Plants of several species, if grown at high relative air humidity (RH ≥85%), develop stomata that fail to close fully in case of low leaf water potential. We studied the effect of a reciprocal change in RH, at different stages of leaf expansion of Rosa hybrida grown at moderate (60%) or high (95%) RH, on the stomatal closing ability. This was assessed by measuring the leaf transpiration rate in response to desiccation once the leaves had fully expanded. For leaves that started expanding at high RH but completed their expansion after transfer to moderate RH, the earlier this switch took place the better the stomatal functioning. Leaves initially expanding at moderate RH and transferred to high RH exhibited poor stomatal functioning, even when this transfer occurred very late during leaf expansion. Applying a daily abscisic acid (ABA) solution to the leaves of plants grown at continuous high RH was effective in inducing stomatal closure at low water potential, if done before full leaf expansion (FLE). After FLE, stomatal functioning was no longer affected either by the RH or ABA level. The results indicate that the degree of stomatal adaptation depends on both the timing and duration of exposure to high RH. It is concluded that stomatal functionality is strongly dependent on the humidity at which the leaf completed its expansion. Since little is known about how the Mediterranean Basin ecosystems are affected by nitrogen deposition, we aimed to understand the use of nitrogen by distinct plant functional groups (PFG: summer semi-deciduous and evergreen sclerophylls) present in the Mediterranean maquis in order to assess which may be more affected by changes in nitrogen availability. The availability of soil inorganic nitrogen, leaf nitrate concentrations and nitrate reductase activity (in vivo and in vitro) were measured during the year in three plant species from each PFG. The patterns of in vitro NRA along the shoot and through the day were also determined. Although summer semi deciduous species occupied soil patches richer in nitrate, their leaf NRA were significantly lower than that of evergreen sclerophylls species. The pattern of nitrate and ammonium availabilities along the year also distinguished the PFG. Results show that each PFG is composed of a number of physiologically similar species. Patterns of NRA varied according to the PFG, which may represent distinct specializations of co-occurring species to access nitrogen. Therefore, the NRA can be used as an indicator of the nitrate availability taking into consideration the time of the year, the plant species and its PFG. 相似文献
This study was carried out to evaluate the molecular pattern of all available Brazilian human T-cell lymphotropic virus type 1 Env (n = 15) and Pol (n = 43) nucleotide sequences via epitope prediction, physico-chemical analysis, and protein potential sites identification, giving support to the Brazilian AIDS vaccine program. In 12 previously described peptides of the Env sequences we found 12 epitopes, while in 4 peptides of the Pol sequences we found 4 epitopes. The total variation on the amino acid composition was 9 and 17% for human leukocyte antigen (HLA) class I and class II Env epitopes, respectively. After analyzing the Pol sequences, results revealed a total amino acid variation of 0.75% for HLA-I and HLA-II epitopes. In 5 of the 12 Env epitopes the physico-chemical analysis demonstrated that the mutations magnified the antigenicity profile. The potential protein domain analysis of Env sequences showed the loss of a CK-2 phosphorylation site caused by D197N mutation in one epitope, and a N-glycosylation site caused by S246Y and V247I mutations in another epitope. Besides, the analysis of selection pressure have found 8 positive selected sites (w = 9.59) using the codon-based substitution models and maximum-likelihood methods. These studies underscore the importance of this Env region for the virus fitness, for the host immune response and, therefore, for the development of vaccine candidates. 相似文献
The aim of this study was to evaluate the regional differences in the surface temperature of Naked Neck hens that were subjected to different temperatures in a semi-arid environment. The surface temperature was measured in four body regions (face, neck, legs and feathered area) of 60 Naked Neck hens. The following environmental variables were measured at the center of the shed: the black globe temperature (TG), air temperature (TA), wind speed (U) and relative humidity (RH). The TA was divided into three classes: 1 (24.0–26.0 °C), 2 (26.1–28.9 °C) and 3 (29.0–31.0 °C). An analysis of variance was performed by the least squares method and a comparison of the means by the Tukey-Kramer test. The results showed a significant effect of TA class, the body region and the interaction between these two effects on the surface temperature. There was no significant difference between the TA classes for the face and neck. The legs and feathered area showed significant differences between the TA classes. Regarding the effect of body regions within each TA class, there was a significant difference among all regions in the three TA classes. In all TA classes the neck had the highest average followed by the face and legs. The feathered area showed the lowest average of the different TA classes. In conclusion, this study showed that there are regional differences in the surface temperature of Naked Neck hens, with the legs acting as thermal windows. 相似文献
The effects of local and regional environmental variables as well as spatial gradients on the plant species composition of two types of alder-dominated forests (riparian forests and alder carrs) with contrasting connectivity were studied across the Western Carpathians from Hungary through Slovakia to Poland. We used large vegetation (240 sampling plots) and environmental (24 variables) datasets, which were accompanied by spatial variables represented by principal coordinates of neighbour matrices. Canonical correspondence analysis (CCA) of the two datasets revealed 13 and 29 variables with significant effects on variation in species composition of alder carrs and riparian alder forests, which jointly explained 41.2% and 36.4% of the variability, respectively. Altitude was the most important factor explaining 7.7% of the variability in the species composition of alder carrs and 8.2% in riparian alder forests. Variation partitioning in CCA revealed that local variables were crucial drivers for species composition patterns in alder carrs, while spatial processes unrelated to the measured environmental variables shaped the vegetation structure of riparian forests.
The patterning and development of multicellular organisms require a precisely controlled balance between cell proliferation, differentiation and death. The regulation of apoptosis is an important aspect to achieve this balance, by eliminating unnecessary or mis-specified cells which, otherwise, may have harmful effects on the whole organism. Apoptosis is also important for the morphogenetic processes that occur during development and that lead to the sculpting of organs and other body structures. Here, we review recent progress in understanding how apoptosis is regulated during development, focusing on studies using Drosophila or Caenorhabditis elegans as model organisms. 相似文献
Monoclonal antibodies (Mabs) against human chorionic gonadotropin hormone (hCG) were raised by hybridoma technology using Sp2/0 myeloma cells as fusion partner. Sixty-five percent of the total culture wells exhibited hybrid growth and 8% of the total wells (13 culture wells) contained anti-hCG secreting hybrids. A positive hybrid cell line secreting antibodies against the free alpha-subunit of hCG was cloned twice by limiting dilution method and eighty four clones were obtained that secreted monoclonal antibodies anti-alpha hCG. One of these hybridoma clones (1C4) secreting monoclonal antibodies against the free alpha-subunit of hCG was selected for purification and characterization purposes. This hybridoma cell line secreted monoclonal antibodies of IgG1 subclass, which were purified by affinity chromatography on Protein A Sepharose CL-4B column with a final relative recovery of antibody activity of 75% and a purification factor of about 12. The purified preparation was analyzed by SDS-PAGE, native PAGE, and IEF. Specificity studies of this Mab revealed that it recognized specifically an epitope on the free alpha-subunits of hCG, FSH, LH, and TSH as determined by enzyme immunoassays. On the other hand, this Mab exhibited crossreactivity with other pituitary hormones either as free subunits or intact molecules as follows: alpha hCG 100%; intact hCG 1.8%; beta hCG 0.14%; alpha FSH 24.5%; intact FSH 0.8%; beta FSH 0.09%; alpha LH 20.5%; intact LH 0.9%; beta LH 0.08%; alpha TSH 50.5%; intact TSH 3.7%; beta TSH 0.07%; The affinity constant (K) of this Mab with respect to free alpha-subunit of hCG was found to be 1.5 x 10(7) I/mol as determined by the simple antibody dilution analysis method. 相似文献
