Caveolins are coat proteins of caveolae, small flask-shaped pits of the plasma membranes of most cells. Aside from roles in caveolae formation, caveolins recruit, retain and regulate many caveolae-associated signalling molecules. Caveolin-protein interactions are commonly considered to occur between a ∼20 amino acid region within caveolin, the caveolin scaffolding domain (CSD), and an aromatic-rich caveolin binding motif (CBM) on the binding partner (фXфXXXXф, фXXXXфXXф or фXфXXXXфXXф, where ф is an aromatic and X an unspecified amino acid). The CBM resembles a typical linear motif - a short, simple sequence independently evolved many times in different proteins for a specific function. Here we exploit recent improvements in bioinformatics tools and in our understanding of linear motifs to critically examine the role of CBMs in caveolin interactions. We find that sequences conforming to the CBM occur in 30% of human proteins, but find no evidence for their statistical enrichment in the caveolin interactome. Furthermore, sequence- and structure-based considerations suggest that CBMs do not have characteristics commonly associated with true interaction motifs. Analysis of the relative solvent accessible area of putative CBMs shows that the majority of their aromatic residues are buried within the protein and are thus unlikely to interact directly with caveolin, but may instead be important for protein structural stability. Together, these findings suggest that the canonical CBM may not be a common characteristic of caveolin-target interactions and that interfaces between caveolin and targets may be more structurally diverse than presently appreciated.  相似文献

Changes in temperature and moisture as a result of climate forcing can impact performance of planted trees. Tree performance may also be sensitive to new soil conditions, for example, brought about by seeds germinating in soils different from those colonized by ancestral populations. Such “edaphic constraint” may occur with natural migration or human‐assisted movement. Pinus ponderosa seedlings, sourced from one location (“home” site), were grown across a field environmental gradient in either their original home soil or in soils from two different “away” sites. Seedlings were inoculated with site‐specific soil organisms by germinating seeds in living soil. After 6 months, the inoculated seedlings were transplanted into sterilized soils from the home or away sites. This experimental design allowed us to uncouple the importance of abiotic and biotic soil properties and test (1) how biotic and abiotic soil properties interact with climate to influence plant growth and stress tolerance, and (2) the role of soil biota in facilitating growth in novel environments. Seedlings grew least in hotter and drier away sites with away soil biota. Home soil biota ameliorated negative impacts on growth of hotter and drier away sites. Measurements of photosynthetic rate, stomatal conductance, and chlorophyll florescence (Fv/Fm) suggest that edaphic constraint reduced growth by increasing tree water stress. Results suggest that success of Ponderosa pine plantings into warming environments will be enhanced by pre‐inoculation with native soil biota of the seed source.  相似文献

Intra-plastid proteases play crucial and diverse roles in the development and maintenance of non-photosynthetic plastids and chloroplasts. Formation and maintenance of a functional thylakoid electron transport chain requires various protease activities, operating in parallel, as well as in series. This review first provides a short, referenced overview of all experimentally identified plastid proteases in Arabidopsis thaliana. We then focus on the Clp protease system which constitutes the most abundant and complex soluble protease system in the plastid, consisting of 15 nuclear-encoded members and one plastid-encoded member in Arabidopsis. Comparisons to the simpler Clp system in photosynthetic and non-photosynthetic bacteria will be made and the role of Clp proteases in the green algae Chlamydomonas reinhardtii will be briefly reviewed. Extensive molecular genetics has shown that the Clp system plays an essential role in Arabidopsis chloroplast development in the embryo as well as in leaves. Molecular characterization of the various Clp mutants has elucidated many of the consequences of loss of Clp activities. We summarize and discuss the structural and functional aspects of the Clp machinery, including progress on substrate identification and recognition. Finally, the Clp system will be evaluated in the context of the chloroplast protease network. Nitric oxide is an important mediator that participates in reduction-oxidation (redox) mechanisms and in cellular signal transduction pathways. Two types of post-translational modifications are induced by nitric oxide: S-nitrosylation of cysteine residues and nitration of tyrosine residues. Two-dimensional gel electrophoresis-based Western blotting was used to detect, and liquid chromatography (LC)-tandem mass spectrometry (MS/MS) to determine the amino acid sequence of, several different nitrated proteins in the human pituitary. Proteins from several 2D gel spots, which corresponded to the strongly positive anti-nitrotyrosine Western blot spots, were subjected to in-gel trypsin-digestion and LC-MS/MS analysis. MS/MS, SEQUEST analysis, and de novo sequencing were used to determine the nitration site of each nitrated peptide. A total of four different nitrated peptides were characterized and were matched to four different proteins: synaptosomal-associated protein, actin, immunoglobulin alpha Fc receptor, and cGMP-dependent protein kinase 2. Those nitrotyrosyl-proteins participate in neurotransmission, cellular immunity, and cellular structure and mobility.  相似文献

