Root exudation from Hordeum vulgare in response to localized nitrate supply   总被引:2,自引:0,他引:2  
Root proliferation as a response to exploit zones of nutrient enrichment in soil has been demonstrated for a wide range of plant species. However, the effectiveness of this as a strategy to acquire nutrients is also dependent on interactions with the soil microbial community. Specifically, C-flow from roots modifies microbial activity and probably the balance between nutrient mineralization and immobilization processes in the rhizosphere. In this study, near-natural abundance 13C-labelling and gene-reporter methods were applied to determine the effects of uneven nitrate supply to roots of Hordeum vulgare on assimilate partitioning and root exudation. Plants were initially grown in uniform nitrate supply in split-root, sand microcosms after which one treatment continued to receive uniform supply, and the other received nitrate to one root compartment only. At the time of imposing the treatments, the CO2 supplied to the plants was switched to a cylinder source, providing a distinct delta13C-signature and allowing the fate of new assimilate within the plants to be determined. The labelling approach allowed quantification of the expected preferential allocation of new C-assimilate to roots in enriched nitrate, prior to any measurable effect on whole biomass or root architecture. Biosensor (lux-marked Pseudomonas fluorescens 10586 pUCD607) bioluminescence, quantified spatially by CCD imaging, demonstrated that root exudation was significantly increased for roots in enriched nitrate. This response of root exudation, being primarily associated with root apices and concurrent with enhanced assimilate supply, strongly suggests that C-flow from roots is an integral component of the proliferation response to nitrate.  相似文献

For the first time, the cyanobacterial diversity from microbial mats in lakes of Eastern Antarctica was investigated using microscopic and molecular approaches. The present study assessed the biogeographical distribution of cyanobacteria in Antarctica. Five samples were taken from four lakes spanning a range of different ecological environments in Larsemann Hills, Vestfold Hills and Rauer Islands to evaluate the influence of lake characteristics on the cyanobacterial diversity. Seventeen morphospecies and 28 16S rRNA gene-based operational taxonomic units belonging to the Oscillatoriales, Nostocales and Chroococcales were identified. The internal transcribed spacer was evaluated to complement the 16S rRNA gene data and showed similar but more clear-cut tendencies. The molecular approach suggested that potential Antarctic endemic species, including a previously undiscovered diversity, are more abundant than has been estimated by morphological methods. Moreover, operational taxonomic units, also found outside Antarctica, were more widespread over the continent than potential endemics. The cyanobacterial diversity of the most saline lakes was found to differ from the others, and correlations between the sampling depth and the cyanobacterial communities can also be drawn. Comparison with database sequences illustrated the ubiquity of several cyanobacterial operational taxonomic units and their remarkable range of tolerance to harsh environmental conditions.  相似文献

Xylan is the major component of hemicellulose, and xylan should be fully utilized to improve the efficiencies of a biobased economy. There are a variety of industrial reaction conditions in which an active xylanase enzyme would be desired. As a result, xylanase enzymes with different activity profiles are of great interest. We isolated a xylanase gene (xyn10) from a Flavobacterium sp. whose sequence suggests that it is a glycosyl hydrolase family 10 member. The enzyme has a temperature optimum of 30°C, is active at cold temperatures, and is thermolabile. The enzyme has an apparent Km of 1.8 mg/ml and kcat of 100 sec−1 for beechwood xylan, attacks highly branched native xylan substrates, and does not have activity against glucans.  相似文献

A series of cationic, half-sandwich ruthenium complexes with the general formula [(η6-p-cymene)RuCl(MeSC6H42-NCHAr)][PF6] (3a-h), have been prepared from the reaction of [(η6-p-cymene)RuCl2]2 with various N,S-donor Schiff base ligands derived from 2-(methylthio)aniline and several substituted benzaldehydes. The related aniline complex [(η6-p-cymene)RuCl(MeS-C6H4-2-NH2)][PF6] (4) was synthesized from 2-(methylthio)aniline. All of the ruthenium complexes were characterized by IR, 1H NMR, and UV/Vis spectroscopies. The molecular structure of complex 4 was determined by X-ray crystallography.  相似文献

