The substantial loss in the stratospheric ozone layer and consequent increase in solar ultraviolet radiation on the earth’s surface have augmented the interest in searching for natural photoprotective compounds in organisms of marine as well as freshwater ecosystems. A number of photoprotective compounds such as mycosporine-like amino acids (MAAs), scytonemin, carotenoids and several other UV-absorbing substances of unknown chemical structure have been identified from different organisms. MAAs form the most common class of UV-absorbing compounds known to occur widely in various marine organisms; however, several compounds having UV-screening properties still need to be identified. The synthesis of scytonemin, a predominant UV-A-photoprotective pigment, is exclusively reported in cyanobacteria. Carotenoids are important components of the photosynthetic apparatus that serve both light-harvesting and photoprotective functions, either by direct quenching of the singlet oxygen or other toxic reactive oxygen species or by dissipating the excess energy in the photosynthetic apparatus. The production of photoprotective compounds is affected by several environmental factors such as different wavelengths of UVR, desiccation, nutrients, salt concentration, light as well as dark period, and still there is controversy about the biosynthesis of various photoprotective compounds. Recent studies have focused on marine organisms as a source of natural bioactive molecules having a photoprotective role, their biosynthesis and commercial application. However, there is a need for extensive work to explore the photoprotective role of various UV-absorbing compounds from marine habitats so that a range of biotechnological and pharmaceutical applications can be found.  相似文献

Abstract The effects of solar irradiation on chlorophyll a fluorescence and photosynthetic oxygen production of three Cryptomonas species, Euglena gracilis and Scenedesmus cf. quadricauda were investigated in comparative field studies in Erlangen (280 m above sea level) and at Zugspitze (2957 m above sea level). The experiments showed that the decrease of fluorescence and the inhibition of photosynthetic oxygen production occurred after shorter times of exposure to solar radiation at Zugspitze compared to Erlangen in all tested organisms. Cryptomonas maculata was more sensitive towards radiation than the other organisms: fluorescence decreased earlier, while Scenedesmus cf. quadricauda seemed to be much less sensitive since fluorescence and photosynthetic oxygen production decreased later and to a smaller extent compared to the other organisms. The results of the present study indicate that increased solar radiation (with an increased level of UV-B radiation) at higher geographical altitudes may have significant effects on phytoplankton populations.  相似文献

Mycosporine-like amino acids (MAAs) are a family of more than 20 compounds having absorption maxima between 310 and 362 nm. These compounds are well known for their UV-absorbing/screening role in various organisms and seem to have evolutionary significance. In the present investigation we tested four cyanobacteria, e.g., Anabaena variabilis PCC 7937, Anabaena sp. PCC 7120, Synechocystis sp. PCC 6803 and Synechococcus sp. PCC 6301, for their ability to synthesize MAA and conducted genomic and phylogenetic analysis to identify the possible set of genes that might be involved in the biosynthesis of these compounds. Out of the four investigated species, only A. variabilis PCC 7937 was able to synthesize MAA. Genome mining identified a combination of genes, YP_324358 (predicted DHQ synthase) and YP_324357 (O-methyltransferase), which were present only in A. variabilis PCC 7937 and missing in the other studied cyanobacteria. Phylogenetic analysis revealed that these two genes are transferred from a cyanobacterial donor to dinoflagellates and finally to metazoa by a lateral gene transfer event. All other cyanobacteria, which have these two genes, also had another copy of the DHQ synthase gene. The predicted protein structure for YP_324358 also suggested that this product is different from the chemically characterized DHQ synthase of Aspergillus nidulans contrary to the YP_324879, which was predicted to be similar to the DHQ synthase. The present study provides a first insight into the genes of cyanobacteria involved in MAA biosynthesis and thus widens the field of research for molecular, bioinformatics and phylogenetic analysis of these evolutionary and industrially important compounds. Based on the results we propose that YP_324358 and YP_324357 gene products are involved in the biosynthesis of the common core (deoxygadusol) of all MAAs.  相似文献