Macrophages are important mediators of the immune response to infection by virtue of their ability to secrete cytokines that trigger inflammation. Toll-like receptors (TLRs) are largely responsible for meditating the activation of macrophages by pathogens. IRAK-1 is a proximal protein kinase in TLR signalling pathways and hence its activation must be tightly regulated. However, the mechanisms which control the activation of IRAK-1 are poorly understood. IRAK-1 contains a death domain at its N-terminus that mediates its interaction with other death domain containing proteins, a central Ser/Thr kinase domain, and a C-terminal domain that contains binding motifs for TRAF6. We show here that deletion of the death domain or the majority of the C-terminal domain markedly enhanced the capacity of IRAK-1 to activate NF-κB in a TLR-independent manner in RAW 264.7 macrophages. Furthermore, the C-terminal truncation mutant spontaneously oligomerised and formed complexes with the negative regulator IRAK-M in the absence of TLR activation. In contrast to the binding of IRAK-M to IRAK-1, the death domain of IRAK-1 was not required for the interaction of IRAK-4 with IRAK-1. On the basis of these results we propose a model in which IRAK-1 is held in a closed, inactive conformation via an intramolecular mechanism involving its C-terminal domain and possibly the death domain. Phosphorylation of IRAK-1 by IRAK-4 in response to TLR activation may then release IRAK-1 from the inhibitory constraint exerted by its C-terminal domain.  相似文献

The maternal effect mutant grandchildless in Drosophila subobscura has been analyzed with the electron microscope. The original mutation was linked to a visible genetic marker and established in a balanced stock. Oocytes and early embryos were examined by both transmission and scanning electron microscopy. The earliest defect is seen in mutant eggs and occurs at the end of oogenesis. In the cortex, at both the anterior and the posterior tips, regions appear which are free of ribosomes, mitochondria, and other cytoplasmic organelles. Most of the polar granules are included in these regions at the posterior tip. Following oviposition, this cytoplasmic segregation is no longer observed and most polar granules have disappeared. The few remaining granules are presumed to derive from the peripheral polar plasm which does not become segregated. During embryogenesis there is a retarded movement of nuclei to the anterior and posterior cortices. At the posterior tip nuclei are delayed in reaching the lateral sides and never move directly into the posterior polar plasm. Pole cells never form. After the last syncytial division the lateral nuclei move under the posterior polar plasm to complete the blastoderm. The posterior polar plasm itself protrudes during blastoderm formation as long cytoplasmic extensions which separate from the blastoderm as cytoplasmic blebs. Neither polar granules nor mitochondria are found in these blebs. The grandchildless phenotype is due to the failure of nuclei to migrate directly into the posterior polar plasm. The defect in the polar plasm presumably is related to the process in mature eggs whereby portions of the cortex become segregated at both anterior and posterior tips. This process may change the properties of the posterior polar plasm so that nuclei do not penetrate into it.  相似文献

Domestication is one of the strongest forms of short-term, directional selection. Although selection is typically only exerted on one or a few target traits, domestication can lead to numerous changes in many seemingly unrelated phenotypes. It is unknown whether such correlated responses are due to pleiotropy or linkage between separate genetic architectures. Using three separate intercrosses between wild and domestic chickens, a locus affecting comb mass (a sexual ornament in the chicken) and several fitness traits (primarily medullary bone allocation and fecundity) was identified. This locus contains two tightly-linked genes, BMP2 and HAO1, which together produce the range of pleiotropic effects seen. This study demonstrates the importance of pleiotropy (or extremely close linkage) in domestication. The nature of this pleiotropy also provides insights into how this sexual ornament could be maintained in wild populations.  相似文献

A series of cationic, half-sandwich ruthenium complexes with the general formula [(η6-p-cymene)RuCl(MeSC6H42-NCHAr)][PF6] (3a-h), have been prepared from the reaction of [(η6-p-cymene)RuCl2]2 with various N,S-donor Schiff base ligands derived from 2-(methylthio)aniline and several substituted benzaldehydes. The related aniline complex [(η6-p-cymene)RuCl(MeS-C6H4-2-NH2)][PF6] (4) was synthesized from 2-(methylthio)aniline. All of the ruthenium complexes were characterized by IR, 1H NMR, and UV/Vis spectroscopies. The molecular structure of complex 4 was determined by X-ray crystallography.  相似文献