Mono- and multi-segmental testing methods are required to identify segmental motion patterns and evaluate the biomechanical behaviour of the spine. This study aimed to evaluate a new testing system for multisegmental specimens using a robot combined with an optical motion analysis system. After validation of the robotic system for accuracy, two groups of calf specimens (six monosegmental vs. six multisegmental) were mounted and the functional unit L3-4 was observed. Using rigid body markers, range of motion (ROM), elastic zone (EZ) and neutral zone (NZ), as well as stiffness properties of each functional spine unit (FSU) was acquired by an optical motion capture system. Finite helical axes (FHA) were calculated to analyse segmental movements. Both groups were tested in flexion and extension. A pure torque of 7.5 Nm was applied. Statistical analyses were performed using the Mann-Whitney U-test. Repeatability of robot positioning was -0.001±0.018 mm and -0.025±0.023° for translations and rotations, respectively. The accuracy of the optical system for the proposed set-up was 0.001±0.034 mm for translations and 0.075±0.12° for rotations. No significant differences in mean values and standard deviations of ROM for L3-4 compared to literature data were found. A robot-based facility for testing multisegmental spine units combined with a motion analysis system was proposed and the reliability and reproducibility of all system components were evaluated and validated. The proposed set-up delivered ROM results for mono- and multi-segmental testing that agreed with those reported in the literature. Representing the FHA via piercing points determined from ROM was the first attempt showing a relationship between ROM and FHA, which could facilitate the interpretation of spine motion patterns in the future.  相似文献

Currently there are several neonicotinoid insecticide seed treatments registered for use on soybean (Glycine max L.), with disparity in adoption rates in the eastern United States. A complex of seedling insect pests is found in mid-south soybean, but thrips are the primary early season pest of soybean in Virginia and North Carolina. Published knowledge regarding their impact on soybean yield is minimal, as is the impact of thrips on soybean yield; thrips species composition is also understudied. In 2008 through 2010, nine field experiments in Virginia and North Carolina were conducted to evaluate the impact on thrips population dynamics; the influence on yield of neonicotinoid seed treatments, imidacloprid and thiamethoxam, was reported from nine of these experiments. Moreover, thrips species abundance was recorded in three of these experiments. Both imidacloprid and thiamethoxam reduced thrips densities compared with untreated soybean. Thiamethoxam was more effective than imidacloprid in reducing adult thrips densities at 5 wk after planting. Adult densities peaked at 3 wk after planting, followed by larval densities, which peaked at 4 wk after planting. The most abundant thrips species was Frankliniella fusca (Hinds), followed by Neohydatothrips variabilis (Beach). Other common species included F. occidentalis (Pergande) and F. tritici (Fitch). In general, F. fusca was more common earlier in the season, while N. variabilis was more common later in the season. There were no significant differences in yield among any of the treatments or in the untreated controls. Although neonicotinoid insecticides reduced thrips abundance, data collected in these studies demonstrated that there was no positive yield response.  相似文献

Isothermal calorimetric studies of the binding of iron(III) citrate to ferric ion binding protein from Neisseria gonorrhoeae suggested the complexation of a tetranuclear iron(III) cluster as a single step binding event (apparent binding constant K(app) (ITC) = 6.0(5) × 10(5) M(-1)). High-resolution Fourier transform ion cyclotron resonance mass spectrometric data supported the binding of a tetranuclear oxo(hydroxo) iron(III) cluster of formula [Fe(4)O(2)(OH)(4)(H(2)O)(cit)](+) in the interdomain binding cleft of FbpA. The mutant H9Y-nFbpA showed a twofold increase in the apparent binding constant [K(app) (ITC) = 1.1(7) × 10(6) M(-1)] for the tetranuclear iron(III) cluster compared to the wild-type protein. M?ssbauer spectra of Escherichia coli cells overexpressing FbpA and cultured in the presence of added (57)Fe citrate were indicative of the presence of dinuclear and polynuclear clusters. FbpA therefore appears to have a strong affinity for iron clusters in iron-rich environments, a property which might endow the protein with new biological functions.  相似文献
