5-Androstene-3β,7β,17β-triol (β-AET), an active metabolite of dehydroepiandrosterone (DHEA), reversed glucocorticoid (GC)-induced suppression of IL-6, IL-8 and osteoprotegerin production by human osteoblast-like MG-63 cells and promoted osteoblast differentiation of human mesenchymal stem cells (MSCs). In a murine thermal injury model that includes glucocorticoid-induced osteopenia, β-AET significantly (p<0.05) preserved bone mineral content, restored whole body bone mineral content and endochondral growth, suggesting reversal of GC-mediated decreases in chondrocyte proliferation, maturation and osteogenesis in the growth plate. In men and women, levels of β-AET decline with age, consistent with a role for β-AET relevant to diseases associated with aging. β-AET, related compounds or synthetic derivatives may be part of effective therapeutic strategies to accelerate tissue regeneration and prevent or treat diseases associated with aging such as osteoporosis.  相似文献

Despite recent progress in our understanding of carotenogenesis in plants, the mechanisms that govern overall carotenoid accumulation remain largely unknown. The Orange (Or) gene mutation in cauliflower (Brassica oleracea var botrytis) confers the accumulation of high levels of beta-carotene in various tissues normally devoid of carotenoids. Using positional cloning, we isolated the gene representing Or and verified it by functional complementation in wild-type cauliflower. Or encodes a plastid-associated protein containing a DnaJ Cys-rich domain. The Or gene mutation is due to the insertion of a long terminal repeat retrotransposon in the Or allele. Or appears to be plant specific and is highly conserved among divergent plant species. Analyses of the gene, the gene product, and the cytological effects of the Or transgene suggest that the functional role of Or is associated with a cellular process that triggers the differentiation of proplastids or other noncolored plastids into chromoplasts for carotenoid accumulation. Moreover, we demonstrate that Or can be used as a novel genetic tool to induce carotenoid accumulation in a major staple food crop. We show here that controlling the formation of chromoplasts is an important mechanism by which carotenoid accumulation is regulated in plants.  相似文献

We used X-rays from a linear accelerator and from a low energy therapeutic source to calibrate the single cell gel electrophoresis (comet assay), a widely used method to measure DNA damage. γ-Rays from 60Co, with known efficiency in inducing DNA breakage, were used as reference. Human lymphocytes and one murine tumour cell line, F10-M3 cells, were irradiated under different experimental conditions. A similar relationship between radiation dose and induced DNA damage was obtained with γ- and X-rays. A calibration curve was constructed to convert the comet assay raw data into break frequency. The median levels of DNA breaks and oxidative damage in circulating lymphocytes from healthy volunteers were calculated to be 0.76 and 0.80 breaks/109 Da, respectively, (0.50 and 0.52 breaks/106 bp). The values of oxidative DNA damage were in the same order of magnitude as those found by others with HPLC methods.  相似文献

BACKGROUND: Endometriosis is considered a premalignant process whose association with carcinoma is well documented. We discuss a case of clear cell carcinoma with an unusual presentation in that it was located outside the abdominal cavity and was the only lesion noted clinically and radiologically. The histopathologic diagnostic criteria will be reviewed, as will the association of carcinomas with endometriosis. Furthermore, we will review the current literature of extraovarian clear cell carcinoma associated with endometriosis with regard to clinical outcome. CASE: A 42-year-old Hispanic woman presented to the Fine Needle Aspiration (FNA) Clinic at Parkland Hospital, Dallas, Texas, in August 2005 secondary to a growing area of firmness associated with her midline abdominal hysterectomy scar. A single mass anterior to the abdominal wall was identified radiologically. The FNA sample was reported as highly atypical cells suspicious for adenocarcinoma. Excision of this mass revealed a clear cell carcinoma in a background of endometriosis. CONCLUSION: Clear cell carcinoma is one of the most prevalent carcinomas associated with endometriosis, whether identified in the ovary or extraovarian site. This case is perplexing because the mass was a solitary lesion and the patient never had documented endometriosis. The unusual presentation may make survival difficult to predict.  相似文献

The encystment of Physarum polycephalum plasmodia, also called spherulation, involves the synthesis of many specific mRNAs and proteins. Most of these molecules accumulate at the onset of the major morphological and physiological changes typical of this differentiation pathway and are not present during the other two transitions leading to dormancy in Physarum, namely sporulation and encystment of amoebae. The nucleotide sequences of apparently full-length cDNA copies of the four major encystment-specific mRNAs were determined. The four sequences included the entire coding regions and at least 26 nucleotides of the 5'-nontranscribed leaders. The encoded proteins were named spherulins. We found that spherulins 1a and 1b are 81% homologous and are thus members of a gene family. They both possess putative signal peptides and N-glycosylation sites, suggesting that they are cell-wall glycoproteins. Spherulin 2a and spherulin 3a are non-homologous proteins. The absence of signal peptides suggests that they are intracellular structural proteins. Low-stringency Southern hybridizations showed that each also belongs to a two-member gene family.  相似文献

Virginiamycin is an antibiotic composed of two synergistic factors, M and S, which stop growth and protein synthesis in procaryotic cells. The two virginiamycin components, separately and in combination, do not alter the multiplication of algae in heterotrophic media. However, virginiamycin M inhibits chlorophyll formation, and virginiamycin S, which alone has no apparent effect, increases this inhibitory action of M.Virginiamycin M produces bleaching of Euglena gracilis: this phenotypic change is temporary in the absence of S, but permanent if S is present. Characteristic alterations of chloroplast structure occur in the presence of virginiamycin M: disappearance of the pyrenoid, and appearance of free-thylakoids. In the presence of both virginiamycins, chloroplasts loose their spindle shape and their lamellar systems, and are converted into reticulated bodies. There is, thus, a relationship between morphological, biochemical and genetic alterations of the chloroplasts.On the other hand, mitochondria from virginiamycin-treated cells appear intact. The reason for such difference between chloroplasts and mitochondria is unknown.A theory explaining the induction of cytoplasmic mutants by protein inhibitors is proposed.
