Plant density varies naturally, from isolated plants to clumped individuals, and this can influence pollinator foraging behaviour and plant reproductive success. In addition, the effect of conspecific density on reproduction may depend on the pollination system, and deceptive species differ from rewarding ones in this regard, a high density being often associated with low fruit set in deceptive plants. In our study, we aimed to determine how local conspecific density and floral display size (i.e. number of flowers per plant) affect fruit set in a deceptive orchid (Orchis militaris) through changes in pollinator visitation. We measured fruit set in a natural population and recorded pollinator abundance and foraging behaviour within plots of different O. militaris densities. Detailed data were recorded for the most abundant potential pollinators of O. militaris, i.e. solitary bees. Floral display size was negatively correlated to fruit set in medium‐density plots, but uncorrelated in low‐ and high‐density plots. Plot density had no effect on solitary bee abundance and visitation, which may be due to low pollinator abundance within the study site. The proportion of visited flowers per inflorescence was negatively influenced by floral display size, which is in line with previous studies. In addition, solitary bees spent decreasing time in successive flowers within an inflorescence, and the time spent per flower was negatively affected by ambient temperature. Our results suggest that pollinator behaviour during visitation is poorly linked to pollen deposition and reproductive success in O. militaris.  相似文献

The histone acetyl transferase Tip60 (HTATIP) shares many properties with the tumor suppressor p53 (TP53). Both proteins are involved in the cellular response to DNA damage, are subjected to proteasomal digestion following Mdm2-mediated ubiquitination, and accumulate after UV irradiation. We found here that knock-down of Tip60 affects the p53-dependent response following actinomycin D treatment, most likely because it inhibits p21 (CDKN1A) accumulation. Moreover, Tip60 is required for p53 to activate the endogenous p21 promoter, suggesting that it functions as a p53 co-activator. However, we also found that knock-down of Tip60 increases the turnover rate of p53 under normal growth conditions. Tip60 interferes with Mdm2-mediated degradation of p53, probably because it affects its subcellular localization. Taken together, our results suggest that Tip60 plays a double role in the p53 pathway: under normal growth conditions, Tip60 contributes to maintain a basal pool of p53 by interfering with its degradation; following DNA damage, Tip60 functions as p53 co-activator. That these two distinct roles are linked during the p53-dependent response is an attractive hypothesis.  相似文献

Recycling is a limiting step for receptor‐mediated endocytosis. We first report three in vitro or in vivo evidences that class III PI3K/VPS34 is the key PI3K isoform regulating apical recycling. A substractive approach, comparing in Opossum Kidney (OK) cells a pan‐class I/II/III PI3K inhibitor (LY294002) with a class I/II PI3K inhibitor (ZSTK474), suggested that class III PI3K/VPS34 inhibition induced selective apical endosome swelling and sequestration of the endocytic receptor, megalin/LRP‐2, causing surface down‐regulation. GFP‐(FYVE)x2 overexpression to sequester PI(3)P caused undistinguishable apical endosome swelling. In mouse kidney proximal tubular cells, conditional Vps34 inactivation also led to vacuolation and intracellular megalin redistribution. We next report that removal of LY294002 from LY294002‐treated OK cells induced a spectacular burst of recycling tubules and restoration of megalin surface pool. Acute triggering of recycling tubules revealed recruitment of dynamin‐GFP and dependence of dynamin‐GTPase, guidance directionality by microtubules, and suggested that a microfilamentous net constrained endosomal swelling. We conclude that (i) besides its role in endosome fusion, PI3K‐III is essential for endosome fission/recycling; and (ii) besides its role in endocytic entry, dynamin also supports tubulation of recycling endosomes. The unleashing of recycling upon acute reversal of PI3K inhibition may help study its dynamics and associated machineries.  相似文献

