Approximately 40% of patients with type 2 diabetes present with concurrent hypertension at the time of diabetes diagnosis. Increases in peripheral vascular resistance and correspondingly enhanced vasoconstrictor capacity could have profound implications for the development of hypertension and the progression of insulin resistance to overt diabetes. The purpose of this study was to determine whether skeletal muscle arteriolar vasoconstrictor dysfunction precedes or occurs concurrently with the onset of diabetes and hypertension. Male Zucker diabetic fatty (ZDF) rats were studied at 7, 13, and 20 wk of age to represent prediabetic and short-term and long-term diabetic states, respectively. Conscious mean arterial pressure (MAP), fasted plasma insulin and glucose, vasoconstrictor responses, and passive mechanical properties of isolated skeletal muscle arterioles were measured in prediabetic, diabetic, and age-matched control rats. Elevated MAP was manifest in short-term diabetes (control 117 +/- 1, diabetic 135 +/- 3 mmHg) and persisted with long-term diabetes (control 113 +/- 2, diabetic 135 +/- 3 mmHg). This higher MAP was preceded by augmented arteriolar vasoconstrictor responses to norepinephrine and endothelin-1 and followed by diminished beta-adrenergic vasodilation and enhanced myogenic constriction in long-term diabetes. Furthermore, we demonstrate that diminished nitric oxide (NO) signaling underlies the increases in vasoconstrictor responsiveness in arterioles from prediabetic and diabetic rats. Arteriolar stiffness was not different between control and prediabetic or diabetic rats at any time point studied. Collectively, these results indicate that increases in vasoconstrictor responsiveness resulting from diminished NO signaling in skeletal muscle arterioles precede the development of diabetes and hypertension in ZDF rats.  相似文献

First-order lateral roots originating in the upper part of the taproot of a woody species, usually termed surface roots, grow close beneath the soil surface, even on irregular or sloping ground. In slope condition, in fact, the surface roots can assume upward as well as downward growth. Existing knowledge on the controls over root direction does not fully explain these field observations.

Two different soil types and sloping conditions were selected in field condition to explore the behaviour of the surface roots in the woody species Spartium junceum L. The root system 3D architecture was measured with a 3D digitizer and the angle of growth (0° = vertically downwards) and the radial direction (0° = horizontally downslope or northwards) of all root segments measured.

Surface roots were more numerous in clay soil than in loam soil, independently from the slope inclination. They had initial angles larger than 90°, i.e. they grew upwards only in clay soil. The subsequent angles of growth maintained this value only in steep-slope condition, showing a clear soil type x slope inclination interaction. The initial angle of all first-order lateral roots decreased linearly with depth of origin on the taproot always in relation to the soil type, with this relationship being stronger in clay soil.

These findings showed that the liminal angle (the preferred angle of growth) of surface roots was mainly affected by the soil type rather than the soil surface inclination. Thus, upward growth must stand in the plasticity of the plagiotropic response of these secondary laterals rather than in a strong internal control.  相似文献

Thelazia gulosa, Thelazia rhodesi and Thelazia skrjabini are nematodes transmitted by some species of Musca (Diptera: Muscidae) which cause ocular infestations in bovines. Differences in the rDNA of these species were determined by a PCR using different sets of relatively conserved oligonucleotide primers. PCR on the first internal transcribed spacer (ITS-1) revealed differences in size in Thelazia species (437 bp for T. gulosa, 370 bp for T. rhodesi and 506 bp for T. skrjabini) while the DNA control of Musca spp. was not amplified. The ITS-1 amplicons of the three species were sequenced and then analysed. The GC contents ranged from 26 to 36% and the level of differences in the nucleotide sequences of ITS-1 was lower between T. skrjabini and T. gulosa (39%) than the latter and T. rhodesi (49–56%). Restriction fragment length polymorphism (RFLP) of ITS-1 amplicons was also carried out and the restriction profiles compared. Clear genetic differences among the three Thelazia examined were demonstrated by using the enzymes HpaII, CpoI and SspI. This PCR–RFLP for the delineation of T. gulosa, T. rhodesi and T. skrjabini offers prospects as a molecular epidemiological tool to study parasite transmission patterns and prevalence.  相似文献

In organic solar cells continuous donor and acceptor networks are considered necessary for charge extraction, whereas discontinuous neat phases and molecularly mixed donor–acceptor phases are generally regarded as detrimental. However, the impact of different levels of domain continuity, purity, and donor–acceptor mixing on charge transport remains only semiquantitatively described. Here, cosublimed donor–acceptor mixtures, where the distance between the donor sites is varied in a controlled manner from homogeneously diluted donor sites to a continuous donor network are studied. Using transient measurements, spanning from sub‐picoseconds to microseconds photogenerated charge motion is measured in complete photovoltaic devices, to show that even highly diluted donor sites (5.7%–10% molar) in a buckminsterfullerene matrix enable hole transport. Hopping between isolated donor sites can occur by long‐range hole tunneling through several buckminsterfullerene molecules, over distances of up to ≈4 nm. Hence, these results question the relevance of “pristine” phases and whether a continuous interpenetrating donor–acceptor network is the ideal morphology for charge transport.  相似文献

High photon energy losses limit the open‐circuit voltage (VOC) and power conversion efficiency of organic solar cells (OSCs). In this work, an optimization route is presented which increases the VOC by reducing the interfacial area between donor (D) and acceptor (A). This optimization route concerns a cascade device architecture in which the introduction of discontinuous interlayers between alpha‐sexithiophene (α‐6T) (D) and chloroboron subnaphthalocyanine (SubNc) (A) increases the VOC of an α‐6T/SubNc/SubPc fullerene‐free cascade OSC from 0.98 V to 1.16 V. This increase of 0.18 V is attributed solely to the suppression of nonradiative recombination at the D–A interface. By accurately measuring the optical gap (Eopt) and the energy of the charge‐transfer state (ECT) of the studied OSC, a detailed analysis of the overall voltage losses is performed. EoptqVOC losses of 0.58 eV, which are among the lowest observed for OSCs, are obtained. Most importantly, for the VOC‐optimized devices, the low‐energy (700 nm) external quantum efficiency (EQE) peak remains high at 79%, despite a minimal driving force for charge separation of less than 10 meV. This work shows that low‐voltage losses can be combined with a high EQE in organic photovoltaic devices.  相似文献

The involvement of the ascorbate (AsA) system in the response of pumpkin (Cucurbita pepo L.) roots to aluminium stress was studied. The treatment of 5-day-old pumpkin seedlings with 50 microM aluminium sulphate resulted in approximately 60% inhibition of root growth within 48-60 h of treatment, while aluminium accumulated in the roots reaching a maximum within 48h. During the same period, the hydrogen peroxide content of the roots was strongly enhanced. The increased level of hydrogen peroxide was matched by both increased ascorbate peroxidase (APX) (EC 1.11.1.11) activity and ascorbate free radical reductase (AFRR) (EC 1.1.5.4) activity, while dehydroascorbate reductase (DHAR) (EC 1.8.5.1) and glutathione reductase (GR) (EC 1.6.4.2) did not change. The levels of AsA in the roots were also increased by the Al treatment. It was concluded that an oxidative burst is probably involved in the toxicity of Al in pumpkin roots and that plants react to the enhanced production of reactive oxygen species by expressing higher levels of scavenging systems such as the AsA-APX system.  相似文献
