Cell proliferation and apoptosis regulate germ cells stock and sperm production, eliminate anomalous gametes, and are essential parameters to consider in fish farming. Herein, spermatogenic activity as well as germ cell proliferation and apoptosis were assessed in Leporinus taeniatus, a seasonal breeding species from the São Francisco River basin, Brazil. Testes of 24 adult fishes from a farming station were sampled between December and July and processed for light and transmission electron microscopy and immunohistochemistry for PCNA and TUNEL assay. The gonadosomatic index and seminiferous tubule diameters presented higher values during the breeding season (December/January and February/March), and then significantly reduced during the regression and resting stages (April/May and June/July). Phagocytosis of spermatozoa by Sertoli cells was evident during gonadal regression, but a significant number (up to 30%) remained at the tubular lumen during the resting stage. A higher PCNA/TUNEL ratio occurred in the breeding period, leading to an elevated proportion (%) of spermatogonia (GA and GB) in resting. Moreover, a higher TUNEL/PCNA ratio indicates the contribution of apoptosis to the reduction of germ cells during testicular regression. Together, these results indicate a shift in the balance between cell proliferation and apoptosis that contributes to the regulation of the spermatogenic cycle and germ cells pool of L. taeniatus kept in captivity. 相似文献

Locular pressure was monitored during ripening of tomato (Lycopersicon esculentum Mill.) fruit and the anatomy of the endocarp surface examined using scanning electron microscopy. The manometric pressure of the locule tissue increased from 0 in mature-green fruit to 10 to 50 Pa at the turning or pink stages, and then subsided in ripe fruit. Nonclimacteric fruit containing the ripening inhibitor (rin) mutation showed a similar pattern of internal pressure accumulation during senescence. Build-up of locular tissue pressure occurred in fruit ripening, on or off the plant, as well as in fruit with different susceptibility to cuticle cracking. Apertures ranging from 18-31 μm in width and 33-41 μm in length, with densities ranging from 6.7 to 47.9 apertures · mm−2 were observed in the endocarp of mature-green fruit. These apertures were progressively occluded during early ripening and were absent in late ripening fruit. Aperture occlusion might result in reduced gas exchange between the locule and external fruit atmosphere, resulting in modification of the locular gas composition. 相似文献

Understanding the social conditions and immunological characteristics that allow some
human immunodeficiency virus (HIV)-exposed patients to remain uninfected represents
an on-going challenge. In this study, the socio-demographic and sexual behaviour
characteristics and immune activation profiles of uninfected individuals exposed to
HIV-infected partners were investigated. A confidential and detailed questionnaire
was administered and venous blood was tested using HIV-1/enzyme immunoassays, plasma
HIV-1 RNA levels/bDNA and immunophenotyping/flow cytometry to determine the
frequencies of CD4 and CD8 T cells expressing activation markers. The data analysis
showed significant differences (p < 0.05) for immune parameters in individuals who
were uninfected, albeit exposed to HIV-infected partners, compared with unexposed
individuals. In particular, the exposed, uninfected individuals had a higher
frequency (median, minimum-maximum) of CD4+HLA-DR+ (4.2,
1.8-6.1), CD8+HLA-DR+ (4.6, 0.9-13.7),
CD4+CD45RO+ (27.5, 14.2-46.6),
CD4+CD45RO+CD62L+ (46.7, 33.9-67.1),
CD8+CD45RA+HLA-DR+ (12.1, 3.4-35.8) and
CD8+CD45RO+HLA-DR+ (9.0, 3.2-14.8) cells, a
decreased percentage of CD8+CD28+ cells (11.7, 4.5-24.0) and a
lower cell-surface expression of Fcγ-R/CD16 on monocytes (56.5, 22.0-130.0). The
plasma HIV-1 RNA levels demonstrated detectable RNA virus loads in 57% of the
HIV-1+ female partners. These findings demonstrate an activation
profile in both CD4 and CD8 peripheral T cells from HIV-1 exposed seronegative
individuals of serodiscordant couples from a referral centre in Belo Horizonte, state

The ability of extracts and fractions of the red seaweed Plocamium brasiliense to inhibit hemorrhagic, edematogenic, hemolytic, clotting and proteolytic activities of Lachesis muta snake venom was analyzed. In Brazil, snakebites by L. muta are low (2 %) when compared to Bothrops genus (90 %); however, their lethality indexes are three times higher than Bothrops. Envenomation by L. muta venom results in hemorrhage, pain, necrosis, hemolysis, myotoxicity, and death. Since antivenom does not efficiently neutralize local effects, a large number of researchers have attempted to identify molecule(s) from natural sources to inhibit such activities to use them as an alternative treatment for snakebite. We tested four extracts of seaweed P. brasiliense obtained with solvents of increasing polarities: n-hexane (HEX), dichloromethane (DCM), ethyl acetate (ACE), and hydroalcohol (HYD). Extracts of alga or fractions were incubated with L. muta venom, and then, biological assays were performed. The extracts, except the HYD, inhibited all the assays but with different potencies. The DCM extract fully inhibited all activities. Moreover, DCM and HEX extracts inhibited hemolysis induced by a phospholipase A2 isolated from L. muta venom (LM-PLA2-I). A fraction from HEX enriched in cholesterol isolated from HEX extract inhibited proteolysis by L. muta venom and hemolysis by LM-PLA2-I; in contrast, monoterpenes isolated from DCM extract did not inhibit both activities. Seaweeds may be a promising source of natural inhibitors of the toxic effects caused by snakebite by L. muta venom, and they may be used to develop new strategies for antivenom treatment. 相似文献