The elastic properties of membrane bilayers are key parameters that control its deformation and can be affected by pharmacological agents. Our previous atomic force microscopy studies revealed that the macrolide antibiotic, azithromycin, leads to erosion of DPPC domains in a fluid DOPC matrix [A. Berquand, M. P. Mingeot-Leclercq, Y. F. Dufrene, Real-time imaging of drug-membrane interactions by atomic force microscopy, Biochim. Biophys. Acta 1664 (2004) 198-205.]. Since this observation could be due to an effect on DOPC cohesion, we investigated the effect of azithromycin on elastic properties of DOPC giant unilamellar vesicles (GUVs). Microcinematographic and morphometric analyses revealed that azithromycin addition enhanced lipid membranes fluctuations, leading to eventual disruption of the largest GUVs. These effects were related to change of elastic moduli of DOPC, quantified by the micropipette aspiration technique. Azithromycin decreased both the bending modulus (k(c), from 23.1+/-3.5 to 10.6+/-4.5 k(B)T) and the apparent area compressibility modulus (K(app), from 176+/-35 to 113+/-25 mN/m). These data suggested that insertion of azithromycin into the DOPC bilayer reduced the requirement level of both the energy for thermal fluctuations and the stress to stretch the bilayer. Computer modeling of azithromycin interaction with DOPC bilayer, based on minimal energy, independently predicted that azithromycin (i) inserts at the interface of phospholipid bilayers, (ii) decreases the energy of interaction between DOPC molecules, and (iii) increases the mean surface occupied by each phospholipid molecule. We conclude that azithromycin inserts into the DOPC lipid bilayer, so as to decrease its cohesion and to facilitate the merging of DPPC into the DOPC fluid matrix, as observed by atomic force microscopy. These investigations, based on three complementary approaches, provide the first biophysical evidence for the ability of an amphiphilic antibiotic to alter lipid elastic moduli. This may be an important determinant for drug: lipid interactions and cellular pharmacology.  相似文献

Multivariate analysis tools are exploited on a data set composed of quantitative characteristics collected on 35 populations of plants of theDactylorhiza maculata (L.)Soó group from Western-Europe. These samples lead to four well-defined clusters; this, together with qualitative, cytological and ecological arguments, allows for the recognition of four specific entities:D. maculata s.str.,D. fuchsii (Druce)Soó,D. saccifera (Brongn.)Soó andD. caramulensis (Vermeulen)Tyteca. It is concluded that the floral characters play an essential role in the taxonomical distinction. It also appears that the set of characters measured, as well as the methods exploited, are especially well-suited and valuable tools for the morphological study of the genusDactylorhiza.  相似文献

Arthropod generalist predators can be effective natural control agents of pests and weeds in agroecosystems. Their activity and contribution to biocontrol may increase in response to more complex agricultural habitats. In this study, we investigated the effects of winter wheat-pea mixed intercropping on the biocontrol potential of generalist predators compared with the respective mono-crops. We evaluated not only the effects during the intercropping season but also the pre-crop values of the mixture for the subsequent barley crop. Furthermore, we evaluated the influence of different long-term soil organic carbon and fertility management regimes on activity and biocontrol potential of predators. Field work was conducted over two seasons in a field experiment located in Gembloux, Belgium. A set of proxies for ecosystem functions were measured using the Rapid Ecosystem Function Assessment approach. We measured attack and predation rates of sentinel prey and weed seeds artificially placed in the field. Furthermore, we assessed activity density of the main groups of generalist predators during the exposure of the baits. Our results showed that crop type affected activity and biocontrol potential of predators. Predation rates were much lower in wheat than pea and wheat-pea. The mixture wheat-pea had a positive effect on predator activity density compared to wheat mono-crop, while pea supported an intermediate activity of epigeal predators. In the second season of the field work, we found the highest biocontrol potential by predators in barley plots cultivated after pea. Finally, our results failed to find any differences in biocontrol potential of predators between long-term soil organic carbon and fertilisation management strategies. These results suggest that crop type has a major relevance in influencing the activity of generalist predators, and the mixed intercropping wheat-pea may represent a valid strategy to enhance biological pest control in comparison to wheat cultivated as mono-crop. Furthermore, we show that the cultivation of pea as mono-crop may have an important pre-crop value within the rotation increasing the provision of ecosystem services such as biocontrol.  相似文献
