匀浆TXB2浓度、TXB2/6-keto-PGF1a比值显著增高,6-keto-PGF1a浓度显著下降;红花注射液组与慢性低P2高CO2相比血浆和肺匀浆TXB2浓度、TXB2/6-keto-PGF1a显著下降,6-keto-PGF1a显著升高。③光镜下慢性低O2高CO2组与对照组相比,肺细小动脉管壁面积/管总面积(WA/TA)和肺细小动脉中膜厚度(PAMT)均显著增高。红花注射液组WA/TA和PAMT显著降低。④电镜下慢性低O2高CO2组大鼠肺细小动脉内皮细胞吞饮小泡增多,血管壁增厚,中膜平滑肌细胞增生,纤维细胞增多,肺泡Ⅱ型上皮细胞微绒毛脱落;红花注射液组肺细小动脉中膜平滑肌细胞增生减轻,纤维细胞少,胶原纤维减少,肺泡Ⅱ型上皮细胞微绒毛丰富、结构清。结论:红花注射液有减轻慢性低O2高CO2性肺动脉高压和肺血管结构重建的作用,可能与抑制TXA2的合成,保护血管内皮细胞,使TXA2/PGI2比值降低有关.  相似文献

选取广西弄拉典型峰丛洼地生态系统中不同地貌类型的青冈栎 (Cyclobalanopsisglauca)叶片进行形态解剖特征比较分析 ,结果表明 :( 1 )峰丛洼地生态系统中不同地貌青冈栎叶片形态解剖特征差异显著。山顶青冈栎叶片部分表皮结构 (角质膜、表皮毛、表皮细胞、气孔 )趋向旱化。山顶青冈栎下表皮毛比山腰青冈栎的浓密且长。两者叶片的厚度和宽度、上表皮细胞个数、气孔指数、下表皮厚度、栅栏组织厚度、海绵组织厚度存在着显著性差异 ;位于山顶的青冈栎受到水分的胁迫 ,在形态解剖上呈现出中生偏旱的结构。 ( 2 )青冈栎的各种形态解剖指标说明在岩溶区青冈栎的抗旱性主要是通过抵御干旱来适应水分的胁迫 ,并主要取决于减少蒸腾失水和维持水分吸收能力两个方面  相似文献

目的:通过分析六种新疆陆地棉棉籽脂肪酸成分与含量的变化,为更好的开发,利用丰富的棉籽资源提供实验数据.方法:采用索氏提取法提取了6种新疆陆地棉棉籽油,其棉籽脂肪酸经甲酯化后用气相色谱-质谱联用仪进行分析.结果:不同品种棉籽油所含脂肪酸成分基本相同,但各脂肪酸成分的相对含量差异较大,其中亚油酸含量变化范围为50.30%~57.25%,棕榈酸、油酸含量的变化范围分别为22.24%~30.98%和12.93%~16.84%.不同棉花品种棉籽的多不饱和脂肪酸(PUFA)与饱和脂肪酸(SFA)相对含量比值也不同,变化范围为1.66~2.59.结论:不同品种棉籽,其营养价值及经济价值存在较大差异,其中,新陆早27号棉籽具有相对更好的开发利用价值.  相似文献

目的:探讨超声诊断胰腺脂肪浸润与血脂、脂肪肝的关系。方法:回顾性分析63例胰腺脂肪浸润者与148例同期检查无胰腺脂肪浸润者的病历资料,研究胰腺脂肪浸润的超声特点以及与脂肪肝、血脂浓度、年龄、血压等的关系。结果:胰腺脂肪浸润超声显示为胰腺饱满,边缘模糊,回声增强且明显强于肝脏,与正常胰腺超声影像容易鉴别。胰腺脂肪浸润组血浆甘油三酯浓度2.17±1.22mmol/L,比非浸润组的1.35±1.06mmol/L明显高(P〈0.05),胰腺脂肪浸润组患者100%(63/63)合并脂肪肝明显高于非浸润组的20.95%(31/148)(P〈0.05)。二组在胆固醇浓度、高血压发病率及年龄方面无显著差异(P〉0.05)。结论:胰腺脂肪浸润与脂肪肝、血浆甘油三酯浓度有着一定的相关性,超声诊断胰腺脂肪浸润对于提示其他相关疾病与异常的存在具有重要临床参考价值。  相似文献

Accurate assessment of human epidermal growth factor receptor (HER) 2 is essential for efficient selection of patients who may benefit from therapies targeting this surface receptor (e.g., trastuzumab). Intratumoral heterogeneity of HER2 expression may potentially contribute to inaccurate assessment of HER2 status. To clarify intratumoral heterogeneity of HER2 expression and its potential clinical impact on assessment of HER2 status, we analyzed 148 endoscopic biopsy specimens and 117 excisional tumor specimens collected from 148 patients with primary gastric cancer. Specifically, we assessed HER2 protein overexpression and gene amplification using, respectively, immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH). There were 28 IHC-positive cases and 25 FISH-positive cases among these 148 patients. Heterogeneous HER2 protein expression was demonstrated in 23 of 29 (79.3%) IHC-positive cases, while gene expression heterogeneity was found in 11 of 25 (44.0%) FISH-positive cases. Intratumoral heterogeneity was the main reason of discordant results between IHC and FISH or between endoscopic biopsy and excisional tumor specimens. The clinical significance and impact of intratumoral HER2 expression heterogeneity on treatment outcome in gastric cancer require further studies.  相似文献