We investigated the live morphology, infraciliature, and small subunit rRNA gene sequences of an unusual population of Stentor polymorphus without symbiotic algae that was isolated from the southeastern region of Brazil. The morphological and molecular data confirmed the identity of this strain as S. polymorphus. The Brazilian S. polymorphus organism is 850–2,000 μm in length in vivo and has colorless cortical granules, a moniliform macronucleus with 6–12 nodules, somatic ciliature composed of 50–60 kineties, a single contractile vacuole located to the left of the cytostome, and a conspicuous oral pouch, and it does not build a lorica. Based on the phylogenetic analyses, the Brazilian S. polymorphus was located within a cluster consisting of four other S. polymorphus sequences, with high support values using both the Bayesian inference and maximum likelihood algorithms. Our study presents the first report of a S. polymorphus population without its symbionts under natural conditions. On the basis of our findings, we propose that the presence or absence of symbiotic algae should not be used as a taxonomic character for the identification of Stentor species. 相似文献

The influence of the allelochemicals ferulic (FA) and vanillic (VA) acids on peroxidase (POD, EC 1.11.1.7) and phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) activities and their relationships with phenolic acid (PhAs) contents and root growth of soybean (Glycine max (L.) Merr.) were examined. Three-day-old seedlings were cultivated in nutrient solution containing FA or VA (0.1 to 1 mM) for 48 h. Both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) and increased PhAs contents. At 0.5 and 1 mM, FA increased soluble POD activity (18% and 47%, respectively) and cell wall (CW)-bound POD activity (61% and 34%), while VA increased soluble POD activity (33% and 17%) but did not affect CW-bound POD activity. At 1 mM, FA increased (82%) while VA reduced (32%) PAL activities. The results are discussed on the basis of the role of these compounds on phenylpropanoid metabolism and root growth and suggest that the effects caused on POD and PAL activities are some of the many mechanisms by which allelochemicals influence plant growth. 相似文献

Aspartic proteinases are present in a variety of organisms including plants. Common features of aspartic proteases include an active site cleft that contains two catalytic aspartic residues, acid pH optima for enzymatic activity, inhibition by pepstatin A. Plant aspartic proteinases occur in seeds and may be involved in the processing of storage proteins. Many of them have been purified and characterized. The presence of aspartic proteases in seeds of Centaurea calcitrapa during germination was investigated by measuring the activity on enzyme extracts. The aspartic proteases are present mainly in the beginning of seed germination suggesting that they could initiate the degradation of protein reserves in germinating seeds.
These proteases were purified by salt precipitation followed by anion-exchange chromatography. Purified aspartic proteases have an optimal pH between 3.5 and 4.5, using FTC-hemoglobin as substrate and an optimal temperature at 52 °C. The ability of seed extracts for milk clotting was tested and the clotting time that was achieved is in the same range found for flower extracts appropriated for special cheeses in which weak clotting agents are required. 相似文献

Here, we evaluate the potential of the bumble bee Bombus impatiens Cresson, obtained from commercial colonies, as a pollen vector for lowbush blueberry, Vaccinium augustifolium Aiton. We wanted to gain insight into the relationships between pollination by introduced bumble bees, the categories of seeds produced, and the weight and the maturity of the blueberries. The effect of B. impatiens foraging in blueberry stands was measured quantitatively through fruit set and seed set analysis. We created a density gradient of B. impatiens by clustering 72 small hives at the southern extremity of blueberry fields located in Girardville (49 degrees 00' N, 72 degrees 33' W), Quebec, Canada. Observers recorded plant and insect data in 52 plots of 1 by 10 m, distributed at distances ranging from 25 to 1,500 m from the hives. From these data, we evaluated the fruit set rate, the weight and maturity of the berries, and the number of seed structures per berry, including true seeds of large, medium, and small size, pseudo-seeds, and ovules. Positive correlations were found between the density of B. impatiens and fruit set as well as the number of large seeds per berry. Large seeds influenced the weight and maturity of berries. B. impatiens acted as a "near-nest central forager" and increased fruit set and seed production up to 100 and 150 m respectively, from the hives. 相似文献

Limnoperna fortunei (Bivalvia, Mitylidae) was introduced into South America in 1991 in the La Plata River (Argentina). It arrived in the ballast water of ships coming
from Asia, where this species is native. It was first observed in 1998 in the Paraguay River. Limnoperna was introduced into the Pantanal region as hull fouling of vessels using the Paraguay–Parana waterway. This study describes
how L. fortunei came to the Pantanal region, and provides details of its occurrence, density, and impacts. From 1999 to 2002, observations
and sampling on natural and artificial substrates in the Paraguay River were made. Some aspects of the spread and impacts,
based on local community information, were also analyzed. On artificial substrate the density reached 523.8 individuals m−2 and on natural substrate (rocks), up to 10,000 individuals m−2 were found. The densities observed were quite low compared to those found in Southern Brazil, where values up to 100,000 individuals m−2 have been recorded in the last 3 years. In the Paraguay River, the population density of L. fortunei can be negatively impacted by periodic low levels of dissolved oxygen and decreases in pH to between 5 and 6. Such conditions
are frequently present during the periodic flooding or inundation of this area. Under these conditions, a high mortality of
L. fortunei was recorded in March of 2002, on both natural and artificial substrates. Despite low densities, L. fortunei can colonize water cooling systems of boats, obstructing water circulation and causing motor overheating. Accumulation in
water supply equipment, such as pumps and pipes has also been observed.
