Plasminogen is a key component of the haemostatic system in man and the plasma-derived protein molecule has been actively investigated. Within the last few years cDNA and the gene encoding plasminogen have been cloned and the protein has been expressed in a number of eukaryotic or prokaryotic systems. Yields of expressed plasminogen are frequently low. Currently available assays for plasminogen generally rely on the determination of antigen or utilize tripeptide substrates for measuring functional activity, and they have certain limitations. Assays employing relevant protein substrates offer an alternative way to measure function and overcome the drawbacks associated with the other tests. The use of fibrin films for the assay of low levels of recombinant plasminogen has not been described fully before. The two fibrin film-based assays described in this paper are significant additions to the array of assays available for plasminogen molecules.  相似文献

We examined the effects of external mechanical loading on glottic dimensions in 13 normal subjects. When flow-resistive loads of 7, 27, and 48 cmH2O X l-1 X s, measured at 0.2 l/s, were applied during expiration, glottic width at the mid-tidal volume point in expiration (dge) was 2.3 +/- 12, 37.9 +/- 7.5, and 38.3 +/- 8.9% (means +/- SE) less than the control dge, respectively. Simultaneously, mouth pressure (Pm) increased by 2.5 +/- 4, 3.0 +/- 0.4, and 4.6 +/- 0.6 cmH2O, respectively. When subjects were switched from a resistance to a positive end-expiratory pressure at comparable values of Pm, both dge and expiratory flow returned to control values, whereas the level of hyperinflation remained constant. Glottic width during inspiration (unloaded) did not change on any of the resistive loads. There was a slight inverse relationship between the ratio of expiratory to inspiratory glottic width and the ratio of expiratory to inspiratory duration. Our results show noncompensatory glottic narrowing when subjects breathe against an expiratory resistance and suggest that the glottic dimensions are influenced by the time course of lung emptying during expiration. We speculate that the glottic constriction is related to the increased activity of expiratory medullary neurons during loaded expiration and, by increasing the internal impedance of the respiratory system, may have a stabilizing function.  相似文献

DNA sequence comparisons of two mitochondrial DNA genes were used to infer phylogenetic relationships among 17 Felidae species, notably 15 in the previously described pantherine lineage. The polymerase chain reaction (PCR) was used to generate sequences of 358 base pairs of the mitochondrial 12S RNA gene and 289 base pairs of the cytochrome b protein coding gene. DNA sequences were compared within and between 17 felid and five nonfelid carnivore species. Evolutionary trees were constructed using phenetic, cladistic, and maximum likelihood algorithms. The combined results suggested several phylogenetic relationships including (1) the recognition of a recently evolved monophyletic genus Panthera consisting of Panthera leo, P. pardus, P. onca, P. uncia, P. tigris, and Neofelis nebulosa; (2) the recent common ancestry of Acinonyx jubatus, the African cheetah, and Puma concolor, the American puma; and (3) two golden cat species, Profelis temmincki and Profelis aurata, are not sister species, and the latter is strongly associated with Caracal caracal. These data add to the growing database of vertebrate mtDNA sequences and, given the relatively recent divergence among the felids represented here (1-10 Myr), allow 12S and cytochrome b sequence evolution to be addressed over a time scale different from those addressed in most work on vertebrate mtDNA.   相似文献

About 1% of Staphylococcus aureus cells survived the production of gelatin sheets containing nutrient broth. Those cells which survived showed no evidence of injury. Growth occurred in rubbery state gelatin with a w values of 0.98 and 0.93; viability decreased during storage at a w values of 0.89, 0.62 and 0.36 but there was little loss of viability in gelatin at an a w of 0.25 over 27 d storage at 26°C. Assays for enterotoxin A detected no synthesis of new toxin but no loss in pre-formed toxin. The results suggest that high levels of Staph. aureus and its toxins should be excluded from glassy and rubbery state food products in order to prevent illness.  相似文献

