The complement component C4 of mammals.   总被引:6,自引:0,他引:6  
Human complement component C4 is coded by tandem genes located in the HLA class III region. The products of the two genes, C4A and C4B, are different in their activity. This difference is due to a degree of 'substrate' specificity in the covalent binding reactions of the two isotypes. Mouse also has a duplicated locus, but only one gene produces active C4, while the other codes for the closely related sex-limited protein (Slp). In order to gain some insight into the evolutionary history of the duplicated C4 locus, we have purified C4 from a number of other mammalian species, and tested their binding specificities. Like man, chimpanzee and rhesus monkey appear to produce two C4 types with reactivities similar to C4A and C4B. Rat, guinea pig, whale, rabbit, dog and pig each expresses C4 with a single binding specificity, which is C4B-like. Sheep and cattle express two C4 types, one C4B-like, the other C4A-like, in their binding properties. These results suggest that more than one locus may be present in these species. If this is so, then the duplication of the C4 locus is either very ancient, having occurred before the divergence of the modern mammals, or there have been three separate duplication events in the lines leading to the primates, rodents and ungulates.  相似文献

A few years ago Clark et al. (1973) published a modification of Cole's hematoxylin (Cole 1943) which had several advantages. Not the least of these was the progressive nature of the stain and the use of stable stock solutions from which the stain could be prepared volumetrically. A distinct disadvantage was the necessity of preparing the stain 24 hours before use. In some other work it was recently found that Lugol's solution was a more powerful oxidant than iodine alone. On this basis we developed another variant of Cole's hematoxylin.  相似文献

Experiments conducted on samples collected from a large oligotrophic lake revealed the following: (1) excretion rates of PO inf4 sup3– by single Daphnia thorata were below detection (5 pmol animal–1 min–1) in 20 ml of oligotrophic lake water over a period of 10 min, (2) experimental addition of D. thorata to 20 ml aliquots of lake water decreased community-wide microbial uptake of PO inf4 sup3– on two occasions (as measured by 32PO inf4 sup3– incorporation), and (3) the presence of D. thorata increased uptake by organisms smaller than 1µm, and decreased uptake by large phytoplankton. The specific mechanism for these responses remains unclear, but the results imply that when phytoplankton larger than 1µm encounter cm scale patches of water recently occupied by Daphnia they may experience decreased PO inf4 sup3– availability rather than elevated concentrations of PO inf4 sup3– caused by excretion. We show that 32P uptake experiments using natural plankton assemblages can be influenced by the presence or absence of large zooplankton, and that neither grazing, turbulence, nor PO inf4 sup3– excretion can account for this influence.  相似文献

A quantitative trait locus (QTL) study was carried out in two countries, recording live animal and carcass composition traits. Back-cross calves (385 heifers and 398 steers) were generated, with Jersey and Limousin breed backgrounds. The New Zealand cattle were reared on pasture to carcass weights averaging 229 kg, whilst the Australian cattle were reared on grass and finished on grain (for at least 180 days) to carcass weights averaging 335 kg. From 11 live animal traits and 31 carcass composition traits respectively, 5 and 22 QTL were detected in combined-sire analyses, which were significant ( P  <   0.05) on a genome-wise basis. Fourteen significant traits for carcass composition QTL were on chromosome 2 and these were traits associated with muscling and fatness. This chromosome carried a variant myostatin allele (F94L), segregating from the Limousin ancestry. Despite very different cattle management systems between the two countries, the two populations had a large number of QTL in common. Of the 18 traits which were common to both countries, and which had significant QTL at the genome-wise level, eight were significant in both countries.  相似文献

Over thirty-six different xenobiotic carboxylic acids have been reported to form xenobiotic lipids. The majority form triacylglycerol analogs or cholesterol esters with fewer reports of polar lipids being formed. As yet there is insufficient information to deduce a relationship between the structure of the xenobiotic acid and its activity as a substrate for lipid biosynthesis, although the ability to form a CoA ester appears to be important. The action of monoacylglycerol acyltransferase, diacylglycerol acyltransferase, lecithin cholesterol acyltransferase and a carboxylesterase in synthesizing xenobiotic lipids has been demonstrated. One xenobiotic lipid has been shown to be the cause of granulomatous changes and there are some indications that others may prove to be of toxicological or pharmacological significance. Detailed investigations into several aspects of xenobiotic lipid biochemistry are still required.  相似文献
