Two glasshouse experiments were done to assess the development and metabolic activity of mycorrhizas formed by isolates of
arbuscular mycorrhizal fungi (AMF) from three different genera, Acaulospora, Gigaspora and Glomus on either Pueraria phaseoloides L. or Desmodium ovalifolium L. plants. The second of the two experiments included three levels of a localised phosphate source in the pots. Alkaline
phosphatase (ALP), stained histochemically in the intra-radical mycelium (IRM) of AMF over sequential harvests, did not provide
a direct marker for the efficiency of AMF in mobilising phosphorus (P) for plant growth and development. The ability of the
extra-radical mycelium (ERM) to scavenge a localised phosphate source, determined by its extraction from buried 35-μm mesh
pouches, was dependent on the species of AMF tested. This work indicates that AMF from different genera have unique patterns
of mycelial development when forming mycorrhizas with tropical hosts in the presence of a localised phosphate source. AMF
also appear to have different mechanisms for the control of P transfer, within the mycelium, to the host. The significance
of the architecture of the ERM is discussed as well as the localisation of ALP in the IRM in determining the efficiency of
AMF in terms of P accumulation in planta and subsequent growth of plants.

Background
South Africa has one of the highest per capita rates of tuberculosis (TB) incidence in the world. In 2012, the South African government produced a National Strategic Plan (NSP) to control the spread of TB with the ambitious aim of zero new TB infections and deaths by 2032, and a halving of the 2012 rates by 2016.Methods
We used a transmission model to investigate whether the NSP targets could be reached if immediate scale up of control methods had happened in 2014. We explored the potential impact of four intervention portfolios; 1) “NSP” represents the NSP strategy, 2) “WHO” investigates increasing antiretroviral therapy eligibility, 3) “Novel Strategies” considers new isoniazid preventive therapy strategies and HIV “Universal Test and Treat” and 4) “Optimised” contains the most effective interventions.Findings
We find that even with this scale-up, the NSP targets are unlikely to be achieved. The portfolio that achieved the greatest impact was “Optimised”, followed closely by “NSP”. The “WHO” and “Novel Strategies” had little impact on TB incidence by 2050. Of the individual interventions explored, the most effective were active case finding and reductions in pre-treatment loss to follow up which would have a large impact on TB burden.Conclusion
Use of existing control strategies has the potential to have a large impact on TB disease burden in South Africa. However, our results suggest that the South African TB targets are unlikely to be reached without new technologies. Most perfusion techniques rely on mechanical means to provide blood flow to the isolated organ for maintaining its physiological conditions. The approach usually requires a complicated mechanical system with the associated problems of blood type matching and prevention of blood cell damage. This paper describes a gastrointestinal tract perfusion technique that uses the rabbit's own cardiopulmonary system as the autologous blood supply source. The technique allows for the removal of the complete intestinal loop from the abdominal cavity of the rabbit, and maintains its blood circulation through silastic tubing connections of the catheterized portal vein and cranial and caudal mesenteric arteries. An alternative perfusion site that uses the aorta as the arterial blood supply and the vena cava as the venous return also is described. The isolated perfused GI tract may then be placed in a separate test environment for controlled experiments. For an acute animal test, the approach was found to be a convenient alternative to conventional approaches. 相似文献

The numbers of coliforms, Enterobacteriaceae and Staphylococcus aureus and the extent of their colonization of the fingers of the defeathering machinery within a poultry processing plant were increased after changes in machinery design and a reduction in the level of chlorination. Summary Tadpoles and young toads of Xenopus laevis were maintained in aqueous solutions of potassium perchlorate or thiourea (0.005% or 0.01% w/v) for up to 20 months. Metamorphosis of tadpoles was inhibited. Within a short time large well-vascularized goitres developed in both tadpoles and toads. An ultrastructural investigation of some of the follicular epithelial cells of these goitres revealed some unusual cytoplasmic membranous inclusions and a morphological account of these structures is presented. It is suggested that some of these complex membranous inclusions may give rise to cytosomes.