We analyzed soil water data from three sites with different soil textures in the shortgrass steppe of northeastern Colorado, USA. Our objective was to evaluate the relationship between the occurrence of plant functional types and the effect of soil texture on soil water availability. Soil water availability was greatest in the upper soil layers at all three sites, but the loamy sand site had significantly greater soil water availability than the sandy clay loam and sandy clay sites in wetter years at depths below 60 cm. Calculations of proportional water availability by layer using both field data and fifty-year soil water model simulations, showed that the sandy clay loam and sandy clay soils on average had greater water availability in layers 30 cm and above, but that the loamy sand had the greatest water availability in layers beneath this, particularly at 105 cm. This observation can be linked to the occurrence of a fine textured subsoil at this site. The textural pattern in the loamy sand profile effectively creates two water resources: a shallow pool accessible to all plants; and a deep pool accessible only to deep-rooted plants. This is offered as an explanation for the co-dominance of the two main plant functional types at the loamy sand site. At the other two sites, shallow-rooted shortgrass vegetation dominated, being more consistent with the general pattern for the area. Thus the patterns of vegetation structure at the three sites were consistent with the hypothesis. Aboveground net primary production data for the three sites, along with transpiration estimates from the model simulations, indicated that the additional water availability in the coarse textured soil was associated with higher overall plant productivity.Nomenclature: Taxonomic nomenclature follows R. The physiological response of plants to different irrigation frequencies may affect plant growth and water use efficiency (WUE; defined as shoot biomass/cumulative irrigation). Glasshouse‐grown, containerized Pelargonium × hortorum BullsEye plants were irrigated either daily at 100% of plant evapotranspiration (ET) (well‐watered; WW), or at 50% ET applied either daily [frequent deficit irrigation (FDI)] or cumulatively every 4 days [infrequent deficit irrigation (IDI)], for 24 days. Both FDI and IDI applied the same irrigation volume. Xylem sap was collected from the leaves, and stomatal conductance (gs) and leaf water potential (Ψleaf) measured every 2 days. As soil moisture decreased, gs decreased similarly under both FDI and IDI throughout the experiment. Ψleaf was maintained under IDI and increased under FDI. Leaf xylem abscisic acid (ABA) concentrations ([X‐ABA]leaf) increased as soil moisture decreased under both IDI and FDI, and was strongly correlated with decreased gs, but [X‐ABA]leaf was attenuated under FDI throughout the experiment (at the same level of soil moisture as IDI plants). These physiological changes corresponded with differences in plant production. Both FDI and IDI decreased growth compared with WW plants, and by the end of the experiment, FDI plants also had a greater shoot fresh weight (18%) than IDI plants. Although both IDI and FDI had higher WUE than WW plants during the first 10 days of the experiment (when biomass did not differ between treatments), the deficit irrigation treatments had lower WUE than WW plants in the latter stages when growth was limited. Thus, ABA‐induced stomatal closure may not always translate to increased WUE (at the whole plant level) if vegetative growth shows a similar sensitivity to soil drying, and growers must adapt their irrigation scheduling according to crop requirements.  相似文献

Capsule Inter-annual survival rates of juvenile Dunlin Calidris alpina increased with wet weather conditions but decreased with the density of conspecifics in winter, ranging from 0.34 to 0.62 in the best model. Adult inter-annual survival remained high and constant at 0.72.

Aims To estimate inter-annual survival rates of both adult and juvenile Dunlin in North Wales. To quantify the effects on survival rates of weather conditions and conspecific density.

Methods Cormack–Jolly–Seber models in Program MARK were used to analyse 21 years of ringing data (1990–2011) from Traeth Lafan, North Wales. Models were constructed using a two age-class structure. The influence of a range of weather conditions and WeBS census data on survival were examined.

Results Adult survival was best modelled at a constant rate of 0.72 (se 0.008) across the 21 years. Juvenile survival rates were found to vary most strongly with total rainfall (mm) during the season and the number of conspecifics present. Survival rates varied from 0.34 (se 0.06) to 0.62 (se 0.09), with higher survival in years with high rainfall and low numbers of birds.

Conclusion Survival rates of juvenile Dunlin are affected by both weather and density of conspecifics. These results have implications for the future of Dunlin in the UK when considering both climate change and habitat loss.  相似文献

This study examined the abiotic and biotic characteristics of ecosystems that allow expression of a life history called ferox trout, the colloquial name given to brown trout Salmo trutta adopting a piscivorous life history strategy, an apex predator in post‐glacial lakes in northern Europe. One hundred and ninety‐two lakes in Scotland show evidence of currently, or historically, supporting ferox S. trutta; their presence was predicted in logistic models by larger and deeper lakes with a large catchment that also support populations of Arctic charr Salvelinus alpinus.  相似文献

Experimental research involving Arabidopsis thaliana often involves the quantification of phenotypic traits during cultivation on compost or other growing media. Many commercially-available growing media contain peat, but peat extraction is not sustainable due to its very slow rate of formation. Moreover, peat extraction reduces peatland biodiversity and releases stored carbon and methane into the atmosphere. Here, we compared the experimental performance of Arabidopsis on peat-based and several types of commercially-available peat-free growing media (variously formed from coir, composted bark, wood-fibre, and domestic compost), to provide guidance for reducing peat use in plant sciences research with Arabidopsis. Arabidopsis biomass accumulation and seed yield were reduced by cultivation on several types of peat-free growing media. Arabidopsis performed extremely poorly on coir alone, presumably because this medium was completely nitrate-free. Some peat-free growing media were more susceptible to fungal contamination. We found that autoclaving of control (peat-based) growing media had no effect upon any physiological parameters that we examined, compared with non-autoclaved control growing media, under our experimental conditions. Overall, we conclude that Arabidopsis performs best when cultivated on peat-based growing media because seed yield was almost always reduced when peat-free media were used. This may be because standard laboratory protocols and growth conditions for Arabidopsis are optimized for peat-based media. However, during the vegetative growth phase several phenotypic traits were comparable between plants cultivated on peat-based and some peat-free media, suggesting that under certain circumstances peat-free media can be suitable for phenotypic analysis of Arabidopsis.  相似文献