Abstract: The effects of solar radiation on photosynthetic oxygen production and pulse amplitude modulated (PAM) fluorescence were measured in the marine brown macroalga Padina pavonia harvested from different depths from the Greek coast near Korinth. In fluence rate-response curves the light compensation point for photosynthetic oxygen production increased and the saturation level decreased with increasing exposure time to solar radiation. Cutting off the UV-B wavelength range (280–315 nm) from solar radiation reduced the inhibition of photosynthesis, and the organisms were less affected when all of the UV radiation was filtered out. Algae collected from 7 m depth were much more prone to photoinhibition than those harvested from rock pools exposed to unfiltered solar radiation. During continuous exposure to solar radiation, rock pool algae showed photoinhibition after longer periods of time than specimens from 7 m or from dark adapted habitats. When subjected to unfiltered solar radiation the ratio of the variable fluorescence to the maximal fluorescence     (Fv = Fm− Fo) rapidly declined with increasing exposure time. However, again algae from 7 m depth were more prone to photoinhibition than rock pool algae. The differences between the two ecological strains were less obvious when UV-B or total UV was removed from solar radiation. Only in the latter case a complete recovery was observed after 2 h while, when exposed to unifiltered sunlight, only the rock pool algae recovered completely within that time.  相似文献

When rotated horizontally in a cuvette in a strong lateral light beam, the flagellateEuglena gracilis effectively corrects its course and shows negative phototaxis, provided the angular velocity does not exceed 200s–1. Faster rotations cannot be corrected effeciently. In two strong light beams of equal illuminance perpendicular to each other, the cells move along the resultant away from the light beams. Decreasing the illuminance of one beam causes increasing numbers of the organisms to orient with respect to the stronger light source. In two perpendicular low illuminance beams (>200lx), the population splits into two components moving towards either light source. The percentage of cells in each component depends on the relative illuminances. The results can be explained by the shading hypothesis combined with a dichroic orientation of the photoreceptor molecules perpendicular to the long axis of the cells. Externally applied electric dc fields have no effect on positive or negative phototaxis; this supports the hypothesis that electrical potential changes are not involved in the sensory transduction chain of photoorientation inEuglena.  相似文献

We present an improved method for genomic DNA extraction from cyanobacteria by updating the earlier method from our group (Sinha et al. 2001) that does not require lysozyme treatment or sonication to lyse the cells. This method use lysis buffer to lyse the cells and also skips the initial treatments to remove the exopolysaccharides or to break the clumps. To test the efficacy of the method DNA was extracted from the freshwater cyanobacteria Anabaena variabilis PCC 7937, Anabaena sp. PCC 7120, Synechocystis sp. PCC 6803, Synechococcus sp. PCC 6301 and Rivularia sp. HKAR-4 (Accession number: FJ939128). The spectrophotometric and gel electrophoresis analysis revealed high yield and high quality of genomic DNA extracted by this method. Furthermore, the RAPD resulted in the amplification of unidentified genomic regions of various lengths; however, rDNA amplification gave only one band of 1.5 kb in all studied cyanobacteria. Thymine dimer detection study revealed that thymine dimers are induced only by UV-B radiation in A. variabilis PCC 7937 and there is no effect of PAR and UV-A on its genome. Collectively, all these findings put forward the applicability of this method in different studies and purposes.  相似文献

The mycosporine-like amino acid (MAA) profile of a rice-field cyanobacterium, Anabaena doliolum, was studied under PAR and PAR + UVR conditions. The high-performance liquid chromatographic analysis of water-soluble compounds reveals the biosynthesis of three MAAs, mycosporine-glycine (lambda (max) = 310 nm), porphyra-334 (lambda (max) = 334 nm) and shinorine (lambda (max) = 334 nm), with retention times of 4.1, 3.5 and 2.3 min, respectively. This is the first report for the occurrence of mycosporine-glycine and porphyra-334 in addition to shinorine in Anabaena strains studied so far. The results indicate that mycosporine-glycine (monosubstituted) acts as a precursor for the biosynthesis of the bisubstituted MAAs shinorine and porphyra-334. Mycosporine-glycine was under constitutive control while porphyra-334 and shinorine were induced by UV-B radiation, indicating the involvement of UV-regulated enzymes in the biotransformation of MAAs. It seems that A. doliolum is able to protect its cell machinery from UVR by synthesizing a complex set of MAAs and thus is able to survive successfully during the summer in its natural brightly lit habitats.  相似文献
